Exploring Mechanisms of Panax notoginseng Saponins in Treating Coronary Heart Disease by Integrating Gene Interaction Network and Functional Enrichment Analysis

Objective： To investigate the mechanisms of Panax notoginseng saponins（PNS） in treating coronary heart disease（CHD） by integrating gene interaction network and functional enrichment analysis. Methods： Text mining was used to get CHD and PNS associated genes. Gene–gene interaction networks of CHD and PNS were built by the Gene MANIA Cytoscape plugin. Advanced Network Merge Cytoscape plugin was used to analyze the two networks. Their functions were analyzed by gene functional enrichment analysis via DAVID Bioinformatics. Joint subnetwork of CHD network and PNS network was identified by network analysis. Results： The 11 genes of the joint subnetwork were the direct targets of PNS in CHD network and enriched in cytokine-cytokine receptor interaction pathway. PNS could affect other 85 genes by the gene–gene interaction of joint subnetwork and these genes were enriched in other 7 pathways. The direct mechanisms of PNS in treating CHD by targeting cytokines to relieve the inflammation and the indirect mechanisms of PNS in treating CHD by affecting other 7 pathways through the interaction of joint subnetwork of PNS and CHD network. The genes in the 7 pathways could be potential targets for the immunologic adjuvant, anticoagulant, hypolipidemic, anti-platelet and anti-hypertrophic activities of PNS. Conclusion： The key mechanisms of PNS in treating CHD could be anticoagulant and hypolipidemic which are indicated by analyzing biological functions of hubs in the merged network.

An initial study on left ventricular diastolic function in patients with hypertrophy cardiomyopathy using single-beat, real-time, three-dimensional echocardiography

Objectives To assess the regional diastolic function in patients with hypertrophic cardiomyopathy （HCM） by using single-beat, real-time, three-dimensional echocardiography （RT-3DE）. Methods Sixty-five patients with HCM in sinus rhythm together with fifty age- and gender-matched normal controls were studied by two dimensional echocardiography （2DE） and RT-3DE. The parameters analyzed by RT-3DE included： left ventrieular （LV） volumes, left ventricular ejection fraction （LVEF）, end diastolic sphericity index （EDSI）, diastolic dyssynchrony index （DDI）, dispersion end diastole （DISPED）, and normalized 17 segmental volume-time curves. Results Evaluated by RT-3DE, LVEF was slightly lower compared with 2DE （63.2 ± 6.8% vs. 59.1 ± 6.4%, P 〈 0.0001）. Normal subjects had relatively uniform volumetric curves for all LV segments. In HCM patients, the segmental volumetric curves were dyssynchronous. Increased DDI and DISPED in end diastole were observed in patients with HCM （9.95 ± 3.75, 41.76 ± 17.19, P 〈 0.0001）, and not all abnormal volumetric segments occurred in the hypertrophic regions. Conclusions Patients with HCM have presented regional diastolic dyssynchrony in the diastole phase, and this preclinical lesion can be recognized by single-beat RT-3DE.

Purification and functional assessment of smooth muscle cells derived from mouse embryonic stem cells

Objective： To establish the diagnosis scale of blood stasis syndrome （BSS） and explore the idea and method of using scale to research the quantitative diagnosis of chinese medicine （CM） syndrome. Methods： Combining the modern epidemiology, consulting the access of quality of life scale, and colligating multi-angle methods to make the scale. Results： The scale had relatively good reliability and validity and could be used to estimate the degree of stasis and analyse the curative effect. Conclusions： It was a reference for CM syndrome diagnosis that combines screening methods of scale entry with quantitative diagnosis to establish the quantitative diagnosis scale.

Plasma homocysteine levels are independently associated with alterations of large artery stiffness in men but not in women

Objectives To investigate the associations of the plasma homocysteine levels with the alterations in arterial stiffness in a commu- nity-based cohort. The gender differences in these associations were examined. Methods We evaluated the relationship between plasma homocysteine levels to three measures of vascular ftmction [carotid-femoral pulse wave velocity （CF-PWV）, carotid-ankle PWV （CA-PWV） and heart rate corrected augmentation index （AI）] in 1680 participants （mean age： 61.5 years; 709 men, 971 women） from communities of Beijing, China. Results In univariate analysis, plasma homocysteine levels was positively related to the CF-PWV （r = 0.211, P 〈 0.0001） and CA-PWV （r = 0.148, P 〈 0.0001）, whereas inversely associated with AI （r = -0.052, P = 0.016）. In multiple linear regression models adjusting for covariants, plasma homocysteine remained positively related to the CF-PWV （standardized 13 = 0.065, P = 0.007） in total cases. When the groups of men and women were examined separately, plasma homocysteine remained positively associated with the CF-PWV （standardized β = 0.082, P = 0.023） in men, whereas the relations between homocysteine and any of the arterial stiffness indices were not further present in women. Conclusions In chinese population, plasma homocysteine levels are independently associated with alterations of large artery stiffness in men but not in women.

Rare occurrence of simultaneous coronary artery perforation and intracoronary thrombus formation following angioplasty

Both coronary artery perforation and intracoronary thrombus formation are life-threatening complications of percutaneous coronary interventions, which rarely occur simultaneously during *** herein report a case of stent-related, left circumflex artery perforation, and subsequently acute left main artery thrombosis after the leakage was embolized with 7 microcoils. Intracoronary thromboectomy and systemic anticoagulant therapy were carefully used with good results. This case also represents some of our uncertainties regarding the best management of the patient.

Biomedical Mechanisms of Blood Stasis Syndrome of Coronary Heart Disease by Systems Biology Approaches

The prevalence of coronary heart disease （CHD） is increasing, and has been a severe burden on society and family worldwide. New ideas need to be achieved for developing more efficacious and safe therapies to treat CHD. chinese medicine （CM） uses multicomponent drugs to prevent disease and ameliorate symptoms based on patients＇ different syndromes. The benefit of CM in CHD has recently been proven by increasing clinical evidence. More importantly, linking CM syndrome differentiation and biomedical diagnosis might provide innovative thinking for treating CHD. According to epidemiological investigations, blood stasis syndrome （BSS） is the major type of syndrome in CHD. Investigating the biomedical mechanisms of BSS of CHD is a topic of CM research. Because the holistic perspective of systems biology is well matched with CM, the application of omics techniques and other integrative approaches appears inherently appropriate. A wide range of omics techniques, including transcriptomics and proteomics, have been used in studies of BSS of CHD to search for a common ground of understanding. These approaches could be useful for understanding BSS of CHD from clinical and biological viewpoints. Nevertheless, current studies mainly contain results from a single approach, and they have not achieved the holistic, systematic and integrative concept of system biology. Therefore, we discuss the progress and challenges in exploring the biomedical mechanisms of BSS of CHD by systems biology approaches. With further development of systems biology, a better platform to study BSS of CHD may be provided, and biomarkers for BSS of CHD and therapeutic targets may be found. The study of BSS of CHD by systems biology approaches will also be beneficial for developing personalized treatment for BSS of CHD patients.
