Inhibitory effect of ginsenoside rb1 on calcineurin signal pathway in cardiomyocyte hypertrophy induced by prostaglandin F2α

Objective:Neutrophil gelatinase-associated lipocalin(NGAL),a member of the lipocalin family,was implicated in the progression of several tumor ***, experimental evidence linking NGAL expression to the development of esophageal squamous cell carcinoma(ESCC) is ***,some experiments were designed to explore the role of NGAL in the progression of ***: NGAL was stably overexpressed in the human ESCC cell line EC109 which normally expresses only low levels of NGAL, and these cells were used to measure cell proliferation by MTT assay and colony formation assay,the ability of cell migration and invasion of Matrigel were evaluated by cell invasiveness and chamber-migration *** was used to measure matrix metalloproteinase-9(MMp-9) activity. Western blotting was used to assess phosphorylation of ErK,and immunofluorescence was used to visualize the Factin cytoskeleton.A mouse model established by injecting EC109 cells expressing NGAL into the tail vein of severe combined immunodeficiency(SCID) mice was employed to address the effect of NGAL on body weight,food intake, rate of cachexia,and survival of ***:NGAL overexpression increased the proliferation,migration,invasiveness, and matrix metalloproteinase-9(MMp-9) activity of EC109 cells and triggered rearrangement of the cytoskeleton and activation of ErK in the *** vivo experiments, many SCID mice inoculated with NGAL-transfected cells displayed a severe weakness,a hunchback,reduction in food intake and weight loss,all symptoms of cachexia,before dying. But,fewer SCID mice injected with the control cells appeared to have this cachexia-like syndrome,and there were significantly different in body weight loss,the rates of cachexia and the survival of ***:These data suggest a model for ESCC in which NGAL acts through the ErK/MApK pathway to stimulate rearrangement of the cytoskeleton and alteration of cell proliferation,migration,and invasiveness,which results in a

Aim:To examine the antihypertrophic effect of ginsenoside rb1（rb1）induced byprostaglandin F2α(pGF2α)in vitro and to investigate the possible mechanismsinvolved in the calcineurin（CAN）signal transduction ***:Thecardiomyocyte hypertrophy induced by pGF2αand the antihypertrophic effect ofrbiwere evaluated in primary culture by measuring the cell diameter,proteincontent,and atrial natriuretic peptide（ANp）mrNA *** and CaNmrNA expressions,CaN and its downstream effectors NFAT3and GATA4 proteinexpressions,and the intracellular free Ca2+concentration([Ca2+]i)were assayedby rT-pCr,Western blot,and fluorescent determination using Fura 2/AM,***:pGF2α（100 nmol/L）significantly increased the cardiomyo-cyte diameter,protein content and[Ca2+]i,and promoted ANp,CaN mrNA,andCaN/NFAT3/GATA4protein expressions,which were inhibited by either rb1in aconcentration-dependent manner（50,100,and 200μg/mL）or L-arginine（1 mmol/L）.NG-nitro-L-arginine-methyl ester,a nitric oxide synthase inhibitor,could abolishthe effects of L-arginine,but failed to change the effects of rb1in the ***:The present data implicate that rb1attenuates cardiachypertrophy,the underlying mechanism may be involved in the inhibition of theCa2+-CaN signal transduction pathway.

Aim: To investigate the anticancer effects and the molecular mechanisms of deguelin on human U937 leukemia cells, and to explore the underlying mechanism regulating nucleoporin 98 （Nup98） and nucleoporin 88 （Nup88） in vitro. Methods: The effects of deguelin on the growth of U937 cells were studied by MTT assay. The effect of deguelin on the cell cycle of U937 cells was studied by using a propidium iodide method. The localization of the nuclear pore complex proteins Nup98 and Nup88 was investigated by using immunofluorescence and immunoelectron microscopy. The expression of Nup98 and Nup88 in U937 cells was investigated by using flow cytometry and Western blot. results: The prolif eration of U937 cells was inhibited in the deguelin-treated group, with a 24-h IC50value of 21.61 nmol/L and a 36-h IC50value of 17.07 nmol/L. U937 cells treated with deguelin had reduced percentages of cells in the G0/G1phase, whereas cells accumulated in the S and G2/M phases. Nup88 and Nup98 were found on both the nuclar and cytoplasmic sides of the U937 cells by using immunofluorescence and immunoelectron microscopy. The expression of Nup98 was upregulated and that of the Nup88 protein was downregulated in U937 cells treated with deguelin. Conclusion: Deguelin is able to inhibit the proliferation of U937 cells by regulating the cell cycle such that cells are arrested at the S and G2/M phases, so that the proportion of cells in the G0/G1phase decreases. The antitumor effects of deguelin are related to upregulating the expression of Nup98 and downregulating the expression of Nup88 protein in U937 cells.

以往在研究由促癌物 12 o 十四烷酰佛波醇 13 乙酯 (12 o tetradecanoylphorbol 13 acetate,TpA)诱导的人永生化食管上皮细胞恶性变中基因的差异表达情况时 ,曾获得中性粒细胞明胶酶相关lipocalin (neutrophilgelatinase associatedlipocalin ,NGAL)等新的食管癌癌变相关基因 .为深入研究这些癌变相关基因在食管癌中以蛋白质蛋白质或蛋白质 DNA相互作用为基础的功能网络调控关系 ,建立了一个食管癌cDNA酵母杂交文库 .采用Trizol试剂从一种新的人食管癌细胞 (SHEEC ,由人永生化食管上皮细胞转化而来 )中提取细胞总rNA ,采用OligotexmrNAKit从细胞总rNA中制备polyA+ mrNA ,采用SuperScriptTM ChoiceSystemForcDNASynthesisKit合成cDNA ,以polyA+ mrNA为探针 ,化学发光法监测cDNA合成的质量 ,以 pGADT7为载体构建了SHEEC细胞的cDNA酵母杂交文库 .文库的滴度为 1 19× 10 9cfu/ml,重组片段大小主要集中在 0 5～6 0kb ,重组率为 5 0 % .在此基础上 ,应用酵母单杂交技术手段对该文库中的NF κB元件结合因子进行了筛选 ,在SD/ his/ leu/[+15mmol/L 3 AT ]缺陷培养基平板上共获得了约 36 0个单克隆 .按照平板上酵母单克隆直径的大小 ,选择直径大于 2mm者 91个 ,提取质粒进行酶切鉴定和一对一酵母单杂交验?

Aim:To investigate the effects of angiotensin receptor blocker (ArB) telmisartanon the expression and distribution of protein kinase C (pKC)-α in the kidneys ofdiabetic ***:Diabetic mice were induced with streptozotocin and agroup of them were randomly selected for treatment with *** 6weeks,the expression and localization of pKC-α in the renal cortex,and the outerand inner medulla were assessed by immunohistochemistry and semiquantitativeWestern *** addition,expressions of pKC-α,transforming growth factor-β1 (TGF-β1),and vascular endothelial growth factor (VEGF) in glomeruli weremeasured by semiquantitative ***:Diabetic and nor-mal mice showed similar distributions of pKC-α in the *** expression ofpKC-α was found in glomeruli,epithelial cells of proximal tubules,and medullary-collecting duct,while not in the medullary and cortical thick ascending limb,andwas different in the epithelial cells of proximal tubules of diabetic nephropathy(DN) mice,pKC-α was mostly translocated from the basement membrane to theapical membrane,whereas it was largely translocated from the apical membrane tothe basement membrane in epithelial cells of the inner medullary-collecting *** blotting detected increased expression of pKC-α in the renal cortex andouter medulla,but not in the inner medulla of DN *** expressions ofpKC-α,TGF-β1,and VEGF were shown in the glomeruli of DN mice,where pKC-αexhibited a correlation to VEGF,but no correlation to TGF-β*** telmisartanattenuated alterations of pKC-α as mentioned earlier in the DN ***:Our findings suggest that pKC-α may play a role in the pathogenesis of DN,andthat the nephroprotective effects of ArB telmisartan may be partly associatedwith its influence on pKC-α.

The phylogenetic origin of Beckmannia remains *** genus has been placed within the Chlorideae,Aveneae(Agrostideae),poeae,or treated as an isolate lineage,*** the present study, we used nuclear internal transcribed spacer(ITS)and chloroplast trnL-F sequences to examine the phylogenetic relationship between Beckmannia and those genera that have assumed to be *** the basis of the results of our studies,the following conclusions could be drawn:(i)Beckmannia and Alopecurus are sister groups with high support;and(ii)Beckmannia and Alopecurus are nested in the poeae clade with high *** results of our analysis suggest that Beckmannia should be placed in poeae.
