AIM: To investigate the relationship betweenhyposmotic membrane stretch and muscarinic receptoragonist-induced depolarization of membrane potentialin antral gastric circular myocytes of ***: Using whole cell patch-clamp techniquerecorded membrane potential and current in singlegastric myocytes isolated by collagena ***: Hyposmotic membrane stretch hyperpolarizedmembrane potential from -60.0mV±1.0mV to -67.9mV±1.0mV. TEA (10mmol/L), a nonselective potassiumchannel blocker significantly inhibited hyposmoticmembrane stretch-induced hyperpolarization. After KCIin the pipette and NaCI in the external solution werereplaced by CsCI to block the potassium current,hyposmotic membrane stretch depolarized the membranepotential from -60.0 mV±-1.0mV to -44.8 mV±2.3mV(P＜0.05), and atropine (1 pmol/L) inhibited thedepolarization of the membrane potential. Muscarinicreceptor agonist Carbachol depolarized membranepotential from -60.0mV±1.0mV to -50.3 mV±0.3mV(P＜0.05) and hyposmotic membrane stretchpotentiated the depolarization. Carbachol inducedmuscarinic current (Icch) was greatly increased byhyposmotic membrane ***: Hyposmotic membrane stretchpotentiated muscarinic receptor agonist-induceddepolarization of membrane potential, which is relatedto hyposmotic membrane stretch-induced increase ofmuscarinic current.

Lancaster Surecrop (LSC) germplasm in inbreds of the Lancaster heterotic group in china was mainly provided by the two inbreds Mol7 and Oh43. Furthermore, the source of LSC germplasm was severely restricted in two inbreds, C103 and Oh40B. The contents of LSC germplasm are 50% or less in inbreds of the group, with the existence of more than 20 non-LSC germplasms that results in the abundant genetic diversity in the group. Most inbreds of Mol7 subgroup contain 25% - 50% LSC, 12.5% - 25% Reid Yellow Dent (RYD) and 12.5%- 25% Iowa Goldmine germplasms in the ratio of 2:1:1, and most inbreds of Zi330 subgroup contain 12.5% - 25% LSC, 6.25% - 12.5% RYD, 6.25% - 12.5% Minnesota # 13 and 25% - 50% Creole germplasms in the ratio of 2:1:1 4. AH the facts indicated that the germplasm base of the Lancaster heterotic group has stupendously changed and evolved away from actual LSC germplasm.

Advantages of breeding schemes using genetic marker information and/or multiple ovulation and embryo transfer(MOET) technology over the traditional approach were extensively evaluated through *** yield was the trait of interest and QTL was the genetic marker *** dairy cattle breeding scenarios were considered,i.e.,traditional progeny testing breeding scheme(denoted as STANPT),GASPT scheme including a pre-selection of young bulls entering progeny testing based on their own QTL information,MOETPT scheme using MOET technology to generate young bulls and a selection of young bulls limited within the full-sib family,GAMOPT scheme adopting both QTL pre-selection and MOET technology,COMBPT scheme using a mixed linear model which considered QTL genotype instead of the BLUP model in GAMOPT,and three non-progeny testing schemes,*** MOET,GAMO and COMB schemes,corresponding to MOETPT,GAMOPT and COMBPT with progeny testing being part of the *** were selected based on their breeding value which was estimated under an animal model *** selection over 17 years was performed in the simulations and 30 replicates were designed for each *** influences of using QTL information and MOET technology on favorable QTL allele frequency,true breeding values,polygenetic breeding values and the accumulated genetic superiority were extensively evaluated,for five different populations including active sires,lactating cows,bull dams,bull sires,and young *** results showed that the combined schemes significantly outperformed other approaches wherein accumulated true breeding value *** difference between schemes exclusively using QTL information or MOET technology was not *** STANPT scheme was the least efficient among the 8 *** schemes using MOET technology had a higher polygenetic response than others in the 17th *** increases of frequency of the favorable QTL allele varied more greatly across t

Aim: To study effects of arachidonic acid （AA） and its metabolites on the hyposmotic membrane stretch-induced increase in calcium-activated potassium currents (IKCa) in gastric myocytes. Methods: Membrane currents were recorded by using a conventional whole cell patch-clamp technique in gastric myocytes isolated with collagenase. Results: Hyposmotic membrane stretch and AA increased both IK（Ca）and spontaneous transient outward currents significantly. Exogenous AA could potentiate the hyposmotic membrane stretch-induced increase in IK（Ca）The hyposmotic membrane stretch-induced increase inK（Ca）was significantly suppressed by dimethyleicosadienoic acid （100 μmol/L in pipette solution）, an inhibitor of phospholipase A2. Nordihydroguaiaretic acid, a lipoxygenase inhibitor, significantly suppressed AA and hyposmotic membrane stretch-induced increases in IK（Ca）. External calcium-free or gadolinium chloride, a blocker of stretch-activated channels, blocked the AA-induced increase in IK（Ca）significantly, but it was not blocked by nicardipine, an L-type calcium channel blocker. Ryanodine, a calcium-induced calcium release agonist, completely blocked the AA-induced increase inK（Ca）; however, heparin, a potent inhibitor of inositol triphosphate receptor, did not block the AA-induced increase in IK（Ca）. Conclusion: Hyposmotic membrane stretch may activate phospholipase A2, which hydrolyzes membrane phospholipids to ultimately produce AA; AA as a second messenger mediates Ca2+influx, which triggers Ca2+-induced Ca2+release and elicits activation of IK（Ca）in gastric antral circular myocytes of the guinea pig.

Substituted benzyl azids were synthesized through the reaction of substituted benzyl chloride and sodium azid, which subsequently underwent cyclization with ethyl propiolate and amidation to give thirteen 1-substituted benzyl-N-substituted-1, 2, 3-triazole-4-formamide derivatives (3a-3m) . The structure of the synthesized compounds was confirmed by IR, 1H-NMR, MS and elemental analysis. Their ant icon vulsant activity against maximal electrolshock (MES) induced seizure was tested and the result showed that all these compounds possess anticonvulsant activity in different degrees. Among those, the compounds containing chloro atoms on the phenyl ring were less potent in anticonvulsant activity, while introducing one or two fluorin atoms on benzyl system increased its activity. Furthermore, their activity decreased when there was substituent on the nitrogen atom of carboxamide, and the larger the substituent, the lower the activity.

Skin graft rejection is a typical cellular immune response,mainly mediated by T *** lymphocyte associated antigen 4-immunoglobin (CTLA4-Ig) extends graft survival by blocking the T cellco-stimulation pathway and inhibiting T cell *** investigate the efficacy of CTLA4-Ig in prolong-ing skin graft survival,human CTLA4-Ig (hCTLA4-Ig) was engineered to overexpress in mouse skin bytransgenesis using the K14 *** transcription-polymerase chain reaction (RT-PCR) and West-ern blot assay indicated that the expression of CTLA4-Ig remained skin-specific and relatively constantcompared to the internal control protein,AKT,through seven *** presence and concentrationof the hCTLA4-Ig protein in transgenic mouse sera was determined by enzyme-linked immunosorbent assay(ELISA),and the results indicated that the serum CTLA4-Ig concentration also remained constant *** of transgenic mouse skins grafted onto rat wounds was remarkably prolonged com-pared to that of wild-type skins from the same mouse strain,and remained comparable among all *** suggested that the bioactive hCTLA4-Ig protein was stably expressed in transgenical micethrough at least seven generations,which was consistent with the stable skin-specific CTLA4-Ig *** results demonstrated that the transgenic expression of hCTLA4-Ig in skin driven by the K14 promoterremained constant through generations,and a transgenic line can be established to provide transgenic skinwith extended survival reproducibly.

The Rice dwarf virus(RDV)P7 structural protein is the key protein in the RDV *** P7 protein was digested partially or completely by Staphylococcus aureus V8 protease and/orPseudomonas fragi Asp-N *** molecular mass and the N-terminal amino acid sequence of thepolypeptide fragments of the P7 protein were determined by SDS-PAGE and the Edman degradation method,*** the polypeptides were located in the deduced amino acid sequence of the RDV P7 proteinbased on the nucleotide sequence information,with the knowledge of the specific cleavage sites of theStaphylococcus aureus V8 and Pseudomonas fragi Asp-N protease,and the two RNA-binding domains in theP7 protein were *** 1 was located in the residue 128-249 containing 122 amino acids anddomain 2 was located in the residue 325-355 containing 31 amino ***,these two domains may playan important role in the virus particle assembly by contributing to the packaging of viral dsRNAs inside *** two domains may be novel RNA-binding domains,because no amino acid sequences highlysimilar to the conservative sequences of known dsRNA-binding domains reported so *** similaritybetween the motif of domain 1 and the motif of the DNA-binding protein suggests that the DNA-bindingactivity of the RDV P7 protein may be due to this *** similarity between the motif of domain 1 andthe motif of the RNA polymerase domain suggests that the P7 protein may also play a role in RNA synthesis,besides its function in the assembly and subsequent packaging of viral dsRNA into core particles.
