Presence of GABAergic neuron progenitors in the mouse neocortex and spinal cord

摘 要:正It has been suggested that the main source of GAB Aergic neurons in the rodent neocortex and spinal cord is in the subpallium and the ventricular zone of the spinal cord,*** study revealed a production of GABAergic neurons in the mouse neocortex and the spinal mantle *** immunohistochemistry,we detected that about 25%of the GFP-positive cells in the cortical subventricular zone(SVZ) of GAD67-GFP knock-in mouse at E18 were positive for Ki- 67 and some of them were undergoing mitosis or in the DNA synthesis *** cells obtained from the deeper half of the neocortex in GAD67-GFP knock-in mouse at E18 were further used for immunocytochemistry and single-cell RT-PCR. Immunostaining results showed that all GFP-positive cells were positive for *** were also positive for MAP2(99.1%) and Tuj1(95.2%).We then found some GFP-positive cells were characterized by cell cycle makers Ki-67(14. 4%),P-H3(4.7%) and BrdU(0.8%).Single-cell PCR showed that 18.5%of GFP-positive cells expressed Ki-67 and all GFP/Ki- 67 cells also expressed GAD67,MAP2 and *** also detected that about 1.6%,0.3%and 0.5%of GFP-positive cells in the spinal mantle layer at E17 were positive for Ki-67,P-H3 and BrdU,*** also revealed that some GFP-positive cells dissociated from the E17spinal mantle layer were positive for Ki-67(2.1%),P-H3(0.5%) and BrdU(0.8%).The present results indicate that neuronal and cell-cycle markers are colocalized in GABAergic-neuron progenitors that contribute to the production of neocortical and spinal GABAergic interneurons.
