Optimization of transfection efficiency of small interfering RNA in purified human prolactinoma cells

Background Control of hypersecretion of certain hormones is one of the key targets in the treatment of pituitary adenomas. RNA interference has been shown to inhibit protein expression, and thus it may represent a promising method for the treatment of pituitary adenomas. In the present study, transfection efficiency of small interfering RNA （siRNA） was optimized in human prolactinoma cells. Methods First, a method was optimized to extract highly purified human prolactinoma cells in vitro. The extracted cells were verified to retain the physiological features of prolactin （PRL） secretion. Second, three conditions for siRNA transfection were tested by the evaluation of transfectfon efficiency and cell viability. The proper transfection condition was verified for human prolactinoma cells. Third, the siRNA for prolactin was transfected into the human prolactinoma cells, and the suppression of PRL mRNA was evaluated by quantitative real-time reverse transcription-PCR. Results The siRNA of 100 pmol with Lipofectamine 2.00 of 5 μl for 1×10^6 cells was proved preferable, with transfection efficiency being 53.3% and cell viability being 69.7%. In the preliminary experiment the siRNA against PRL decreased the mRNA of PRL by 34.0%. Conclusion It is possible to inhibit hormone hypersecretion by RNA interference, that may eventually enable therapeutic siRNA drugs developed.

China is extremely poor in mineral resources of Platinum Group Metals (PGMs), productive output of PGMs from mineral resource is 2.5 tons per year. At the same time, China is the biggest PGMs consumption country in the world, the mineral resource of PGMs is critical shortage, it shows the importance of recycling the secondary resource of PGMs. Sino-Platinum Metals Resource (Yimen) Co., Ltd. is the leader in recycling of PGMs from secondary resource, and has made outstanding contributions to China PGMs secondary resources recycling. This article elucidates the current situation of secondary resources recovery and development of metallurgical technology for PGMs.

Important immunohistochemical markers for identifying sweat glands

he major adnexal structures of the skin, the sweat glands, regulate the body temperature. There are twotypes of sweat glands in the human body, the eccrine type, which produces the NaCl-rich solution in response to heat, and the apocrine type, which releases a lipid-rich secretion. Human sweat glands have many immunohistochemical markers which have close relationship with sweat-related diseases. Recently, although numerous immunohistochemical markers appeared in sweat glands, these biomarkers scattered in the oceans of literature without a systemic review for the relationship between the immunohistochemical marker and their functions in the counterparts of the sweat glands. It is important for identifying sweat gland-derived neoplasms that originate from or differentiate into the ducts or secretory elements of the sweat glands by immunohistochemical markers. Therefore, it is necessary to make a review of the immunohistochemical markers systematically. Actually, we try to classify the immunohistochemical markers of the sweat glands into four groups according to their protein characters and the functions associated with their presumed physiological counterparts of the sweat apparatus.

Salvage liver transplantation in the treatment of hepatocellular carcinoma:A Meta-analysis

AIM:To evaluate survival and recurrence after salvage liver transplantation(SLT) for the treatment of hepatocellular carcinoma(HCC) compared with primary liver transplantation(PLT) using a ***:Literature on SLT versus PLT for the treatment of HCC published between 1966 and July 2.11 was retrieved.A meta-analysis was conducted to estimate pooled survival and disease-free rates.A fixed or random-effect model was established to collect the ***:The differences in overall survival and disease-free survival rates at 1-year,3-year and 5-year survival rates were not statistically significant between SLT group and PLT group(P > 0.05).After stratifying the various studies by donor source and Milan criteria,we found that:(1) Living donor liver transplantation recipients had significantly higher 1-year survival rate,lower 3-year and 5-year survival rates compared with deceased-donor liver transplantation(DDLT) *** in DDLT recipients they had better 1-year and 5-year disease-free survival rate in SLT group;and(2. No difference was seen in 1-year,3-year and 5-year survival rates between two groups who beyond Milan criteria at the time of liver ***:SLT can be effectively performed for patients with recurrence or deterioration of liver function after hepatectomy for *** does not increase the perioperative mortality and has a similar long-term survival rates compared to PLT.

Accuracy evaluation of a new three-dimensional reproduction method of edentulous dental casts, and wax occlusion rims with jaw relation

Development of Quantitative Real-time Polymerase Chain Reaction for the Detection of Vibrio vulnificus Based on Hemolysin (vvhA) Coding System

Objective To establish a TaqMan real-time fluorescent quantitative PCR to detect Vibrio vulnificus based on the hemolysin gene （vvhA） coding cytolysin. Methods Primers and probes in the conserved region of the vvhA gene sequence were designed for the TaqMan real-time PCR to detect 100 bp amplicon from V. vulnificus DNA. Recombinant plasmid pMD19-vvhA100 was constructed and used as a positive control during the detection. Minimal amplification cycles （Ct value） and fluorescence intensity enhancement （ARn value） were used as observing indexes to optimize the reaction conditions of TaqMan real-time PCR. The TaqMan assay for the detection of Vbirio vulnificus was evaluated in pure culture, mice tissue which artificially contaminated Vibrio vulnificus and clinical samples. Results The established TaqMan real-time PCR showed positive results only for Vibrio vulnificus DNA and pMD19-vvhA100. The standard curve was plotted and the minimum level of the vvhA target from the recombinant plasmid DNA was 103 copies with a Ct value of 37.94±0.19, as the equivalent of 0.01 ng purified genomic DNA of Vibrio vulnificus. The results detected by TaqMan PCR were positive for the 16 clinical samples and all the specimens of peripheral blood and subcutaneous tissue of mice which were infected with Vibrio vulnificus. Conclusion TaqMan real-time PCR is a rapid, effective, and quantitative tool to detect Vibro vulnificus, and can be used in clinical laboratory diagnosis of septicemia and wound infection caused by Vibrio vulnificus.

Mechanics Principle and Engineering Application of Split Layer and Bed Separation of Mining Overburden

To control land surface subsidence caused the underground mineral exploitation and the catastrophic phenomena such as serious damage of buildings, waterbodies, cultivated lands, railways, bridges caused by land subsidence, bed separation grouting technology of overburden is put forward. To provide theoretical support for the technology, the characteristics and the mechanics mechanism of mining overburden from layer-split to formation of bed separation are studied. On the basis of elastic sheet board theory, calculation formula of rock sheet deflection is presented, and the mechanics criteria of the separation formation and the calculation formula of bed separation volume are set up. Finally, the applications and technics of bed separation grout technology of mining overburden to control land subsidence in china are introduced.

过江盾构隧道选线过程中因避让不良地质段,或地形上满足两端进洞及展线要求而常产生局部弯曲,如苏州市海太过江盾构隧道。而隧道曲率对其动力响应影响规律在现有研究中鲜有提及,为探究在动力条件下不同曲率的盾构隧道纵向方向管环的轴力、张开量等变化规律,运用纵向反应位移法思想,基于长大隧道非一致激励动力分析方法,建立多质点-梁-弹簧模型,开展多工况地震动输入下的不同曲率盾构隧道纵向动力响应分析。结果表明:数值模型与计算方法能够得到合理的隧道结构动力响应;盾构隧道动力响应随隧道曲率增加而增加;不同曲率盾构隧道在进入土层交界区段其轴力和张开量出现显著的差异。该研究结果可为盾构隧道选线论证及动力响应分析提供更为可靠完善的计算模型与设计依据。

Local excision carcinoma in early stage

AIM: To assess the validity of local excision for the early stage low rectal cancer as an effective treatment alternative to radical ***: A retrospective medical chart review was done in 47 patients with early stage low rectal carcinoma who underwent local excision from November 1980 through November 1999 at Cancer Hospital of Chinese Academy of Medical Sciences (CAMS). The patients were treated by either transanal (40 cases), trans-sacral (5 cases), or trans-vaginal (2.cases) excision of tumors and no death was related to surgery. Sixteen patients received postoperative ***: T1 and T2.lesion was found in 36 (76.6 %) and 11 patients (2..4 %) respectively. The overall local tumor recurrence rate was 14.9 % (7/47), with an average recurrence time of 2. months. Among these 7 recurrent patients, there were 4 T1 and 3 T2.lesions. Microscopically,the surgical incisal margin was negative in 45 (95.7 %) and positive in 2.patients (4.3 %); Both of the later had developed local recurrence. The overall 5-year survival rate was 91.7 %,in which there were 94.4 % for T1 and 83.3 % for T2.tumors.T stage, intravessel tumor thrombosis, lymphocytic infiltration and histological grade were not found to be significant by related to the local recurrence and survival (P＞0.05).CONCLUSION: Local tumor excision was a safe procedure for the treatment of early stage low rectal carcinoma with minimal morbidity and mortality, which might serves as one of the primary surgical treatment methods for the disease of this kind.
