Identification and Expression of Some Plant Cell Wall-Degrading Enzymes Present in Three Ontogenetics Stages of Thecaphora frezii, a Peanut (<i>Arachis hypogaea</i>L.) Pathogenic Fungus

Peanuts can be affected by the presence of pathogenic microorganisms. The fungus Thecaphora frezii (T. frezii), which belongs to the taxonomic class Ustilaginomycetes, is the causal agent of the disease known as “peanut smut”. The life cycle of this fungus includes three stages, namely teliospores, basidiospores and hyphae. In the hyphae stage, infection occurs in the peanut plant, which requires the involvement of some enzymes secreted by the fungus. These include the Plant Cell Wall-Degrading Enzymes (PCWDEs), which degrade various polysaccharides. This study aimed to identify the presence of transcript for enzymes belonging to the PCWDEs from three stages of T. frezii. For this, total RNA was extracted from the three ontogenetic stages of T. frezii. These samples were analyzed using an RNA-Seq approach and some transcripts were quantified using real Time PCR. The analysis of the data provided by the RNA-Seq of the three T. frezii stages, it was possible to identify some transcripts that could encode enzymes compatible with polysaccharides degradation that are part of the plant cell wall. In T. frezii transcriptome, 40 deduced proteins would be enzymes with functions of PCWDEs were identified. They were divided into 27 glycoside hydrolases;two polysaccharide lyases;three carbohydrate esterases and eight enzymes with auxiliary activities. In addition, the fungal SNF1 gene was identified whose activity could be affected by high glucose level, and indirectly influence the levels of some PCWDEs. The analysis of the PCWDEs could help to understand part of the fungal infection process and possibly find substances that can control its development.

Physiopathological Mechanism and Assessment of Fetal Asphyxia

Treatment and outcome of childbirth depend on the acidobasic balance of the fetal blood related to the oxygen and carbon dioxide level. Hypoxemia could lead to asphyxia that is why fetal monitoring and biochemical parameters assessment are mandatory. Although there are compensatory mechanisms that temporarily protect the fetus, there are also other factors that interfere with the oxygenation of the fetus and determine the development of the fetus and the newborn. Actually, the level of the oxygen, the carbon dioxide, the acidobasic balance and the pH are the cornerstones of the well-being of the fetus.

This work presents a moving mesh methodology based on the solution of a pseudo flow *** mesh motion is modeled as a pseudo Stokes problem solved by an explicit finite element projection *** mesh quality requirements are satisfied by employing a null divergent velocity *** methodology is applied to triangular unstructured meshes and compared to well known approaches such as the ones based on diffusion and pseudo structural *** of the test cases is an airfoil with a fully meshed domain.A specific rotation velocity is imposed as the airfoil boundary *** other test is a set of two cylinders that move toward each other.A mesh quality criteria is employed to identify critically distorted elements and to evaluate the performance of each mesh motion *** results obtained for each test case show that the pseudo-flow methodology produces satisfactory meshes during the moving process.

Resolution of metabolic syndrome after following a gluten free diet in an adult woman diagnosed with celiac disease

Adult celiac disease(CD) presents with very diverse symptoms that are clearly different from those typically seen in pediatric patients,including ferropenic anemia,dyspepsia,endocrine alterations and elevated transaminase concentration. We present the case of a 51-year-old overweight woman with altered basal blood glucose,hypercholesterolemia,hypertriglyceridemia and persisting elevated transaminase levels,who showed all the symptoms for a diagnosis of metabolic syndrome. Because she presented iron deficiency anemia,she was referred to the gastroenterology department and subsequently diagnosed with celiac disease after duodenal biopsies and detection of a compatible HLA haplotype. Gluten-free diet(GFD) was prescribed and after 6 mo the patient showed resolution of laboratory abnormalities(including recovering anemia and iron reserves,normalization of altered lipid and liver function parameters and decrease of glucose blood levels) . No changes in weight or waist circumference were observedand no significant changes in diet were documented apart from the GFD. The present case study is the first reported description of an association between CD and metabolic syndrome,and invites investigation of the metabolic changes induced by gluten in celiac patients.

Determination of sildenafil by preconcentration on surfactant coated polymeric resin followed by spectrofluorimetry

The illicit addition of phosphodiesterase type-5 （PDE-5） inhibitors like sildenafil （Viagra） in product offered as herbal medicine or dietary supplement for male erectile dysfunction has concerned authorities in recent times. In this paper, we proposed a sensitive surfactant-coated Amberlite XADTM resin for sildenafil preconcentration method with spectrofluorimetric detection. Retention capacity of micellar coated XAD resin for sildenafil was studied and the obtained eluate was measured by spectrofluorometer at excitation and emission wavelengths of 350 and 430 nm, respectively. This method allowed the detection of sildenafil at 0.15 ng/mL with linear range of 0.0003-7.0 μg/mL. The method has been successfully applied to the analysis of some local commercially available herbal medicines and urine.

Induction of Extracellular Lytic Enzymes by <i>Fusarium solani</i>

Fusarium solani is a necrotrophic parasitic fungus that causes wilt in some plants, causing severe economic losses in some areas of the country. The objective of this work was to analyze the induction of extracellular lytic enzymes produced by a strain of F. solani, isolated from a culture of tomato, in Villa de Arista, S.L.P. México. Polygalacturonase activity has a greater induction time at 10 days, and the xylanase has two times higher activity at 8 and 13 days of incubation at 28?C. Also, the xylanase activities A and B were very stable at 4?C. After 7 days of incubation, it has an activity of 100% and 96%, respectively, while polygalacturonase retains 61% of its initial activity. Both activities are better induced with glutamate and urea as nitrogen sources respectively, and both exhibit an initial pH optimum of 5.5. Finally, we didn’t find cellulase activity in the analyzing conditions.

Hexavalent Chromium Removal by Citrus limonium Shell

We studied the Chromium(VI) removal capacity in aqueous solution by the lemon shell, using the diphenylcarbazide method to evaluate the metal concentration. So, the highest biosorption of the metal (50 mg/L) occurs within 100 minutes, at pH of 1.0, and 28°C. According to temperature, the highest removal was observed at 60°C, in 11 minutes, when the metal (1 g/L) is completely adsorbed. At the analyzed concentrations of Cr(VI), lemon shell, showed excellent removal capacity, besides it removes efficiently the metal in situ (97.2% removal, 7 days of incubation, 5 g of biomass). After 1 hour of incubation the studied biomass reduces 1.0 g of Cr(VI) with the simultaneous production of Cr(III);so it can be used to eliminate it from industrial wastewater.

Ectopic lignification in primary cellulose-deficient cell walls of maize cell suspension cultures

Maize （Zea mays L.） suspension-cultured cells with up to 7o% less cellulose were obtained by stepwise habituation to dichlobenil （DCB）, a cellulose biosynthesis inhibitor. Cellulose deficiency was accompanied by marked changes in cell wall matrix polysaccharides and phenolics as revealed by Fourier transform infrared （FTIR） spectroscopy. Cell wall compositional analysis indicated that the cellulosedeficient cell walls showed an enhancement of highly branched and cross-linked arabinoxylans, as well as an increased content in ferulic acid, diferulates and p-coumaric acid, and the presence of a polymer that stained positive for phloroglucinol. In accordance with this, cellulose-deficient cell walls showed a fivefold increase in Klason-type lignin. Thioacidolysis/GC-MS analysis of cellulose-deficient cell walls indicated the presence of a lignin-like polymer with a Syringyl/Guaiacyl ratio of 1.45, which differed from the sensu stricto stress-related lignin that arose in response to shortterm DCB-treatments. Gene expression analysis of these ceils indicated an overexpression of genes specific for the biosynthesis of monolignol units of lignin. A study of stress signaling pathways revealed an overexpression of some of the iasmonate signaling pathway genes, which might trigger ectopic lignification in response to cell wall integrity disruptions. In summary, the structural plasticity of primary cell walls is proven, since a lignification process is possible in response to cellulose impoverishment.

Bioactive Compounds and Insecticidal Activity of Hysterionica pinifolia, a Native South American Plant

Phytochemical investigation of the purified fractions of the active dichloromethane extract of Hysterionica pinifolia (Poir.) Baker led to the identification of five compounds. New acetylenic alcohol (E)-undec-3-en-5,7-diyne-1-ol (1) and three other compounds (3), (4), and (5) were reported for the first time in this species. Furthermore, forty-six components from the volatile fraction of H. pinifolia were identified. These compounds were elucidated using 1D and 2D NMR spectroscopy as well as MS-ESI and GC-FID-MS experiments. The dichloromethane extract, its fractions, and the methanolic extract were tested for insecticidal activity against Tribolium castaneum under laboratory conditions. The dichloromethane extract and the fraction F2 were found to be active, showing high larval mortality. The dichloromethane extract was also active against T. castaneum adults. The results have shown that H. pinifolia could be considered, in a near future, as a potential source for the development of a botanical insecticide for pest control.

Performance of cold-preserved rat liver Microorgans as the biological component of a simplified prototype model of bioartificial liver

AIM To develop a simplified bioartificial liver(BAL) device prototype, suitable to use freshly and preserved liver Microorgans(LMOs) as biological component. METHODS The system consists of 140 capillary fibers through which goat blood is pumped. The evolution of hema-tocrit, plasma and extra-fiber fluid osmolality was evaluated without any biological component, to characterize the prototype. LMOs were cut and cold stored 48 h in BG35 and Via Span~? solutions. Fresh LMOs were used as controls. After preservation, LMOs were loaded into the BAL and an ammonia overload was added. To assess LMOs viability and functionality, samples were taken to determine lactate dehydrogenase(LDH) release and ammonia detoxification capacity. RESULTS The concentrations of ammonia and glucose, and the fluids osmolalities were matched after the first hour of perfusion, showing a proper exchange between blood and the biological compartment in the minibioreactor. After 120 min of perfusion, LMOs cold preserved in BG35 and Via Span~? were able to detoxify 52.9% ± 6.5% and 53.6% ± 6.0%, respectively, of the initial ammonia overload. No significant differences were found with Controls(49.3% ± 8.8%, P < 0.05). LDH release was 6.0% ± 2.3% for control LMOs, and 6.2% ± 1.7% and 14.3% ± 1.1% for BG35 and Via Span~? cold preserved LMOs, respectively(n = 6, P < 0.05). CONCLUSION This prototype relied on a simple design and excellent performance. It’s a practical tool to evaluate the detoxification ability of LMOs subjected to different preservation protocols.
