Sequence variability in two mitochondrial DNA regions and internal transcribed spacer among Coenurus infecting animals and humans from different regions of China

摘 要:Objectives Sequence variability in two mitochondrial DNA(mtDNA) regions, namely cox1 and 12 S rRNA, and ITS of rDNA in Coenurus from different geogr-aphical regions in China was examined to analyze the variation of three sequences. Methods A portion of the cox1(pcox1), ITS genes and the 12 S rRNA(p12S) were amplified respectively from individual sample by PCR. Representative amplicons were subjected to sequencing for estimating sequence variability. Results The sequences of pcox1, 12 S rRNA and ITS were 421, 617 bp and 1320 in length, severally. the level of pairwise sequence variation between individual haplotypes were concluded to be 0—1.6% for the pcox1, 0—0.9% for the 12 S rRNA, 0.3%—4.2% for ITS. while, the inter-specific sequence variation were dramatically higher,being 9.9—28.7%, 8.3—26.4%,36—72.34% for pcox1,12 S rRNA and ITS, respectively. Phylogenetic analyses based on the 12 S rRNA sequence data revealed that T. multiceps and T. hydatigena were more closely related to the other members of the Taenia genus, and parasites of genus Echinoco- ccus were sister to the genus Taenia. Conclusion we could make a decision that Coenurus cerebralis and Coenurus skrjabini were belonged to the larval development of Taenia *** findings demonstrated clearly the usefulness of mtDNA and rDNA sequences for population genetic studies of these cestodes of socio-economic importance.
