Construction,Expression,and Characterization of a Recombinant Annexin B1-Low Molecular Weight Urokinase Chimera in Escherichia coli

To produce a thrombi-targeting plasminogen activator, low molecular weight single-chain urokinase gene (scuPA32k) was spliced with the full-length cDNA of annexin B1 gene (anxB1) by overlap extension method. The fused gene anxB1scuPA was ligated into pET28a vector, transformed into E. coli BL21-RIL, and then induced to express under the control of T7 promoter. The AnxB1ScuPA protein ex- pressed amounted to 22% of the total bacterial proteins. The product was refolded, and then purified by using DEAE Sepharose fast flow ion-exchange column and Superdex S-200 gel-filtration column. HPLC analysis revealed that the final purity is about 95%. The specific activity of AnxB1ScuPA, measured as amidolytic activity, reached 100,000 IU/mg. It had a similar S2444 catalytic efficiency (kcat/Km) to ScuPA32k, and also showed high activated-platelet membrane-binding activity and anticoagulant activity, indicating that the chi- mera fully retained the components of enzymatic and membrane-binding activities of the parent molecules. In vivo test revealed that, the dogs administered with AnxB1ScuPA had less reperfusion time, higher reperfu- sion ratio, and less bleeding effects than those with urokinase. These findings indicated that AnxB1ScuPA might have advantages over current available thrombolytic agents.

The Method of Single Peak Fourier Analysis of Measuring the Size of Mosaic Block and Microstrain by X-ray Diffraction

The values of mosaic block size and microstrain measured by the method of Voigt function are sometimes largely erroneous and difficult to be revised. In this paper, the causes of the 'hook effect' which inevitably exists in the method of Fourier analysis are explained; then the method for resolving this effect is put forward; and finally the method of hypothetical function is used to simplify the method of the Fourier analysis from multi-peak to single-peak. By introducing the parameter m, this method can not only indicate the degree of errors, but also revise them as well, it is simple, clear and able to accomplish the intricate pattern calculation quickly by the aid of computer. In the end, we argue the identity between method of the approximate function and the Fourier analysis, and give a more accurate proportional coefficient k.

Second mode unstable disturbance measurement of hypersonic boundary layer on cone by wavelet transform

Experimental investigation of hypersonic boundary layer instability on a cone is performed at Mach number 6 in a hypersonic wind tunnel. Time series signals of instantaneous fluctuating surface-thermal-flux are measured by Pt-thin-film thermocouple temperature sensors mounted at 28 stations on the cone surface in the streamwise direction to investigate the development of the unstable disturbance. Wavelet transform is employed as a mathematical tool to obtain the multi-scale characteristics of fluctuating surfacethermal-flux both in the temporal and spectrum space. The conditional sampling algorithm using wavelet coefficient as an index is put forward to extract the unstable disturbance waveform from the fluctuating surface-thermal-flux *** generic waveform for the second mode unstable disturbance is obtained by a phase-averaging technique. The development of the unstable disturbance in the streamwise direction is assessed both in the temporal and spectrum space. Our study shows that the local unstable disturbance detection method based on wavelet transformation offers an alternative powerful tool in studying the hypersonic unstable mode of laminar-turbulent transition. It is demonstrated that, at hypersonic speeds, the dominant flow instability is the second mode, which governs the course of laminar-turbulent transition of sharp cone boundary layer.

OBSERVATIONS ON DEFORMATION BEHAVIOR OF HIGH PERFORMANCE FIBERS BY POLARIZING OPTICAL MICROSCOPY

By means of polarizing optical microscopy (POM), deformation behavior of four kinds of fibers, i.e, ultra high molecular weight polyethylene (UHMW-PE) fiber, polyvinyl alcohol (PVA) fiber, polyethylene terephthalate (PET) fiber, and wholly aromatic (rho-hydroxybenzoic acid/2-hydroxy-6-naphthoic acid) copolyester [P(HBA/HNA)]/PET (ACPET blend) fiber, in axial compression, axial impacting, and bending was observed. In compression, kink bands formed at an angle of 55-60 degrees ro the fiber axis in 10-times-drawn UHMW-PE fiber, 75-80 degrees in 40-times-drawn sample, 80 degrees in PVA fiber, and 90 degrees in the ACPET blend fiber. In impacting and bending, band angles of UHMW-PE, PVA and PET fibers are nearly the same as those formed in compression, indicating that slip systems do not change. For any of the four kinds of fiber, band spacing exhibits great differences in compression, in impacting, and in bending, which may be attributed to the differences in the degrees of strain or stress concentration.

This review summarizes the labeling technology and applications of fluorescent dye probe in biology,especially the characteristics,modification and applications of cyanine *** on the currently available modification methods of fluorescent dye probe,we discuss the studies of enhancing the water-solubility,improving the degree of biocompatibility and target-labeling,increasing the sensitivity and decreasing the toxicity of fluorescent *** also give a brief introduction on the modification method,that the fluorescent dye is directly introduced onto the cell surfaces by amine derivatives or azides to intensify the transferring information of aberrant *** suggest that fluorescent dye modified with chitosan oligosaccharide can obviously increase the degree of biocompatibility and target-labeling,and decrease the degree of toxicity.

Two-photon spectroscopic behaviors and photodynamic effect on the BEL-7402 cancer cells of the new chlorophyll photosensitizer

The spectroscopic properties of a new chlorophyll derivate photosensitizer (CDP) are studied under the excitation wavelengths at 800 and 400 nm using femtosecond pulses from a Ti: sapphire laser. The damaging effect of CDP on the BEL-7402 cancer cells is also investigated upon two-photon illumination at 800 nm. The normalized fluorescence spectra of CDP in tetrahydrofuran (THF) show that two-photon and one-photon spectra have the same distributions and the same emission bands (675 nm). The lifetimes of two- and one-photon induced fluorescence of this molecule are of the order of 5.0 ns. By comparing the data it is shown that there is some difference between the two lifetimes, but the difference is less than one nanosecond. The two-photon absorption cross section of the molecule is also measured at 800 nm and estimated as about σ'_2 ≈ 31.5x10^(-50)cm^4·s·photon^(-1). The results of two-photon photodynamic therapy (TPPDT) tests show that CDP can kill all of the tested cancer cells according to the usual Eosine assessment. Our results indicate that the two-photon-induced photophysical, photochemical and photosensitizing processes of CDP may be basically similar to those of one-photon excitation. These behaviors of the sample suggest that one may find other possible methods to estimate some photosensitizers' effects in details such as their distribution in cells and the reactive targets of the sub-cellular parts of some tumor cells via two-photon excitation techniques.

Randomization of protein encoding gene during species evolution

Employing chi_square test, the authors observed the bias of 4 kinds of bases’ locating in 3 codon positions in gene sequence, and proved that there is a positive correlation between higher chi_square value and protein coding structure. By this, the observation that genes undergo progressive reduction in chi_values along evolution direction will reflect the progressive randomization of gene structures and the loss of the characteristics of protein coding sequences. The result proposed that during the evolution of life, genes are unexpectedly degenerating.

Electroabsorption spectrum of substituted polyphenylacetylenes

Electroabsorption (EA) measurements on thin films of poly[ o (trimethylsilyl) phenylacetylene] and poly[ o (trimethylsilylmethyl) phenylacetylene] have been performed. The spectral change induced by an exernal electric field (EA spectra) has been observed, which shows the similar shape of the second derivative of the optical absorption spectra. The change in the absorption Δ α is obtained from the EA spectrum, and Δ n is calculated through Kramers_Krong transformation. The optical properties of substituted polyphenylacetylenes (PPAs) were discussed.

Numerical modelling of sand beach evolution around coastal structures

A systematic study of waves, nearshore currents and sediment transport related to the coastal evolution of sand beach under the action of wave, has been carried out. On the basis of the re sults, a numerical model of sand beach evolution around coastal structures has been established. To pre dict the bed topography, the sediment transport rates of bedload and suspended load under the action of wave and nearshore current as well as the effects of wave on the sediment are considered. The sand beach evolutions caused by a breakwater and a settled ship near the shore are simulated.
