Ketamine modulates dopamine release in mice striatum in vivo

摘 要:Ketamine is known as a general anesthetic that administration of subanesthetic doses(R, S)-ketamine(2, 10, 50 mg/kg, i.p.) produces rapid onset antidepressant effects in humans with profound analgesic and hypnotic properties. It is also a popular addictive drug that targeting dopaminergic(DA) system. However, whether and how ketamine modulates DA functions is elusive. Here we determined the effect of ketamine on DA release using electrochemical carbon fiber electrode(CFE) in mouse striatum in vivo. DA release was evoked by electrical stimulation at MFB. Ketamine decreased the DA overflow, while the DA uptake and vesicle pool recycling were not changed. Ketamine effect on DA release was similar in anesthetic and awaking status. These results suggest that ketamine may exert its addictive role is probably independent on MFB-induced DA release in vivo.
