Deciphering the coordinated regulation of gene silencing and pathogenicity by histone methylations in a plant fungal pathogen ***

摘      要:In eukaryotes,previous studies often considered the control of histone H3K9me3 and H3K27me3,two major methylation markers critical for heterochromatin structures and epigenetic regulation of genetic information,as independent pathways,recruited and catalyzed by different complexes with site ***,recent findings have demonstrated that these modifications can co-occur at the same gene locus,suggesting that despite their functional differentiation,H3K9me3 and H3K27me3 exhibit complementarity and synergistic *** the study of the coordinated regulation mechanism of H3K9me3 and H3K27me3 is still in the early stages,and many studies have implicated histone methylation in fungal pathogenesis,we utilized Verticillium dahliae,a fungus possessing both modifications and capable of causing severe plant vascular disease,as a model organism to investigate the coordinated regulation of gene silencing and pathogenicity by histone H3K9 and H3K27 *** mutated the histone H3K9me3 and H3K27me3 methyltransferases Vd KMT1 and Vd EZH2in *** and observed significant upregulation of numerous genes,indicating the crucial roles of both histone H3K9me3 and H3K27me3 in gene *** in situ labeling of Vd KMT1and Vd EZH2,combined with native complex extraction and mass spectrometry analysis,we identified the essential components of the complexes and verified their *** further explore the coordinated regulation mechanism of H3K9me3 and H3K27me3,we employed CUT&Tag-seq to detect the H3K9me3 and H3K27me3 levels in the methyltransferase mutants VdΔKMT1 and VdΔEZH2,respectively,and conducted joint analysis with transcriptome *** results revealed that the regulation of gene expression by H3K9me3 levels is achieved through indirect regulation of H3K27me3 ***,in the absence of Vd EZH2,the composition of a histone chaperone RbAp48 might increase in the Vd KMT1 complex to recruit Vd KMT1 to exert the functions of both H3
