摘 要:lntracellular membrane trafficking is essential for eukaryotic cell ***,we show that RAB37,a small GTPase involved in vesicle transport,is a target of ATG5,a key protein required for ***37activation through GTP binding recruits ATG5-12 to isolation membrane and promotes autophagosome *** analysis reveals that GTP-bound RAB37 exhibits enhanced interactions with ATG5-12 and GDP-stabilized mutation impairs the ***37 recruits ATG5-12 to LC3-Ⅱpuncta in a GTP-dependent manner to promote ***,RAB37 vesicles participate in formation of autophagosomes and subsequent autolysosomesupon starvation *** ablation of RAB37 expression decreases autophagy and increases tumour metastasis in vivo,whereas forced RAB37 expression suppresses cell proliferation and tumour *** results identify RAB37 as a master organiser of vesicle transport and autophagosome formation through a molecular link of RAB37,ATG5-12 up to *** findings have broad implications for understanding me role of RAB37 vesicle transport in autophagy and cancer.
