Expression of calreticulin is associated with infiltration of T-cells in stage ⅢB colon cancer

AIM: To investigate the correlation between expression of calreticulin and infiltration of lymphocytes in stage ⅢB colon cancer. METHODS: Sixty-eight pathologically-confirmed speci-mens were obtained from stage ⅢB (T3N1M0) colon cancer patients who underwent radical resection between January 1999 and May 2002 at the Cancer Center of sun yat-sen university, guangzhou, china. Immuno-histochemical analysis was performed to show infiltration of lymphocytes and expression of calreticulin in colon cancer. Association between calreticulin expression, infiltration of lymphocytes, and 5-year survival rate of patients was assessed. RESULTS: The expression level of calreticulin was lower in cancer nest than in its adjacent normal epithelium since 61.8% (42/68) of the samples were stained with calreticulin in colon cancer. The expression of calreticulin in colon cancer was associated with the infi ltration of CD45RO+ cells rather than with that of CD3+ cells. In addition, the stronger expression of calreticulin and the higher infiltration of CD3+ and CD45RO+ cells in colon cancer were associated with the higher 5-year survival rate of patients. CONCLUSION: Expression of calreticulin is associated with infiltration of T-cells, which implies that a low expression level of molecular marker may represent a new mechanism underlying immune escape in colon cancer.

Experiments were performed on cats under chloralose and flaxedil. Evokedunit discharges recorded in contralateral somato-sensory cortex to noxiousstimulating superficial peroneal nerve (P-ED) were analysed quantitativelywith micro-computer using the method of stochastic point process for signalanalysis in order to study the effects of electro-acupuncture(EA)and morphineon non-time-lock response(N-TLR) and time-lock response (TLR) of nociceptiveunit discharges. N-TLR was shown by interspike interval mean function(ISIMF).TLR was shown by normalized cross-covariance function (NCCVF) reflectingthe relationship between stimulation and response. The results were testedstatistically.

Molecular Mechanism of Bovine Trabecular Meshwork Cells Apoptosis Induced by Dexamethasone and Protection by Pilocarpine

Purpose: To study the molecular mechanism of trabecular meshwork cells apoptosis induced by dexamethasone and the protection of pilocarpine. Methods: Determining mRNA expression with reverse transcription-polymerase chain reaction (RT-PCR), protein expression with Western blots and the percentage of apoptotic cells with fluorescent microscopy. Results: Dexamethasone up-regulated Fas proteins and affected Bax, caspase-8 and caspase-9 proteins in an action of first decrease then increase. Pre -treatment with pilocarpine decreased the four proteins expression, which were increased by dexamethasone. Pilocarpine self could decrease pro-apoptotic factors Bax, caspase-8 and caspase-9 proteins expression. Conclusion: Fas/FasL pathway participated in apoptotic process induced by dexamethasone in trabecular meshwork cells and the process was probably related with both caspase-8 and caspase-9 pathways. Vortex genesis over the Bay of Bengal in spring and its role in the onset of the Asian Summer Monsoon

Physical processes associated with onset of the 1998 Asian summer monsoon were examined in detail using multi-source datasets. We demonstrated that strong ocean-atmosphere-land interaction in the northern Indian Ocean and tropical Asian area during spring is a fundamental factor that induces the genesis and development of a monsoon onset vortex over the Bay of Bengal (BOB), with the vortex in turn triggering onset of the Asian summer monsoon. In spring, strong surface sensible heating over India and the Indochina Peninsula is transferred to the atmosphere, forming prominent in situ cyclonic circulation, with anticyclonic circulations over the Arabian Sea and northern BOB where the ocean receives abundant solar radiation. The corresponding surface winds along the North Indian Ocean coastal areas cause the ocean to produce the in situ offshore currents and upwelling, resulting in sea surface temperature (SST) cooling. With precipitation on the Indochina Peninsula increasing from late April to early May, the offshore current disappears in the eastern BOB or develops into an onshore current, leading to SST increasing. A southwest-northeast oriented spring BOB warm pool with SST >31°C forms in a band from the southeastern Arabian Sea to the eastern BOB. In early May, the Somali cross-equatorial flow forms due to the meridional SST gradient between the two hemispheres, and surface sensible heat over the African land surface. The Somali flow overlaps in phase with the anticyclone over the northern Arabian Sea in the course of its inertial fluctuation along the equator. The convergent cold northerlies on the eastern side of the anticyclone cause the westerly in the inertial trough to increase rapidly, so that enhanced sensible heat is released from the sea surface into the atmosphere. New ancient DNA sequences suggest high genetic diversity for the woolly mammoth (Mammuthus primigenius)

Partial DNA sequences of cytochrome b gene (mtDNA) were successfully retrieved from Late Pleistocene fossil bone of Mammuthus primigenius collected from the Xiguitu County ( Yakeshi), Inner Mongolia Autonomous Region and from Zhaodong, Harbin of Heilongjiang Province in northern china. Two ancient DNA fragments (109 bp and 124 bp) were authenticated by reproducible experiments in two different laboratories and by phylogenetic analysis with other Elephantidae taxa. Phylogenetic analysis using these sequences and published data in either separate or combined datasets indicate unstable relationship among the woolly mammoth and the two living elephants, Elephas and Loxodonta . In addition to the short sequences used to attempt the long independent evolution of Elephantidae terminal taxa, we suggest that a high intra-specific diversity existed in Mammuthus primigenius crossing both spatial and temporal ranges, resulting in a complex and divergent genetic background for DNA sequences so far recovered. The high genetic diversity in the extinct woolly mammoth can explain the apparent instability of Elephantidae taxa on the molecular phylogenetic trees and can reconcile the apparent paradox regarding the unresolved Elephantidae trichotomy.

Non-invasive Prenatal Gene Diagnosis: Progress through Cell-free Fetal DNA and RNA in Maternal Plasma and Urine

Non-invasive prenatal gene diagnosis has been developed rapidly in the recent years, and numerous medical researchers are focusing on it. Such techniques could not only achieve prenatal diagnosis accurately, but also prevent tangential illness in fetuses and thus, reduce the incidence of diseases. Moreover, it is non-invasive prenatal gene diagnosis that prevents potential threaten and danger to both mothers and fetuses. Therefore, it is welcomed by clinical gynecologist and obstetrian, researchers of medical genetics, and especially, pregnancies. This review article touches briefly on the advanced development of using cell-free DNA, RNA in maternal plasma and urine for non-invasive prenatal gene diagnosis.

Hemorrhagic activity and mechanism of FIIa,a fibrinolytic enzyme from Agkistrodon acutus venom

AIM: To study the local hemorrhagic activity of a fibrinolytic enzyme (FIIa) from Agkistrodon acutus venom and its mechanism. METHODS: The local hemorrhagic activity was determined by subcutaneous injection on the back of mouse. The effects of FIIa on factor X, prothrombin, gelatin, and collagen were visualized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Platelet aggregation assays were performed in rat platelet- rich plasma (PRP). Human umbilical vein endothelial cells (HUVEC) were cultured and passaged in complete M199 medium. Cell viability and nuclear morphology change were determined by fluorescein diacetate (FDA) staining and Hoechst 33258 staining, respectively. RESULTS: The minimum hemorrhagic dose (MHD) of FIIa was 89 μg. In vitro, FIIa (0.25 g/L) degraded factor X, prothrombin, collagen, and gelatin, and dose-dependently (0.25, 0.50, 0.75, and 1.00 g/L) inhibited the platelet aggregation induced by ADP in rat PRP. When HUVEC in culture treated with FIIa, HUVEC showed detachment in a dose-dependent manner, but no apoptosis sign was observed. CONCLUSION: FIIahad local hemorrhagic activity, and the mechanism was related to the degradation of factor X, prothrombin, gelatin, and collagen, the inhibition of ADP-induced platelet aggregation, and inducement of HUVEC detachment.2Now in Beijing universityof Chinese Medicine,Beijing 100029,china.3Correspondence to Dr Xiu-xia LIANG.
