Identification and molecular characterization of exosome-like vesicles derived from Taenia asiatica adult worms

摘 要:Taenia asiatica is one of the most important food-borne parasites and posts a great threat to human *** far little is known about a specific infection and immune escape mechanism of ***,exosome-like vesicles have been emerging as a regulator in the interactions between parasites and hosts,which provide a new sight for the research on the infection of ***,exosome-like vesicles were collected from the excretory/secretory products of cultured *** and purified by differential *** purified vesicles,ranging from 50 to 150 nm in size,were identified as exosomes by transmission electron microscope and nanoparticle tracking *** analysis showed that a total of 843 proteins were identified in the exosome-like vesicles(737 from *** and 106 from the host) and enzymes involved in metabolic processes,such as glyceraldehyde 3 phosphate dehydrogenase,fructose 1,6 bisphosphate aldolase,cytosolic malate dehydrogenase,enolase and so on,were enriched in *** abundant proteins from proteomic analysis 14-3-3 and enolase were present in the exosome-like vesicles by Immunogold labeling ***,we validated the expressions of *** mi RNAs,including 6 known mi RNAs(tas-mi R-71,tas-mi R-1,tas-mi R-7,tas-mi R-9,tas-mi R-10,tas-let-7) and 3 novel mi RNAs(tas-m0022-3 p,tas-m0816-3 p,tas-m0082-5 p).The eight mi RNAs expression levels were mainly consistent with those obtained by high-throughput ***,we also demonstrated that these exosomes were internalized by Lo Vo cells in vitro through uptake experiment of exosome-like vesicles labeled with the green fluorescent dye *** study firstly isolated and identified exosome-like vesicles derived from *** adult worms,and described molecular characteristics of some cargoes loaded on these ***,exosome-like vesicles were internalized by Lo Vo *** findings provide new insights into the interaction between
