Degradation of Hyaluronan by Hydrogen Peroxide or by Hyaluronidase

摘      要:Compared to hyaluonan(HA)with high molecular weight,HA with low molecular weight(LMWHA)has different or diverse biological activities.The biological activities of HA are closely related to its molecular weight.Up to now,the molecular weight range of LMWHA has not been defined,but generally HA with average molecular weight of 1×10 to 50×10 is considered as LMWHA.In this paper,the preparation of LMWHA by degrading HA with hydrogen peroxide or hyaluronidase was studied,and the effects of degradation conditions on molecular weight also were investigated,including the concentrations of hydrogen peroxide or hyaluronidase,pH,reaction temperature and time. The results showed that the degradation of HA with hydrogen peroxide was influenced by the concentration of hydrogen peroxide,reaction temperature and pH.The selected degradation condition was as follows:the concentration of hydrogen peroxide was 3.0%,reacted at 50℃with pH 7.0.The LMWHA obtained by hydrogen peroxide oxidation for 7,9,11,13 and 15 h had weight average molecular weight(M)of 21.1×10,7.5×10,5.7×10,5.2×10 and 4.8×10, respectively,as analyzed by high performance gel permeation chromatography(HPGPC),and their polydispersion index(PDI)was about 2.5.While LMWHA hydrolyzed by 30 U/ml hyaluronidase at 50℃with pH 7.0 for 2,3,4,5 and 6 h had M of 9.6×10,8.3×10,7.1×10, 6.3×10 and 5.7×10,respectively,and the PDI was about 2.0.Therefore,this two degradation methods of HA both are effective,and the degradation speed by hyaluronidase is more quick than by hydrogen peroxide.The uronie acid contents of LMWHA have no difference compared with undegraded HA.
