Long noncoding RNA H19 inhibits the growth and invasion of trophoblasts by inactivating Wnt/β-catenin signaling via downregulation of DDX3X

摘 要:In our study, we highlights the importance of lnc RNA-H19 in the restriction of early placenta overgrowth and the transfer of genetic and epigenetic information from the entire genome. We examined the effects of H19 on the proliferation and invasion of human trophoblasts. To further explore the mechanism, RNA pulldown and mass spectrometry were used to identify lnc RNA-H19-binding proteins in human trophoblasts. In addition, changes in methylation of the H19 promoter region were determined using pyrophosphate sequencing *** results indicate that lnc RNA-H19 was upregulated in the spontaneous abortion samples, consistent with the hypomethylation of the differential methylation region(DMR) of lnc RNA-H19. Overexpression of lnc RNA-H19 inhibited trophoblast growth and invasion. Furthermore, our studies demonstrated that H19 inhibited Wnt-β-catenin pathway activation in trophoblasts by affecting DDX3 X function, thus clarifying the mechanism of action of H19.
