DNA barcoding is a molecular tool that uses a standardized DNA region to identify *** preliminary study reported here is the first attempt to specifically focus on universality and attributes of candidate barcodes across a wide systematic range of *** tested eight previously proposed plant barcoding regions(atpF-atpH, ITS2. matK, psbK-psbI, rbcL, rpoB, rpoC1, and trnH-psbA) and two popular phylogenetic markers(rps4 and trnL-trnF of cpDNA) in 49 moss species and 9 liverwort species, representing half of the orders in moss *** ITS2. rbcL, rpoC1, rps4, trnH-psbA and trnL-trnF regions showed good universality, and therefore the efficacy of these loci as DNA barcodes was further evaluated in 36 mosses and 2.liverworts, each of which included two to three individuals per *** five loci, ***, rpoC1, rps4, trnH-psbA and trnL-trnF, were easy to amplify and sequence and showed significant inter-specific genetic variability, making them potentially useful DNA barcodes for *** best performing single loci were the rbcL and rpoC1 coding *** loci showed equivalent performance and combinations of them did not greatly increase their discrimination *** addition, phylogenies generated from each of the separate regions and multi-locus combinations by using best-fit and Kimura 2.parameter models were compared, but no significant difference was found.

An efficient transformation method mediated by PEG-protoplasts was developed for the newly commercial edible mushroom Pleu- rotus nebrodensis. Two plasmids were used to co-transform protoplasts of P. nebrodensis. One plasmid is pAN7-1 containing a positive selectable marker gene hph conferring hygromycin B resistance. Another plasmid is pBlue-GFP containing a reporter gene gfp conferring green fluorescent protein. PCR and Southern blot analysis showed that hph gene or/and gfp gene were integrated into the genome of P. nebrodensis transformants. The transformation efficiency of the positive selectable marker gene hph was 3 transformants per microgram of plasmid pAN7-1 DNA, which was about 30 times higher than that previously reported in thoroughly studied Pleurotus species such as Pleurotus ostreatus. The transformation efficiency of the reporter gene gfp was 9 transformants per microgram of plasmid pBlu-GFP DNA. The co-transformation efficiency was 2..68%. This is the first report that a "reporter" gene, green fluorescent protein gene can be successfully stably expressed in this Pleurotus species.

A new kind of nano-crysta cellulose (NCC) prepared from natural cotton fiber has been obtained by the method ofacid hydrolysis. Compared to most other nanophase materials that derive from inorganic materials, our products are preparedfrom natural cotton fibers. The products are of spherical shape with mixed crystal forms of cellulose I and II. The preparationconditions determine the properties of the products. Prior treatment is a critical procedure. The properties of the products arealso strongly affected by such conditions as the kinds of acids used, the ratio of the acid mixture, the acid concentration, theultrasonic agitation time and hydrolysis temperature. The number average molecular weight of NCC is determined by gelpermeation chromatography (GPC). The particle size and shape were determined by transmission electron microscopy(TEM). X-ray diffraction was used to detect the crystallinity and average crystallite size of the panicle.

Water pricing towards sustainability of water resources: a case study in Beijing

The role of water pricing for managing water resources is widely recognized in many areas of the world because of the increasing scarcity of water resources, a high competition between water uses and environmental degradation. Based on the analysis of cost of water, this paper explores which types of cost should be reflected in the water pricing enhancing the sustainability of water resources. The principle of full cost pricing in which the cost should include supply cost, opportunity cost and externalities is proposed as a means to achieve the sustainability of water resources. In a case study of Beijing, low water price is analyzed as one reason for unsustainable water consumption. Thus water pricing justified is necessary and pressing. It is proposed to justify water price in phased manner and eventually towards full cost pricing. The assessment of impacts on water resources by raising water price shows water pricing could alleviate the conflict between water supply and demand. This paper concludes that water pricing can play an effective role in enhancing the sustainability of water resources in Beijing.

Based on the characteristics of Thin slab casting and rolling (TSCR) process,using test methods such as SEM,EDS and measuring content of inclusions, the influencing factor of controlling inclusions during producing middle and high carbon steel was studied. The technologies of electric furnace smelting, LF refining and controlling inclusions of tundish were proposed. The inclusions control level of middle and high carbon steel in which the carbon content is 0.3-1.0%, The level reached and even exceeded the level of like product producing with conventional process at home and abroad.
