S-nitrosylation has been brought forward to be a prototypic redox-based post-translational modification in parallel with O-phosphorylation. However, the relationship between S-nitrosylation and cell redox status and the details of the S-nitrosylation mechanism involved in the cellular processes remain to be elucidated. In the present study, we investigated the role of S-nitrosylation in the susceptibility of glutathione (GSH) -depleted SH-SY5Y to S-nitrosoglutathione (GSNO) insults. Our results showed that: (1) D,L-buthionine-[s,r]-sulfoximine (BSO) greatly increased the susceptibility of SH-SY5Y cells to GSNO’s insult, and it is intriguing that the total cellular S-nitrosothiols (SNOs) increased significantly in the process. (2) The increased SNOs were primarily contributed by the S-nitrosylated proteins (PrSNO);moreover, the accumulation of PrSNO was dependent on the degree of oxidative status of the cells. (3) Reversible modification of the thiols appeared to mediate the susceptivity of the cells to GSNO’s insults, and reactive oxygen species (ROS) mediated the process. These results deepened our understanding of the S-nitrosylation mechanism in the redox-related physiological and pathological processed.

Objective In the rabbit retina,orientation-selective ganglion cells(OSGCs)were discovered in *** orientation selectivity results from an incomplete antagonistic surround along the preferred orientation. Pharmacological studies confirmed that the inhibition,originated from amacrine cells,shaped the orientation selectivity of retinal ganglion *** in the mouse retina,there is no information about the OSGCs,that is why we try to identify them and characterized their physiological,morphological and pharmacological ***(1) On-cell and whole-cell patch clamp recording were performed to indentify OSGCs and to examine synaptic inputs.(2) Pharmacological experiments were carried out to check which kinds of neurotransmitters are essential for orientation ***(1)In the mouse retina,ganglion cells classed into B3 and Dl subtypes have orientation selectivity.(2)These cells preferred either vertical or horizontal orientation.(3)The orientation selectivity can be reduced apparently by picrotoxin,which is an antagonist of GABA and GABA *** B3 and D1 ganglion cells,who respond to onset and termination of flashing spot,have orientation *** of them are sensitive to GABA blocker.

＜正＞Objective This study represents the most complete morphological classification of mouse retinal displaced ganglion cells（DGCs）in the inner nuclear layer available to date and provides a foundation for further understanding of the correlation of physiology and morphology and ganglion cell function with genetically manipulated ***（1）Flurogold was injected in two adjacent sites or one left site at the SC and the soma of DGCs in the mouse retina was stained by retrograding with Flurogold.（2）The morphology of DGCs was revealed by microinjection with Lucifer yellow.（3）The diameter of soma and dendritic field and distance from soma to the optic nerve were ***（1）The diameter of soma（or dendritic field）and distance from soma to the optic nerve were irrelevant.（2）The DGCs were classified into five groups based on their soma size,dendritic field size,and pattern and level of ***（1）It was proved that all morphological types of GCs in the GCL also existed in the ENL.（2）For monostratified DGCs,there is similar distribution of four morphological types in contralateral projection group and significant preference of B and C types in ipsilateral projection group.

＜正＞All animals are able to perceive space and *** behaviors in space and physiological processes fluctuate to adapt approximately to the 24-hour day/night cycle created by the self-rotation of our *** mammals including humans,this circadian biorhythm is entrained mainly by daily changes in ambient luminance detected by newly-found intrinsically photoreceptive retinal ganglion cells,which project to several brain regions including the hypothalamic suprachiasmatic nucleus and the thalamic lateral geniculate *** multiunit and brain slice recordings have revealed a circadian oscillation of suprachiasmatic activity,but our knowledge is completely lacking about whether and（if yes）how single thalamic cells would synchronize their firing rates to external light/dark cycles. Here we show by single-cell recording from the pigeon that there exist light-activated and light-inhibited neurons in the nucleus opticus principalis thalami,homologue of the lateral geniculate *** photosensitive thalamic neurons not only change firing rates in synchrony with the luminance level of light/dark cycles with various periods applied to the viewing eye but also memorize their own entrained firing pattern and replay spiking rhythmically for up to hours under constant photic *** light that maximally activates intrinsically photoreceptive retinal ganglion cells is much more potent than achromatic light for photoentrainment of thalamic *** cells as master clocks also possess such photoresponse *** results shed new light on the neural principle with which external light/dark cycles are translated into an internal rhythm in the organism and strongly support involvement of luminance encoding thalamic cells in biological rhythmicity.
