Hexokinase 1 and 2 mediates glucose utilization to regulate the synthesis of kappa casein via ribosome protein subunit 6 kinase 1 in bovine mammary epithelial cells

Glucose plays a vital part in milk protein synthesis through the mTOR signaling pathway in bovine mammary epithelial cells(BMEC). The objectives of this study were to determine how glucose affects hexokinase(HK) activity in BMEC and investigate the regulatory effect of HK in kappa casein(CSN3)synthesis via the mechanistic target of rapamycin complex 1(mTORC1) signaling pathway in BMEC. For this, HK1 and HK2 were knocked out in BMEC using the CRISPR/Cas9 system. The gene and protein expression, glucose uptake, and cell proliferation were measured. We found that glucose uptake, cell proliferation, CSN3 gene expression levels, and expression of HK1 and HK2 increased with increasing glucose concentrations. Notably, glucose uptake was significantly reduced in HK2 knockout(HK2KO)BMEC treated with 17.5 mM glucose. Moreover, under the same glucose treatment conditions, the proliferative ability and abundance of CSN3 were significantly diminished in both HK1 knockout(HK1KO)and HK2KO BMEC compared with that in wild-type BEMC. We further observed that the phosphorylation levels of ribosome protein subunit 6 kinase 1(S6K1) were reduced in HK1KO and HK2KO BMEC following treatment with 17.5 mM glucose. As expected, the levels of glucose-6-phosphate and the m RNA expression levels of glycolysis-related genes were decreased in both HK1KO and HK2KO BMEC following glucose treatment. These results indicated that the knockout of HK1 and HK2 inhibited cell proliferation and CSN3 expression in BMEC under glucose treatment, which may be associated with the inactivation of the S6K1 and inhibition of glycolysis.

Various new conductive materials with exceptional properties are utilized for the preparation of electronic devices.Achieving ultra-high conductivity is crucial to attain excellent electrical performance.However,there is a lack of systematic research on the impact of conductor material thickness on device performance.Here,we investigate the effect of conductor thickness on power transmission and radiation in radio-frequency(RF)and microwave electronics based on MXene nanosheets material transmission lines and antennas.The MXene transmission line with thickness above the skin depth exhibits a good transmission coefficient of approximately-3 dB,and the realized gain of MXene antennas exceeds 2 dBi.Additionally,the signal transmission strength of MXene antenna with thickness above the skin depth is higher than 5-μm MXene antenna approximately 5.5 dB.Transmission lines and antennas made from MXene materials with thickness above the skin depth exhibit stable and reliable performance,which has significant implications for obtaining high-performance RF and microwave electronics based on new conductive materials.

Data race is one of the most important concurrent anomalies in multi-threaded programs.Emerging con-straint-based techniques are leveraged into race detection,which is able to find all the races that can be found by any oth-er sound race detector.However,this constraint-based approach has serious limitations on helping programmers analyze and understand data races.First,it may report a large number of false positives due to the unrecognized dataflow propa-gation of the program.Second,it recommends a wide range of thread context switches to schedule the reported race(in-cluding the false one)whenever this race is exposed during the constraint-solving process.This ad hoc recommendation imposes too many context switches,which complicates the data race analysis.To address these two limitations in the state-of-the-art constraint-based race detection,this paper proposes DFTracker,an improved constraint-based race detec-tor to recommend each data race with minimal thread context switches.Specifically,we reduce the false positives by ana-lyzing and tracking the dataflow in the program.By this means,DFTracker thus reduces the unnecessary analysis of false race schedules.We further propose a novel algorithm to recommend an effective race schedule with minimal thread con-text switches for each data race.Our experimental results on the real applications demonstrate that 1)without removing any true data race,DFTracker effectively prunes false positives by 68%in comparison with the state-of-the-art constraint-based race detector;2)DFTracker recommends as low as 2.6-8.3(4.7 on average)thread context switches per data race in the real world,which is 81.6%fewer context switches per data race than the state-of-the-art constraint based race detec-tor.Therefore,DFTracker can be used as an effective tool to understand the data race for programmers.

Identification and functional analysis of CG3526 in spermatogenesis of Drosophila melanogaster

Spermatogenesis is a critical part of reproduction in insects;however,its molecular mechanism is still largely unknown.In this study,we identified a testis-specific gene CG3526 in Drosophila melanogaster.Bioinformatics analysis showed that CG3526 contains a zinc binding domain and 2 C2H2 type zinc fingers,and it is clustered to the vertebrate really interesting new gene(RING)family E3 ubiquitin-protein ligases.When CG3526 was knocked down by RNA interference(RNAi),the testis became much smaller in size,and the apical tip exhibited a sharp and thin end instead of the blunt and round shape in the control testis.More importantly,compared to the control flies,only a few mature sperm were present in the seminal vesicle of C587-Gal4>CG3526 RNAi flies.Immunofluorescence staining of the testis from CG3526 RNAi flies showed that the homeostasis of testis stem cell niche was disrupted,cell distribution in the apical tip was scattered,and the process of spermatogenesis was not completed.Furthermore,we found that the phenotype of CG3526 RNAi flies’testis was similar to that of testis of Stat92E RNAi flies,the expression level of CG3526 was significantly downregulated in the Stat92EF06346 mutant flies,and the promoter activity of CG3526 was upregulated by STAT92E.Taken together,our results indicated that CG3526 is a downstream effector gene in the JAK-STAT signaling pathway that plays a key role in the spermatogenesis of Drosophila.

Text Difficulty Study:Do Machines Behave the Same as Humans Regarding Text Difficulty?

The objective of this study was to optimize the extraction process of total sterols from wild Pyracantha fortuneana leaves (WPFL), and explore their antioxidant and whitening activities. The yield of total sterols (WPFLTPS) was used as indicator, and the single factor experiment was used to investigate the effects of extraction solvent, solid-liquid ratio, ultrasonic temperature and ultrasonic time on the yield of WPFLTPS. Based on this, the orthogonal experimental design was used to optimize the extraction conditions. The DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] and nitrite methods were used to evaluate the antioxidant activity of WPFLTPS in vitro. The whitening activity was evaluated by the inhibitory effect on tyrosinase. The results showed that for optimal extraction process of WPFLTPS, the extraction solvent was absolute ethanol, ultrasonic temperature was 50 ℃, solid-liquid ratio was 1:16 g/mL, and ultrasonic time was 25 minutes. Under these conditions, the yield of WPFLTPS was (11.00± 0.03) mg/g. In the range of 0.5~5 mg/mL, WPFLTPS had strong scavenging effects on DPPH radicals, ABTS+ radicals and nitrite ions, as well as a strong inhibitory effect on tyrosinase, demonstrating a good dose-response relationship. This extraction process was stable and reliable for extracting WPFLTPS. The WPFLTPS can be used as a new, natural antioxidant and tyrosinase inhibitor in functional foods. © 2024 Editorial Department of Science and technology of Food Science. All rights reserved.

With the emergence of pre-trained models,current neural networks are able to give task performance that is comparable to humans.However,we know little about the fundamental working mechanism of pre-trained models in which we do not know how they approach such performance and how the task is solved by the model.For example,given a task,human learns from easy to hard,whereas the model learns randomly.Undeniably,difficulty-insensitive learning leads to great success in natural language processing(NLP),but little attention has been paid to the effect of text difficulty in NLP.We propose a human learning matching index(HLM Index)to investigate the effect of text difficulty.Experiment results show:1)LSTM gives more human-like learning behavior than BERT.Additionally,UID-SuperLinear gives the best evaluation of text difficulty among four text difficulty criteria.Among nine tasks,some tasks’performance is related to text difficulty,whereas others are not.2)Model trained on easy data performs best in both easy and medium test data,whereas trained on hard data only performs well on hard test data.3)Train the model from easy to hard,leading to quicker convergence.

Sb-based materials exhibit considerable potential for sodium-ion storage owing to their high theoretica capacities.However,the bulk properties of Sb-based materials always result in poor cycling and rate performances.To overcome these issues,pyridine-regulated Sb@InSbS3ultrafine nanoplates loaded on reduced graphene oxides （Sb@InSbS3@rGO） were designed and synthesized.During the synthesis process,pyridine was initially adopted to coordinate with In3+,and uniformly dispersed In2S3ultrafine nanoplates on reduced graphene oxide were generated after sulfidation.Next,partial In3+was exchanged with Sb3+,and Sb@InSbS3@rGO was obtained by using the subsequent annealing method.The unique structure of Sb@InSbS3@rGO effectively shortened the transfer path of sodium ions and electrons and provided a high pseudocapacitance.As the anode in sodium-ion batteries,the Sb@InSbS3@rGO electrode demonstrated a significantly higher reversible capacity better stability (445 m Ah·g-1at 0.1 A·g-1after 200 cycles and 212 mAh·g-1at 2 A·g-1after 1200 cycles),and superior rate (210 mAh·g-1at 6.4 A·g-1) than the electrode without pyridine (355 mAh·g-1at 0.1 A·g-1after 55 cycles and 109 mAh·g-1at 2 A·g-1after 770 cycles)Furthermore,full cells were assembled by using the Sb@InSbS3@rGO as anode and Na3V2（PO4）3as cathode which demonstrated good cycling and rate performances and exhibited promising application prospects.These results indicate that adjusting the microstructure of electrode materials through coordination balance is A·good strategy for obtaining high-capacity,high-rate,and longcycle sodium storage performances.

Graph processing has been widely used in many scenarios,from scientific computing to artificial intelligence.Graph processing exhibits irregular computational parallelism and random memory accesses,unlike traditional workloads.Therefore,running graph processing workloads on conventional architectures(e.g.,CPUs and GPUs)often shows a significantly low compute-memory ratio with few performance benefits,which can be,in many cases,even slower than a specialized single-thread graph algorithm.While domain-specific hardware designs are essential for graph processing,it is still challenging to transform the hardware capability to performance boost without coupled software codesigns.This article presents a graph processing ecosystem from hardware to software.We start by introducing a series of hardware accelerators as the foundation of this ecosystem.Subsequently,the codesigned parallel graph systems and their distributed techniques are presented to support graph applications.Finally,we introduce our efforts on novel graph applications and hardware architectures.Extensive results show that various graph applications can be efficiently accelerated in this graph processing ecosystem.

Augmented Reality is a technique that allows users to overlap digital information with their physical world.The Augmented Reality(AR)displays have an exceptional characteristic from the Human–Computer Interaction(HCI)perspective.Due to its increasing popularity and application in diverse domains,increasing user-friendliness and AR usage are critical.Context-aware is one approach since an AR application can adapt to the user,environment,needs and enhance ergonomic principles and functionality.This paper proposes the Intelligent Contextaware Augmented Reality Model(ICAARM)for Human–Computer Interaction systems.This study explores and reduces interaction uncertainty by semantically modeling user-specific interaction with context,allowing personalised interaction.Sensory information is captured from an AR device to understand user interactions and context.These depictions carry semantics to Augmented Reality applications about the user’s intention to interact with a specific device affordance.Thus,this study describes personalised gesture interaction in VR/AR applications for immersive/intelligent environments.
