DEPOLARIZATION-INDUCED RECOVERY OF GABAA RECEPTORS FROM DESENSITIZATION IN α1β2γ2 ANDα1β2HEK293 CELL

摘 要:正 Depolarization mediated reactivation of GABAA receptors from desensitization were investigated using recombinant GABAA receptors stable expressed in rat α1β2γ2 and rat α1β2 with whole cell patch clamp technique. The holding potential was clamped at -60mV and GABA applied at a saturating concentration for 1 min. After a 30 s interval, applied GABA for 1 min a second time. The initial GABA application at -60 mV produced a large GABA response with fast desensitization. A subsequent GABA (1mM).application at -60 mV to the same cell resulted in reduced peak to average 60.9% of initial GABA (n=20). Depolarization to +20 mV, 0 mV, -20 mV for 30sec produced recovery in amplitude of second GABA application,. The peak amplitude was 106.1%,85.26%, 78.8% of initial GABA application (n=14). The effect of depolarization on recovery of desensitization was studied in α1β2 too. Applying GABA for 1min after clamping at +20 mV, 0 mV,-20 mV for 30 s give an average peak amplitude of 83.2%, 82.5%, 55.2% (n=14). Howeve
