YAP(yes-associated protein),a key transcriptional cofactor that is negatively regulated by Hippo pathway,is critical for the development and size control of multiple organs such as liver.However,its role in the brain remains largely unclear.Here we provide evidence for YAP regulation of mouse neocortical astrocytic differentiation and proliferation.YAP was undetectable in neurons,but selectively expressed in neural stem cells(NSCs) and astrocytes.YAP in NSCs was required for neocortical astrocytic differentiation,with little role in selfrenewal or neural differentiation.However,YAP in astrocytes was necessary for astrocytic proliferation.YAP knock-out mice,Yap

To study investigated the frequency and the relationship between behavioral and psychological symptoms of dementia(BPSD) in Alzheimer’s disease patients.The cross-sectional study was derived from the Beijing Dementia Cooperative Study.The diagnosis of dementia was made according to the DSM-IV criteria,and a total of 127 elderly people with mild or moderate dementia assessed with the Clinical Dementia Rating scale was involved.The BPSD was assessed with the Neuropsychiatric Inventory(NPI).The principal component analysis with varimax rotation Was used for factor analysis.Overal1,48.8%of the subjects reported one or more neuropsychiatric symptoms.cluster analysis showed four behavioral subsyndromes,mood/impulsivity(36.2%),psychosis(28.3%),psychosomatic disorder(26.8%)and abnormal behavior(15.0%).The results suggest that there are some behavioral sub-syndromes in dementia,which are more common than any single behavioral disturbance;There may be a common cause or risk factor for the BPSD in same sub-syndrome.

Objective:To explore the association of TNF-α-1031 T/C polymorphism and age of onset and cognitive functions.Methods:We recruit486 inpatients with schizophrenia and 310 age-,sex-and education matched healthy controls.Cognitive functions were measured by RBANS and symptoms were assessed by PANSS.The TNF-α-1031 T/C polymorphisms were measured by PCR-RFLP.Results:There were no significant differences in allelic frequency and genotype frequency of TNF-α-1031 T/C polymorphism between patients and normal controls,but a significant association with age of onset(F=7.321,P=0.007) and significant associations with immediate memory,visuospatial,total score of RBANS(F=9.140,P=0.003;F=6.742,P=0.010;F=8.331,P=0.004).The age of onset was related to attention,immediate memory,delayed memory and total score of RBANS(r=0.179,P=0.002;r=0.102,P=0.041;r=0.154,P=0.008;r=0.134,P=0.021).It is significant for the mediation analysis for the relationship across TNF-α-1031 T/C polymorphism,age of onset and cognitive functions.Discussion:Our results suggested that TNF-α-1031 T/C plays a role in cognitive impairments,age of onset and their association in chronic patients with schizophrenia.

目的:探讨事件相关脑电位(ERP)P300、P50在颅脑外伤伤残评估中的应用。方法:选取60例颅脑外伤后智能损伤患者作为病例组,同时选取60例健康的受试者作为正常对照组,应用脑电生理仪记录并比较两组的P300的差异。结果:患者组P300较对照组潜伏期显著延长(P<0.01)、波幅降低(P<0.01);患者组S2-P50波幅明显高于对照组(P<0.05),S2-P50波幅/S1-P50波幅的比值明显大于对照组(P<0.05)。结论:ERP有助于为颅脑外伤后认知障碍严重程度的司法鉴定提供客观的脑电生理参考指标。

目的:探讨儿童期创伤对健康成人执行功能的影响及其潜在的脑功能机制。方法:选取24名伴儿童期创伤的健康成年人及24名年龄性别严格匹配的不伴儿童期创伤的健康成年人为研究对象。采用威斯康星卡片分类测试、彩色连线测验以及STROOP色词测试测查执行功能。运用比率低频振幅(fALFF)方法对两组受试者静息态功能核磁共振成像(rs-fMRI)数据进行对比分析。以呈现异常fALFF值的脑区作为种子点进行功能连接分析。将大脑fALFF值及脑区间功能连接强度与儿童期创伤严重程度和执行功能进行相关分析。

Bipolar disorder has been proved to be a multigenic disease,and genetic factors play an important role in the pathogenesis of this disease.If there is a correlation between the risk genes and the age of onset of bipolar disorder,we can predict the onset age of bipolar disorder individuals by testing genes,so as to improve the early diagnosis and early intervention.To test this hypothesis,we search out 26 SNP sites which have been demonstrated to be associated with bipolar disorder.Then we test these SNPs’ genotypes of 224 bipolar disorder patients,and analyzed the correlation between the risk genotypes and the age of onset.The results suggest that there is no significant correlation between these SNP sites and the age at onset in the patients with bipolar disorder(P > 0.05).In summary,the study confirmed that bipolar disorder is a multigenic disease,and the single risk gene has no effect on the age of first onset.

a-Synucleinopathies,a spectrum of progressive neurodegenerative disorders,most notably Parkinson’s disease(PD),are characterized by abnormal accumulation of misfolded α-synuclein(aS)-positive cytoplasmic inclusion,as well as neurodegeneration and cognitive impairments,but the molecular mechanisms underlying ccS aggregation and propagation are largely unknown.Using a mouse model of α-synucleinopathy,we recently showed that aberrant adenosine Areceptor(AR) signaling contributes to neurodegeneration and cognitive impairments.Here,we explored the effect of chronic caffeine(a non-selective antagonist of AR) treatment on the aS transmission mouse model of a-synucleinopathy by injection of preformed A53 T aS fibrils into prefrontal cortex region.We demonstrated that phamacologically blocking ARs signaling by chronic caffeine treatment blunted a cascade of pathological events leading to synucleinopathy,including pSer129 α-Syn-rich and p62-positive aggregates,apoptotic neuronal cell death,microglia and astroglia reactivation,NF-kB activation,pronounced neuroinflammation and working memory deficits.Combining our previous AR gene knockout findings,the present studies suggest that caffeine may exerts cognitive improvement and neuroprotective effects against aS pathology by modulating aS aggregation via targeting ARs.Thus,our data provides a neurobiological basis for epidemiological investigation into the possible relationship between caffeine consumption and development of α-Synucleinopathies in humans.

Background:Methamphetamine(METH) has been suggested to produce neuroinflammation.NALPl inflammatory bodies participate in the inflammatory reaction process via a series of products and regulatory pathways.Lipoxin produces both anti-inflammatory and proinflammatory effect.The current study is aimed to investigate effect of NALPl inflammasome and Lipoxins in methamphetamine-induced on neuroinflammation in rats and the regulatory role of the lipoxin to NALPl inflammasome.Methods:METH self-administration model was firstly established.Subsequently,the animals were treated with lipoxins(i.p injections,10 mg/ml) for consecutive 5、 10 and 15 days.The aggregation and expression of NALPl were measured in different brain regions.The expression of caspase-1 and IL-1β were also detected.Results:The animals were treated with methamphetamine and normal saline:the NALPl inflammatory bodies 、 the content of caspase-1 and IL-1β were significantly elevated in different brain regions.The animals were treated with methamphetamine and lipoxin:the NALPl inflammatory bodies 、the content of caspase-1 and IL-1β were decreased significantly in different brain regions,the protein expressed different in different brain regions.Conclusions:The current studies demonstrate that NALPl inflammasome regulates IL-1βthrough its hydrolysis product,caspase-1.Furthermore,this process is involved in METHinduced neuroinflammation.Lipoxin decreased the expression of proinflammatory cytokines and,subsequently,inhibited METH-induced neuroinflammation induced.
