Effects of ischemia on degradation of amyloid beta

摘      要：正Objective To investigate the changes of amyloid precursor protein（APP） processing under ischemic condition and the effects of ischemia on the expression of beta amyloid degradation enzyme neprilysin.Furthermore,to explain the new mechanisms why ischemia exacerbate the pathogenesis of Alzheimer’s disease.Methods Cells were treated with ischemia through transferring them to a ischemic humidity chamber（1%O2）.Intracellular APP metabolites were analyzed by immunoblotting.And the mRNA and protein levels of neprilysin were detected via RT-PCR and immunoblotting respectively.Results（1） Intracellular APP metabolites were increased under ischemic condition.（2） Ischemia decreased the mRNA and protein levels of neprilysin.Conclusion The increase of APP metabolites under ischemic condition is due to the reduction of neprilysin expression.
