Study on Tibetan Chicken embryonic adaptability to chronic hypoxia by revealing differential gene expression in heart tissue

yellow^0,a marker for low body weight in Drosophila melanogaster

Marker-assisted selection(MAS) is an important modern breeding technique,but it has been found that the effect of the markers for quantitative trait loci(QTL) is inconsistent,leading in some cases to MAS failure and raising doubts about its *** the model organism Drosophila melanogaster was employed to study whether an effective marker could be found and applied to *** crossed the stock carrying the y0 marker(a recessive mutation allele of the yellow gene on the X chromosome) with three other stocks carrying corresponding wild-type markers in an F2 design,and found that the y0 marker was in significant association with low body weight(P20 generations of marker-assisted introgression(MAI),and constructed the introgression stock y0(OR)20 in which body weight decreased by 13% and 7%,in female and male,respectively,compared to the wild stock(P20,indicating that the QTL for low body weight is located within this less-than-1.5 cM *** characteristics of the y0 marker by PCR amplification and Southern blotting revealed that yellow gene was deficient in the y0 stock,leading to disappearance of melanin from the cuticle and probably influencing the developmental *** above results confirmed the existence of effective QTL markers applicable to MAS breeding schemes,and their potential application in breeding new stocks.

Activity Calculation in Complex Metallurgical Molten Slag Systems Based on Regular Solution Model

The activity of FetO is very important in ironmaking and steelmaking process. In order to predict the activ- ity of Fe, O and optimize the operation conditions in ironmaking and steelmaking process, by application of regular so lution model in molten slag systems, FeO-Fe2 O3-SIO2 ternary system, FeO-Fe2 O3-SiO2-CaO and FeO-Fe2 O3-SiO2-NiO quaternary systems have been studied by the chemical equilibrium between H2/H20 gas mixture and liquid slag con tained in solid iron. The values of interaction energy between cations concerning steelmaking slags have been deter- mined by application of ferric-ferrous iron equilibrium and iron-ferric iron equilibrium. And then the activity of Fe, O can be calculated. The results show that the relative error is 3.9% in FeO-Fe203-SiO2 system and 18% in FeO- Fe203-SiO2 CaO system. The prediction of activities of FetO in the systems are in good agreement with the measure- ments and the regular solution model is valid for predicting the activity of FetO in complex molten slags systems. The activity of Fe, O in FeO-Fe20a-NiO system have not been tested presently, and the calculated result can not be assessed.

Inducement of chromosome translocation with small alien segments by irradiating mature female gametes of the whole arm translocation line

Haynaldia villosa Schur. (syn. Dasypyrum villosum Candargy, 2n=14, VV) has been proved to be an important genetic resource for wheat improvement. The development of translocation with small alien chromosome segments, especially interstitial translocation, will be helpful for better utilization of its useful genes. Up to now, most of the reported Triticum aestivum – H. villosa translocation lines are involved in a whole arm or large alien fragments. In this paper, we report a highly efficient approach for the creation of small chromosome segment translocation lines. Before flowering, the female gametes of wheat-H. villosa 6VS/6AL translocation line were irradiated by 60CO-γ ray at 160 Rad/M dosage rate and three dosages (1600, 1920, 2240 Rad). Anthers were removed from the irradiated florets on the same day and the florets were pollinated with normal fresh pollens of T. aestivum cv. Chinese Spring after 2-3 days. Genomic in situ hybridization (GISH) at mitosis metaphase of root-tip cell of M1 plants was used to detect the chromosome structural changes involving 6VS of H. villosa. Among the 534 M1 plants screened, 97 plants contained small segment chromosome structural changes of 6VS, including 80 interstitial translocation chromosomes, 57 terminal translocation chromosomes and 55 deletion chromosomes. For the 2240 Rad dosage treatment, the inducement frequencies of interstitial translo-cation, terminal translocation and deletion were 21.02%, 14.01%, and 14.65%, respectively, which were much higher than those previously reported. The M2 seeds were obtained by backcrossing of 74 M1 plants involving 146 chromosomes structural changes of 6VS, and it was found that the structural aberrations in the M1 plants could be transmitted to their progenies. Irradiating mature female gametes of whole arm translocation is a new and highly efficient approach for creation of small segment chromosome struc-tural changes, especially for interstitial translocations.

Simulation study on the efficiencies of MOET nucleus breeding schemes applying marker assisted selection in dairy cattle

Microstructure and Mechanical Properties of Martensitic Stainless Steel 6Cr15MoV

Martensitic stainless steel containing Cr of 12% to 18%（mass percent） are common utilized in quenching and tempering processes for knife and cutlery steel. The properties obtained in these materials are significantly influ- enced by matrix composition after heat treatment, especially as Cr and C content. Comprehensive considered the hardness and corrosion resistance, a new type martensitic stainless steel 6Crl5MoV has been developed. The effect of heat treatment processes on microstructure and mechanical properties of 6Crl5MoV martensitic stainless steel is emphatically researched. Thermo-Calc software has been carried out to thermodynamic calculation; OM, SEM and TEM have been carried out to microstructure observation; hardness and impact toughness test have been carried out to evaluate the mechanical properties. Results show that the equilibrium carbide in 6Cr15MoV steel is M23 C6 car- bide, and the M23 C6 carbides finely distributed in annealed microstructure. 6Crl5MoV martensitic stainless steel has a wider quenching temperature range, the hardness value of steel 6Cr15MoV can reach to HRC 60.8 to HRC 61.6 when quenched at 1 060 to 1 100 ℃. Finely distributed carbides will exist in quenched microstructure, and effectively inhabit the growth of austenite grain. With the increasing of quenching temperature, the volume fraction of undis- solved carbides will decrease. The excellent comprehensive mechanical properties can be obtained by quenched at 1 060 to 1 100℃ with tempered at 100 to 150 ℃, and it is mainly due to the high carbon martensite and fine grain size. At these temperature ranges, the hardness will retain about HRC 59.2 to HRC 61.6 and the Charpy U-notch impact toughness will retain about 17.3 to 20 J. A lot of M23C6 carbides precipitated from martensite matrix, at the same time along the boundaries of martensite lathes which leading to the decrease of impact toughness when tempered at 500 to 540 ℃. Purification of a Novel Antibacterial Short Peptide in Earthworm Eisenia foetida

A novel antimicrobial short peptide was purified from earthworm (Eisenia foetida) by a five-step protocol including ammonium sulfate precipitation, ultrafiltration, DE-52 ion exchange chromatography, Sephadex G-10 column chromatography, and C-18 reversed-phase HPLC techniques. The purified peptide was applied to the MALDI-TOF MS to determine the molecular mass and was also subjected to TOF MS-MS analysis to determine the amino acid sequence. As a result, a novel antibacterial peptide, named OEP3121, was obtained, with the molecular mass of 510.8 Da and the sequence being “ACSAG”.

MANUFACTURING OF ALUMINUM/FLY ASH COMPOSITE WITH LIQUID REACTIVE SINTERING technology

The Al/fly ash composites are fabricated by liquid reactive sintering P/M process with fly ash particles as intensifying phases. The reactivity and newly formed phases during liquid sintering process have been analyzed by combing Thermochemicdl data base calculation and XRD characterization. The results show that some of constituents in fly ash have reacted with liquid aluminum so that the elemental Si, Fe, Ti as well as some amount of intermetallic compounds occur. The properties of aluminum/fly ash composites have been improved. With the fraction of fly ash increase, the composite density decreases; the hardness and the modulus of the composite increases, and the composite wear resistance are significantly increased. The fly ash reinforced composites represent a sort of low cost product with possible widespread applications in the automotive, small engine, and electromechanical machinery sectors.

The distribution of N and morphology of graphite in gray cast iron containing N were studicd by adopting the man-made specimens of Fe-C-Si-Mn alloys with some N added. The N adsorbed along the interfaces between graphite and matrix together with solid- solutioned in graphite is believed to be the principal cause of morpholongical variation of graphite.

A five-fold pig bacterial artificial chromosome library:a resource for positional cloning and physical mapping

A pig BAC library was constructed with genomic DNA from a male Erhualian pig. After partial digestion with HindⅢor BamHⅠthe fragments obtained were cloned into the pBeloBAC11 vector. The library consists of 184320 clones which stored in 480 pieces 384-well plates (20 plates per superpool). A two-step 4-dimension PCR screening system was established to screen the positive clones. An average insert size of 128 kb was estimated from 105 randomly isolated clones, which indicates that the library is more than five times of genomic coverage. For the demonstration of the probability to pick out any unique genes or DNA markers from the library, 10 single-copy genes were screened out and the positive clones were yielded between 1 and 8 with an average of 3.6. Positive superpools were obtained for 32 microsatellite markers selected from different regions of pig genome. The number of positive superpools for each marker varies from 1 to 9 with an average of 4.78. This BAC library provides an additional resource for pig physical mapping and gene identifica-
