Photosystem Ⅱ(PSⅡ)is a multi-subunit protein complex of the photosynthetic electron transport chain that is vital to photosynthesis.Although the structure,composition,and function of PSⅡ have been extensively studied,its biogenesis mechanism remains less understood.Thylakoid rhodanese-like(TROL)provides an anchor for leaf-type ferredoxin:NADP^(+)oxidoreductase.Here,we report the chacterizaton of a second type of TROL protein,TROL2,encoded by seed plant genomes whose function has not previously been reported.We show that TROL2 is a PSⅡ assembly cofactor with essential roles in the establishment of photoautotrophy.TROL2 contains a 45-amino-acid domain,termed the chlorotic lethal seedling(CLS)domain,that is both necessary and sufficient for TROL2 function in PSⅡ assembly and photoautotrophic growth.Phylogenetic analyses suggest that TROL2 may have arisen from ancestral TROL1 via gene duplication before the emergence of seed plants and acquired the CLS domain via evolution of the sequence encoding its N-terminal portion.We further reveal that TROL2(or CLS)forms an assembly cofactor complex with the intrinsic thylakoid membrane protein LOW PSⅡ ACCUMULATION2 and interacts with small PSⅡ subunits to facilitate PSⅡ complex assembly.Collectively,our study not only shows that TROL2(CLS)is essential for photoautotrophy in angiosperms but also reveals its mechanistic role in PSⅡ complex assembly,shedding light on the molecular and evolutionary mechanisms of photosynthetic complex assemblyin angiosperms.

Nucleic acid vaccine or DNA vaccine is a hopeful vac- cine to prevent and treat viral hepatitis. Problems exist in different DNA vaccines for HBV or HCV. Optimal animal model should be established study vaccine against hepatitis. Apart from the strategy to enhance the efficiency of DNA vaccine, combined use of cytokines or chemokines, different routes of inocu- lation, design of optimal vector, ISS insertion in the plasmid vectors, etc to enhance the efficiency of DNA vaccine are reviewed.

Patients with chronic obstructive pulmonary disease(COPD)who exhibit elevated blood eosinophil levels often experience worsened lung function and more severe emphysema.This implies the potential involvement of eosinophils in the development of emphysema.However,the precise mechanisms underlying the development of eosinophil-mediated emphysema remain unclear.In this study,we employed single-cell RNA sequencing to identify eosinophil subgroups in mouse models of asthma and emphysema,followed by functional analyses of these subgroups.Assessment of accumulated eosinophils unveiled distinct transcriptomes in the lungs of mice with elastase-induced emphysema and ovalbumin-induced asthma.Depletion of eosinophils through the use of anti-interleukin-5 antibodies ameliorated elastase-induced emphysema.A particularly noteworthy discovery is that eosinophil-derived cathepsin L contributed to the degradation of the extracellular matrix,thereby leading to emphysema in pulmonary tissue Inhibition of cathepsin L resulted in a reduction of elastase-induced emphysema in a mouse model.Importantly,eosinophil levels correlated positively with serum cathepsin L levels,which were higher in emphysema patients than those without emphysema.Expression of cathepsin L in eosinophils demonstrated a direct association with lung emphysema in COPD patients.Collectively,these findings underscore the significant role of eosinophil-derived cathepsin L in extracellular matrix degradation and remodeling,and its relevance to emphysema in coPD patients.Consequently,targeting eosinophil-derived cathepsin L could potentially offer a therapeutic avenue for emphysema patients.Further investigations are warranted to explore therapeutic strategies targeting cathepsin L in emphysema patients.

Effects of flow velocity on water quality and ammonia excretion in recirculating aquaculture system culturing juvenile largemouth bass(Micropterus salmoides)

Flow velocity plays an important role in recirculating aquaculture systems(RAS)and the growing practice of culturing juvenile largemouth bass(Micropterus salmoides).In this study,the effects of flow velocity on the water quality as well as the ammonia excretion were discussed from the perspective of actual production,and a polynomial model of ammonia nitrogen excretion was established,using the juvenile largemouth bass.Results showed that the range of ammonia nitrogen and nitrite nitrogen decreased with flow velocity increasing,while the number and volume share of large particles increased.According to the polynomial model,compared with the medium flow velocity(11 cm/s,2.45 body length(bl)/s),the ammonia excretion of juvenile largemouth bass at high(18 cm/s,4.00 bl/s),and low(4 cm/s,0.90 bl/s)flow velocity changed faster with time,and the excretion rate peaked at the 6th hour after feeding,earlier than that under medium flow velocity.Therefore,it is suggested to increase the flow velocity at the 5th hour after feeding and then decreased it at the 10th hour,to ensure better water quality in RAS culturing juvenile largemouth bass.

Despite recent progress in crop genomics studies,the genomic changes brought about by modern breeding selection are still poorly understood,thus hampering genomics-assisted breeding,especially in polyploid crops with compound genomes such as common wheat(Triticum aestivum).In this work,we constructed genome resources for the modern elite common wheat variety Aikang 58(AK58).Comparative genomics between AK58 and the landrace cultivar Chinese Spring(CS)shed light on genomic changes that occurred through recent varietal improvement.We also explored subgenome diploidization and divergence in common wheat and developed a homoeologous locus-based genome-wide association study(HGWAS)approach,which was more effective than single homoeolog-based GWAS in unraveling agronomic trait-associated loci.A total of 123 major HGWAs loci were detected using a genetic population derived from AK58 and cs.Elite homoeologous haplotypes(HHs),formed by combinations of subgenomic homoeologs of the associated loci,were found in both parents and progeny,and many could substantially improve wheat yield and related traits.We built a website where users can download genome assembly sequence and annotation data for AK58,perform blast analysis,and run JBrowse.Our work enriches genome resources for wheat,provides new insights into genomic changes during modern wheat improve-.ment,and suggests that efficientmining of elite HHs can make a substantial contribuutionto genomics-assisted breeding in common wheat and other polyploid crops.

Background:Chemotherapy is a common treatment for advanced hepatocellular carcinoma,but the effect is not satisfactory.The study aimed to retrospectively evaluate the effects of adding all-trans-retinoic acid(ATRA)to infusional fluorouracil,leucovorin,and oxaliplatin(FOLFOX4)for advanced hepatocellular carcinoma(HCC).Methods:We extracted the data of patients with advanced HCC who underwent systemic chemotherapy using FOLFOX4 or ATRA plus FOLFOX4 at the Eastern Hepatobiliary Surgery Hospital,First Hospital of Jilin university,and Zhejiang Sian International Hospital and retrospectively compared for overall survival.The Cox proportional hazards model was used to calculate the hazard ratios for overall survival and disease progression after controlling for age,sex,and disease stage.Results:From July 2013 to July 2018,111 patients with HCC were included in this study.The median survival duration was 14.8 months in the ATRA plus FOLFOX4 group and 8.2 months in the FOLFOX4 only group(P<0.001).The ATRA plus FOLFOX4 group had a significantly longer median time to progression compared with the FOLFOX4 group(3.6 monthsvs.1.8 months,P<0.001).Hazard ratios for overall survival and disease progression were 0.465(95%confidence interval:0.298–0.726;P=0.001)and 0.474(0.314–0.717;P<0.001)after adjusting for potential confounders,respectively.Conclusion:ATRA plus FOLFOX4 significantly improves the overall survival and time to disease progression in patients with advanced HCC.

目的观察揿针结合小儿推拿治疗儿童肝郁气滞型便秘的临床疗效。方法将60例肝郁气滞型便秘患儿随机分为联合组和推拿组,每组30例。推拿组采用小儿推拿治疗,联合组在推拿治疗基础上加用揿针治疗。观察两组临床疗效及治疗前后每周自主排便次数评分的变化。结果治疗后联合组治愈率、总有效率高于推拿组,差异有统计学意义(P<0.05)。与同组治疗前比较,两组治疗后每周自主排便次数均增加(P<0.05)。随着治疗疗程的延长,两组患者每周自主排便次数增加,且联合组次数多于推拿组,组间差异有统计学意义(P<0.01)。结论揿针结合小儿推拿治疗儿童肝郁气滞型便秘疗效优于单独小儿推拿治疗。

Natural locomotion such as walking,crawling,and swimming relies on spatially controlled deformation of soft tissues,which could allow efficient interaction with the external environment.As one of the ideal candidates for biomimetic materials,hydrogels can exhibit versatile bionic morphings.However,it remains an enormous challenge to transfer these insitu deformations to locomotion,particularly above complex terrains.Herein,inspired by the crawling mode of inchworms,an isotropic hydrogel with thermoresponsiveness could evolve to an anisotropic hydrogel actuator via interfacial diffusion polymerization,further evolving to multisection structure and exhibiting adaptive deformation with diverse degrees of freedom.Therefore,a dynamic mortise-and-tenon interlock could be generated through the interaction between the self-deformation of the hydrogel actuator and rough terrains,inducing continual multidimensional locomotion on various artificial rough substrates and natural sandy terrain.Interestingly,benefiting from the powerful mechanical energy transfer capability,the crawlable hydrogel actuators could also be utilized as hydrogel motors to activate static cargos to overstep complex terrains,which exhibit the potential application of a biomimetic mechanical discoloration device.Therefore,we believe that this design principle and control strategy may be of potential interest to the field of deformable materials,soft robots,and biomimetic devices.

Let a quadratic system of differential equations （E2） possess a focus of order 3 and a uniquesimple singular point at infinity. Then it possesses another focus with odd number of limit cyclesaround it. By suitable variations of the coefficients of the system,three limit cycles jump out ofthe focus of order 3 so that there exist at least four limit cycles for （E2）, and the global structureis completely determined.
