摘      要:To investigate the role of ATP-sensitive potassium (KATP) channels on blood pressure variability (BPV) in sinoaortic-denervated (SAD) rats。SAD was performed on male Sprague-Dawley rats four weeks before the study。Using real-time quantitive polymerase chain reaction, we found that expression of SUR2 from SAD aorta was significantly increased, whereas Kit6.1 was decreased when compared to the sham-operated rats。For mesenteric artery both SUR2 and Kit6.1 were reduced markedly。No changes were observed about Kit6.2 between SAD and sham-operated rats for both arteries。Westem blotting and laser confocal immunofluorescence assays Of proteins level were in agreement with the result of mRNA。Concentration-response curves to either adenosine or pinacidil on isolated aorta and mesenteric artery between sham-operated and SAD rats were established。The curves of aorta obtained from the SAD shifted significantly to the left。While the curves of mesenteric artery Of the SAD shifted to the right。Different KATP channels openers and blockers on BPV were studied in conscious SAD rats。Not only pinacidil and diazoxide, but also glibenclamide significantly decreased BPV。Either HMR1098 or 5-HD or combined administration of them did not influence BPV。Pretreatment with the combination of HMR1098 and 5-HD did not modify the effects of the openers on BPV。We concluded that after SAD vascular KATP was disparate, this disparity led to different vasomotor activity to agents acting on KATP channels, which might induce greater fluctuation of vascular resistance and produce increased BPV。Relief of this fluctuation by opening or blocking KATP channels, BPV was reduced。
