Homeodomain proteins POU-M2, antennapedia and abdominal-B are involved in regulation of the segment-specific expression of the clip-domain serine protease gene CLIP13 in the silkworm, Bombyx mori

Clip-domain serine proteases (CLIPs) play important roles in insect innate immunity and development. Our previous studies indicated that CLIP13, an epidermis-specific gene, was involved in cuticle remodeling during molting and metamorphosis in the silkworm, Bombyx mori. However, the transcriptional regulatory mechanism and regulatory pathways of CLIP13 remained unclear. In the present study, we investigated CLIP13 expression and the regulation pathway controlled by 20-hydroxyecdysone (20E) in the silkworm. At the transcriptional level, expression of CLIP13 exhibited pronounced spatial and temporal specificity in different regions of the epidermis;homeodomain transcription factors POU-M2, antennapedia (Antp), and abdominal-B (Abd-B) showed similar expression change trends as CLIP13 in the head capsule, thorax, and abdomen, respectively. Furthermore, results of cell transfection assays, electrophoretic mobility shift assays, and chromatin immunoprecipitation demonstrated that POU-M2, Antp, and Abd-B were involved in the transcriptional regulation of CLIP13 by directly binding to their cis-response elements in CLIP13 promoter. RNA interference-mediated silencing of POU-M2, Antp, and Abd-B led to a decrease of CLIP13 expression in the head capsule, the epidermis of the 1st to 3rd thoracic segments and the 7th to 10th abdominal segments, respectively. Consistent with CLIP13, 20E treatment significantly upregulated expression of POU-M2, Antp, and Abd-B in the silkworm epidermis. Taken together, these data suggest that 20E positively regulates transcription of CLIP13 via homeodomain proteins POU-M2, Antp, and Abd-B in different regions of the silkworm epidermis during metamorphosis, thus affecting the molting process. Our findings provide new insight into the functions of homeodomain transcription factors in insect molting.

Alternative splicing of a carboxyl/choline esterase gene enhances the fenpropathrin tolerance of Tetranychus cinnabarinus

Detoxification plays a crucial role in agricultural pests to withstand pesticides,and cytochrome P450s,carboxyl/choline esterases(CCEs),and glutathione-S-transferases are the main proteins responsible for their detoxification *** activity of CCEs can be upregulated,downregulated,or modified by ***,few studies have examined the role of alternative splicing in altering the properties of *** identified 2 variants of TcCCE23 in Tetranychus cinnabarinus:a long version(CCE23-V1)and a short version that is 18 nucleotides shorter than CCE23-V1(CCE23-V2).Whether splicing affects the activity of TcCCE23 remains *** of CCE23-V2 in fenpropathrin-resistant *** revealed that splicing affected the detoxification of fenpropathrin by *** mortality of mites was significantly higher when the expression of CCE23-V2 was knocked down(43.2%±3.3%)via injection of CCE23-dsRNA(double-stranded RNA)compared with the control group injected with green fluorescent protein-dsRNA under fenpropathrin exposure;however,the downregulation of CCE23-V1(61.3%±6.3%)by CCE23-small interfering RNA had no such effect,indicating CCE23-V2 plays a greater role in xenobiotic metabolism than *** tolerance of flies overexpressing CCE23-V2 to fenpropathrin(50%lethal dose[LD_(50)]=19.47μg/g)was significantly higher than that of Gal4/UAS-CCE23-V1 transgenic flies(LD_(50)=13.11μg/g).Molecular docking analysis showed that splicing opened a“gate”that enlarges the substrate binding cavity of CCE23-V2,might enhance the ability of CCE23-V2 to harbor fenpropathrin *** findings suggest that splicing might enhance the detoxifying capability of ***,our data improve the understanding of the diversity and complexity of the mechanisms underlying the regulation of CCEs.

Mechanism of Diabatic Heating on Precipitation and the Track of a Tibetan Plateau Vortex over the Eastern Slope of the Tibetan Plateau

Existing studies contend that latent heating(LH)will replace sensible heating(SH)to become the dominant factor affecting the development of the Tibetan Plateau vortex(TPV)after it moves off the Tibetan Plateau(TP).However,in the process of the TPV moving off the TP requires that the airmass traverse the eastern slope of the Tibetan Plateau(ESTP)where the topography and diabatic heating(DH)conditions rapidly *** LH gradually replaces SH to become the dominant factor in the development of the TPV over the ESTP is still not very *** this paper,an analysis of a typical case of a TPV with a long life history over the ESTP is performed by using multi-sourced meteorological data and model *** results show that SH from the TP surface can change the TPV-associated precipitation distribution by temperature advection after the TPV moves off the *** LH can then directly promote the development of the TPV and has a certain guiding effect on the track of the *** SH can control the active area of LH by changing the falling area of the TPV-associated precipitation,so it still plays a key role in the development and tracking of the TPV even though it has moved out of the main body of the TP.

Formation evaluation and rock type classification of Asmari Formation based on petrophysical-petrographic data:A case study in one of super fields in Iran southwest

The Oligo-Miocene Asmari Formation is one of the most important hydrocarbon reservoirs in the Middle *** oilfield under study is one of the largest oilfields in the Zagros basin with the Asmari Formation being the major reservoir *** this study,petrographic analyses,petrophysical data and neural network clustering techniques were used for identifying rock types in the Asmari *** analysis of the Asmari Formation in the study area has resulted in the definition of 1 clastic lithofacies and 14 carbonate microfacies *** petrophysical logs from 43 wells and their correlation with capillary pressure(Pc)curves,led to the recognition of 7 electrofacies(EF1-EF7).Microscopic evidence of Electrofacies group C1 and S1 show that the sedimentary facies of these electrofacies are most commonly found in restricted and shoal facies belts ***,petrographic studies show that the sedimentary facies of C2,C3,C4,S2 and S3 were formed in the open marine,Lagoon,and Tidal flat facies belt zone of homoclinal ramp sedimentary environment during the Oligo-Miocene based on relative sea level changes *** link between electrofacies and geological data indicated that both sedimentary and diagenetic processes controlled the reservoir quality of the Asmari ***,permeability and water saturation were used to estimate the reservoir quality of each *** 1 and 2 with high porosity and permeability,low water saturation is considered as the best reservoir with regard to sedimentary textures(dolowackestone and dolograinstone)and the effect of diagenetic processes such as dolomitization ***,growth framework and interparticle porosities are major in EF-2,while the intercrystalline porosity is the major type in *** 3 and 4 show low values of porosity,permeability and high percentage of water saturations,which characterizes them as poor reservoir ***,EF-5 is the only electrofacies in the siliciclastic parts of

The dual impact of climate change and human activities has precipitated a sharp decline in primate biodiversity *** is home to the most diverse primate species in the Northern hemisphere,which face severe ecological threats due to the expansion of modern agriculture,extensive exploitation and consumption of natural resources,and excessive land development during its transition from an agricultural to a modern *** response,China has implemented various ecological conservation measures,including habitat restoration and *** efforts have made substantial strides in biodiversity conservation,with certain regions witnessing an increase in primate *** the current study,we conducted a systematic review of historical documents and field research data related to Chinese primates,evaluating the endangered status of primate species in *** improvements in the habitats of most primate species and some population growth,many species still face severe threats,including declining and small *** such as the Myanmar snub-nosed monkey(Rhinopithecus strykeri),eastern black crested gibbon(Nomascus nasutus),and Hainan gibbon(***)remain particularly vulnerable due to their limited distribution ranges and extremely small *** scientific data,fragmented information,and not enough studies in conservation biology further compound the ***,there is a notable lack of detailed population monitoring data for species such as the Bengal slow loris(Nycticebus bengalensis),pygmy slow loris(***),Indochinese gray langur(Trachypithecus crepusculus),Shortridge’s langur(***),and capped langur(***),which hinders the development of practical and targeted conservation management ***,for national biodiversity conservation,there is an urgent need for specialized primate surveys,enhancing habitat protection and restoration,and increasing focus on cross-border conse

Pseudotyped lentiviral vectors:Ready for translation into targeted cancer gene therapy?

Gene therapy holds great promise for curing cancer by editing the deleterious genes of tumor cells,but the lack of vector systems for efficient delivery of genetic material into specific tumor sites in vivo has limited its full therapeutic potential in cancer gene *** the past two decades,increasing studies have shown that lentiviral vectors(LVs)modified with different glycoproteins from a donating virus,a process referred to as pseudotyping,have altered tropism and display cell-type specificity in transduction,leading to selective tumor cell *** feature of LVs together with their ability to enable high efficient gene delivery in dividing and non-dividing mammalian cells in vivo make them to be attractive tools in future cancer gene *** review is intended to summarize the status quo of some typical pseudotypings of LVs and their applications in basic anti-cancer studies across many *** opportunities of translating pseudotyped LVs into clinic use in cancer therapy have also been discussed.
