摘      要:Sample preparation workflows for metabolomics studies of various sample types are often done on the basis of liquid-liquid extraction. Most commonly Bligh and Dyer extraction or one of its variants. This step is generally performed to separate polar endogenous metabolites from non-polar compounds such as lipids. We propose an integrated workflow using a UHPLC-SWATH/MS platform for the metabolomics analysis of plasma samples combined with an automated Bligh and Dyer extraction on a robotic sample preparation system. Lipids are separated into three fractions using a mixed mode μSPE. The resulting three fractions are analyzed on a hyphenated HILICSWATH/MS system. The solid phase extraction offers an attractive alternative to equalize the abundance of the different lipid classes in a sample. As a consequence injection volumes and therefore dynamic ranges can be optimized without saturating the system with higher abundant lipid classes or species.
