Development of NK cell-based cancer immunotherapies through receptor engineering

Natural killer(NK)cell-based immunotherapies are attracting increasing interest in the field of cancer *** clinical trials have shown promising outcomes,alongside satisfactory product efficacy and *** developments have greatly increased the therapeutic potential of NK cells by endowing them with enhanced recognition and cytotoxic *** review focuses on surface receptor engineering in NK cell therapy and discusses its impact,challenges,and future *** approaches are based on engineering with chimeric antigen receptors to allow NK cells to target specific tumor antigens independent of human leukocyte antigen *** approach has increased the precision and potency of NK-mediated recognition and elimination of cancer *** addition,engineering NK cells with T-cell receptors also mediates the recognition of intracellular epitopes,which broadens the range of target *** tumor peptide recognition by NK cells has also been improved by optimizing immunoglobulin constant fragment receptor expression and ***,engineered NK cells have an improved ability to recognize and destroy target cells coated with specific antibodies,thereby increasing their antibodydependent cellular *** ability of NK cell receptor engineering to promote the expansion,persistence,and infiltration of transferred cells in the tumor microenvironment has also been ***-based strategies for sustained NK cell functionality within the tumor environment have also been discussed,and these strategies providing perspectives to counteract tumor-induced ***,receptor engineering has led to significant advances in NK cell-based cancer *** technical challenges are addressed,these innovative treatments will likely reshape cancer immunotherapy.

Understanding the solid target dynamics resulting from the interaction with an ultrashort laser pulse is a challenging fundamental multi-physics problem involving atomic and solid-state physics,plasma physics,and laser *** of the initial interplay of the underlying processes is essential to many applications ranging from lowpower laser regimes like laser-induced ablation to high-power laser regimes like laser-driven ion *** the properties of the so-called pre-plasma formed as the laser pulse’s rising edge ionizes the target is complicated from the theoretical and experimental point of view,and many aspects of this laser-induced transition from solid to overdense plasma over picosecond timescales are still open *** the one hand,laser-driven ion acceleration requires precise control of the pre-plasma because the efficiency of the acceleration process crucially depends on the target properties at the arrival of the relativistic intensity peak of the *** the other hand,efficient laser ablation requires,for example,preventing the so-called“plasma shielding”.By capturing the dynamics of the initial stage of the interaction,we report on a detailed visualization of the pre-plasma formation and ***-thin diamond-like carbon foils are shown to transition from solid to plasma during the laser rising edge with intensities10^(16)W/cm^(2).Single-shot near-infrared probe transmission measurements evidence sub-picosecond dynamics of an expanding plasma with densities above 10^(23)cm^(−3)(about 100 times the critical plasma density).The complementarity of a solid-state interaction model and kinetic plasma description provides deep insight into the interplay of initial ionization,collisions,and expansion.

Dysregulation of G9a,a histone-lysine N-methyltransferase,has been observed in Alzheimer’s disease and has been correlated with increased levels of chronic inflammation and oxidative ***,microRNAs are involved in many biological processes and diseases playing a key role in pathogenesis,especially in multifactorial diseases such as Alzheimer’s ***,our aim has been to provide partial insights into the interconnection between G9a,microRNAs,oxidative stress,and *** better understand the biology of G9a,we compared the global microRNA expression between senescence-accelerated mouse-prone 8(SAMP8)control mice and SAMP8 treated with G9a inhibitor *** found a downregulation of miR-128 after a G9a inhibition treatment,which interestingly binds to the 3′untranslated region(3′-UTR)of peroxisome-proliferator activator receptor γ(PPARG)***,Pparg gene expression levels were higher in the SAMP8 group treated with G9a inhibitor than in the SAMP8 control *** also observed modulation of oxidative stress responses might be mainly driven Pparg after G9a *** confirm these antioxidant effects,we treated primary neuron cell cultures with hydrogen peroxide as an oxidative *** this setting,treatment with G9a inhibitor increases both cell survival and antioxidant ***,up-regulation of PPARγby G9a inhibitor could also increase the expression of genes involved in DNA damage responses and *** addition,we also described that the PPARγ/AMPK axis partially explains the regulation of autophagy markers ***,PPARγ/GADD45αpotentially contributes to enhancing synaptic plasticity and neurogenesis after G9a ***,we propose that pharmacological inhibition of G9a leads to a neuroprotective effect that could be due,at least in part,by the modulation of PPARγ-dependent pathways by miR-128.

An inverse method for characterization of dynamic response of 2D structures under stochastic conditions

The reliable estimation of the wavenumber space(k-space)of the plates remains a longterm concern for acoustic modeling and structural dynamic behavior *** current analyses of wavenumber identification methods are based on the deterministic *** this end,an inverse method is proposed for identifying wave propagation characteristics of twodimensional structures under stochastic conditions,such as wavenumber space,dispersion curves,and band *** proposed method is developed based on an algebraic identification scheme in the polar coordinate system framework,thus named Algebraic K-Space Identification(AKSI)***,a model order estimation strategy and a wavenumber filter are proposed to ensure that AKSI is successfully *** main benefit of AKSI is that it is a reliable and fast method under four stochastic conditions:(A)High level of signal noise;(B)Small perturbation caused by uncertainties in measurement points’coordinates;(C)Non-periodic sampling;(D)Unknown structural *** validate the proposed method,we numerically benchmark AKSI and three other inverse methods to extract dispersion curves on three plates under stochastic *** experiment is then performed on an isotropic steel *** investigations demonstrate that AKSI is a good in-situ k-space estimator under stochastic conditions.

A cheap method allowing fabrication of biocompatible,ultra-small(2-10 nm)and fluorescent(λ_(em)=425-500 nm)nanohybrids(NHs)from coffee wastes is *** gadolinium-doped nanohybrids(GDNHs)or gadolinium-free carbon dots(GFCDs)can be synthesized in a domestic microwave oven according to green synthesis *** sizes,chemical composition,impact on proton magnetic resonance relaxation time and optical properties of the GDNHs and GFCDs were studied in details and *** particular,doping of the NHs with Gd^(3+)ions,up to 1.87%w/w of gadolinium per particles’weight,will allow their application for magnetic resonance imaging(MRI).Furthermore,cell culture tests on human adenocarcinomic alveolar basal epithelial cells line(A549)have shown high biocompatibility of the GDNHs and in a wide concentration range 100-1000μg/ml.

Expanding through the Emerald Isle:exploration and spatial orientation of non-native bank voles in Ireland

Whether introduced into a completely novel habitat or slowly expanding their current range,the degree to which animals can effciently explore and navigate new environments can be key to survival,ultimately determining population establishment and colonization *** tested whether spatial orientation and exploratory behavior are associated with non-native spread in free-living bank voles(Myodes glareolus,N=43)from a population accidentally introduced to Ireland a century *** measured spatial orientation and navigation in a radial arm maze,and behaviors associated to exploratory tendencies and risk-taking in repeated open-feld tests,at the expansion edge and in the source *** voles at the expansion edge re-visited unrewarded arms of the maze more,waited longer before leaving it,took longer to start exploring both the radial arm maze and the open feld,and were more risk-averse compared to conspecifcs in the source *** together,results suggest that for this small mammal under heavy predation pressure,a careful and thorough exploration strategy might be favored when expanding into novel environments.
