The association of isocarbophos and isofenphos with different types of glucose metabolism:The role of inflammatory cells

To investigate the associations between isocarbophos and isofenphos with impaired fasting glucose(IFG)and type 2 diabetes mellitus(T2DM),and to assess the mediation roles of inflammation *** were 2701 participants in the case-control study,including 896 patients with T2DM,900 patients with IFG,905 subjects with *** isocarbophos and isofenphos concentrationsweremeasured using gas chromatography and triple quadrupole tandem mass *** linear models were used to calculate the relationships between plasma isofenphos and isocarbophos levels with inflammatory factor levels and *** cell was used as mediators to estimate the mediating effects on the above *** and isofenphos were positively related with T2DM after adjusting for other *** odds ratio(95%confidence interval)(OR(95%CI))for T2DM was 1.041(1.015,1.068)and for IFG was 1.066(1.009,1.127)per unit rise in *** prevalence of T2DM increased by 6.4%for every 1 unit more of ln-isofenphos(OR(95%CI):1.064(1.041,1.087)).Additionally,a 100%rise in ln-isocarbophos was linked to 3.3%higher ln-HOMA2IR and a 0.029 mmol/L higher glycosylated hemoglobin(HbA1c)(95%CI:0.007,0.051).While a 100%rise in ln-isofenphos was linked to increase in ln-HOMA2 and ln-HOMA2IR of 5.8%and 3.4%,***,white blood cell(WBC)and neutrophilic(NE)were found to be mediators in the relationship between isocarbophos and T2DM,and the corresponding proportions were 17.12%and 17.67%,*** and isocarbophos are associated with IFG and T2DM in the rural Chinese population,WBC and NE have a significant role in this relationship.

The emerging role of mesenchymal stem cell-derived extracellular vesicles to ameliorate hippocampal NLRP3 inflammation induced by binge-like ethanol treatment in adolescence

Our previous studies have reported that activation of the NLRP3(NOD-,LRR-and pyrin domain-containing protein 3)-inflammasome complex in ethanol-treated astrocytes and chronic alcohol-fed mice could be associated with neuroinflammation and brain *** stem cell-derived extracellular vesicles(MSC-EVs)have been shown to restore the neuroinflammatory response,along with myelin and synaptic structural alterations in the prefrontal cortex,and alleviate cognitive and memory dysfunctions induced by binge-like ethanol treatment in adolescent *** the therapeutic role of the molecules contained in mesenchymal stem cell-derived extracellular vesicles,the present study analyzed whether the administration of mesenchymal stem cell-derived extracellular vesicles isolated from adipose tissue,which inhibited the activation of the NLRP3 inflammasome,was capable of reducing hippocampal neuroinflammation in adolescent mice treated with binge *** demonstrated that the administration of mesenchymal stem cell-derived extracellular vesicles ameliorated the activation of the hippocampal NLRP3 inflammasome complex and other NLRs inflammasomes(e.g.,pyrin domain-containing 1,caspase recruitment domain-containing 4,and absent in melanoma 2,as well as the alterations in inflammatory genes(interleukin-1β,interleukin-18,inducible nitric oxide synthase,nuclear factor-kappa B,monocyte chemoattractant protein-1,and C–X3–C motif chemokine ligand 1)and miRNAs(miR-21a-5p,miR-146a-5p,and miR-141-5p)induced by binge-like ethanol treatment in adolescent *** analysis further revealed the involvement of miR-21a-5p and miR-146a-5p with inflammatory target genes and NOD-like receptor signaling *** together,these findings provide novel evidence of the therapeutic potential of MSC-derived EVs to ameliorate the hippocampal neuroinflammatory response associated with NLRP3 inflammasome activation induced by binge drinking in adolescence.

Ozone exposure and cardiovascular disease: A narrative review of epidemiology evidence and underlying mechanisms

Ozone (O3) poses a significant global public health concern as it exerts adverse effects on human cardiovascular health. Nevertheless, there remains a lack of comprehensive understanding regarding the relationships between O3 exposure and the risk of cardiovascular diseases (CVD), as well as the underlying biological mechanisms. To address this knowledge gap, this narrative review meticulously summarizes the existing epidemiological evidence, susceptibility, and potential underlying biological mechanisms linking O3 exposure with CVD. An increasing body of epidemiological studies has demonstrated that O3 exposure heightens the incidence and mortality of CVD, including specific subtypes such as ischemic heart disease, hypertension, and heart failure. Certain populations display heightened vulnerability to these effects, particularly children, the elderly, obese individuals, and those with pre-existing conditions. Proposed biological mechanisms suggest that O3 exposure engenders respiratory and systemic inflammation, oxidative stress, disruption of autonomic nervous and neuroendocrine systems, as well as impairment of coagulation function, glucose, and lipid metabolism. Ultimately, these processes contribute to vascular dysfunction and the development of CVD. However, some studies have reported the absence of associations between O3 and CVD, or even potentially protective effects of O3. Inconsistencies among the literature may be attributed to inaccurate assessment of personal O3 exposure levels in epidemiologic studies, as well as confounding effects stemming from co-pollutants and temperature. Consequently, our findings underscore the imperative for further research, including the development of reliable methodologies for assessing personal O3 exposure, exploration of O3 exposure's impact on cardiovascular health, and elucidation of its biological mechanisms.

Association of Co-Exposure to Polycyclic Aromatic Hydrocarbons and Metal(loid)s with the Risk of Neural Tube Defects:A Case-Control Study in Northern China

Objective Exposure to polycyclic aromatic hydrocarbons(PAHs)or metal(loid)s individually has been associated with neural tube defects(NTDs).However,the impacts of PAH and metal(loid)co-exposure and potential interaction effects on NTD risk remain *** conducted a case-control study in China among population with a high prevalence of NTDs to investigate the combined effects of PAH and metal(loid)exposures on the risk of *** Cases included 80 women who gave birth to offspring with NTDs,whereas controls were 50 women who delivered infants with no congenital *** analyzed the levels of placental PAHs using gas chromatography and mass spectrometry,PAH-DNA adducts with 32P-post-labeling method,and metal(loid)s with an inductively coupled plasma mass *** logistic regression was employed to estimate the associations between individual exposures and *** absolute shrinkage and selection operator(LASSO)penalized regression models were used to select a subset of exposures,while additive interaction models were used to identify interaction *** In the single-exposure models,we found that eight PAHs,PAH-DNA adducts,and 28 metal(loid)s were associated with ***,selenium,molybdenum,cadmium,uranium,and rubidium were selected through LASSO regression and were statistically associated with NTDs in the multiple-exposure *** with high levels of pyrene and molybdenum or pyrene and selenium exhibited significantly increased risk of having offspring with NTDs,indicating that these combinations may have synergistic effects on the risk of *** Our findings suggest that individual PAHs and metal(loid)s,as well as their interactions,may be associated with the risk of NTDs,which warrants further investigation.

For a double array {V_(m,n), m ≥ 1, n ≥ 1} of independent, mean 0 random elements in a real separable Rademacher type p(1 ≤ p ≤ 2) Banach space and an increasing double array {b_(m,n), m ≥1, n ≥ 1} of positive constants, the limit law ■ and in L_p as m∨n→∞ is shown to hold if ■ This strong law of large numbers provides a complete characterization of Rademacher type p Banach spaces. Results of this form are also established when 0 < p ≤ 1 where no independence or mean 0 conditions are placed on the random elements and without any geometric conditions placed on the underlying Banach space.

Genetic and environmental risk factors for primary open-angle glaucoma

Background Primary open-angle glaucoma (POAG) is characterized by optic nerve damage and consists of a group of genetically heterogeneous disorders. This study was to investigate the associations of genetic and environmental factors with POAG in a hospital-based Chinese population Methods Thirty-two adult onset POAG patients and 96 age-sex matched control subjects were studied by multivariable logistic regression analysis for the relationships between POAG and its risk factors including family history, diabetes, hypertension, cardiovascular diseases, cigarette smoking, alcohol consumption and polymorphisms of the myocilin and the optineurin genes Results Univariate analysis showed that POAG was related to family history, cardiovascular disease, alcohol consumption and a myocilin sequence alteration (T353I) ( P <0 04) Multivariable logistic regression analysis confirmed that POAG was significantly associated with family history ( OR =20 2), hypertension ( OR =3 58), cigarette smoking ( OR =10 8), alcohol consumption ( OR =0 028) and T353I ( OR =6 03, all P <0 05) Conclusions Family history, hypertension, cigarette smoking and T353I in the myocilin gene are risk factors for POAG Alcohol consumption, however, has a protective effect

PacBio Sequencing and Its Applications

Single-molecule, real-time sequencing developed by Pacific BioSciences offers longer read lengths than the second-generation sequencing （SGS） technologies, making it well-suited for unsolved problems in genome, transcriptome, and epigenetics research. The highly-contiguous de novo assemblies using PacBio sequencing can close gaps in current reference assemblies and characterize structural variation （SV） in personal genomes. With longer reads, we can sequence through extended repetitive regions and detect mutations, many of which are associated with dis- eases. Moreover, PacBio transcriptome sequencing is advantageous for the identification of gene isoforms and facilitates reliable discoveries of novel genes and novel isoforms of annotated genes, due to its ability to sequence full-length transcripts or fragments with significant lengths. Addition- ally, PacBio＇s sequencing technique provides information that is useful for the direct detection of base modifications, such as methylation. In addition to using PacBio sequencing alone, many hybrid sequencing strategies have been developed to make use of more accurate short reads in conjunction with PacBio long reads. In general, hybrid sequencing strategies are more affordable and scalable especially for small-size laboratories than using PacBio Sequencing alone. The advent of PacBio sequencing has made available much information that could not be obtained via SGS alone.

Though a relatively young discipline, translational bioinformatics （TBI） has become a key component of biomedical research in the era of precision medicine. Development of high-throughput technologies and electronic health records has caused a paradigm shift in both healthcare and biomedical research. Novel tools and methods are required to convert increasingly voluminous datasets into information and actionable knowledge. This review provides a definition and contex- tualization of the term TBI, describes the discipline＇s brief history and past accomplishments, as well as current loci, and concludes with predictions of future directions in the field.

Incorporating functional annotation information in prioritizing disease associated SNPs from genome wide association studies

With recent advances in genotyping and sequencing technologies,many disease susceptibility loci have been ***,much of the genetic heritability remains unexplained and the replication rate between independent studies is still ***,there have been increasing efforts on functional annotations of the entire human genome,such as the Encyclopedia of DNA Elements(ENCODE)project and other similar *** has been shown that incorporating these functional annotations to prioritize genome wide association signals may help identify true association ***,to our knowledge,the extent of the improvement when functional annotation data are considered has not been studied in the *** this article,we propose a statistical framework to estimate the improvement in replication rate with annotation data,and apply it to Crohn’s disease and DNase I hypersensitive *** results show that with cell line specific functional annotations,the expected replication rate is improved,but only at modest level.
