Expression of CDC42 in cervical squamous cell carcinoma and its correlation with clinicopathologic characteristics

Objective： The high expression of cell division cycle 42 protein （CDC42） may be involved in the occurrence and progression of several tumors. However, the expression and function of CDC42 in cervical squamous cell carcinoma remains unclear. This study aimed to investigate the expression of CDC42 in cervical squamous cell carcinoma and its correlation with clinicopathologic characteristics. Methods： The expression of CDC42 in 162 cervical squamous cell carcinoma tissue samples and 33 normal cervical tissue samples was investigated by immunohistochemistry. The CDC42 mRNA expression was detected by reverse transcription-polymerase chain reaction （RT-PCR）. Results： The cervical squamous cell carcinoma group showed a significantly higher CDC42 positive rate, compared to the normal cervical tissues （P〈0.05）. Fttrthermore, the tissues of stage Ⅱ-Ⅳ carcinoma patients showed higher CDC42 expression levels compared to stage I patients （P=0.05）. In addition, the expression of CDC42 was not correlated to age of patients, differentiation degree of cancer cells, or lymph node metastasis （P〉0.05）. Furthermore, compare with normal cervical tissues, the CDC42 mRNA expression in cervical cancer had no significant difference. Conclusions： CDC42 was up-regulated at protein level, but not mRNA level, in cervical squamous cell carcinoma. The high expression of CDC42 was correlated to the clinical stage of the patients, indicating that CDC42 might contribute to the progression of cervical squamous cell carcinoma.

Downregulation of the nucleosome-binding protein 1 （NSBP1） gene can inhibit the in vitro and in vivo proliferation of prostate cancer cells

This studay is to construct a lentiviral vector harbouring an RNA interference （RNAi） sequence that targets the gene encoding the human high-mobility group nucleosomal binding protein 1 （NSBP1）; to study its role in inducing G2/M phase arrest and apoptosis in prostate cancer （PCa） DU145 cells; and to assess the effect of its knockdown on cell proliferation in vitro and in vivo. RNAi was applied to knock down NSBP1 expression in the PCa cell line DU145 by lentiviral plasmids producing an NSBP1 small hairpin RNA. After NSBP1 knockdown in DU145 cells, the growth rate of cells was analyzed by MTT, and G2/M cell cycle arrest and apoptosis were assessed using a FAC- Scalibur flow cytometer. Tumour growth was assessed in nude mice. The mRNA and protein expression levels of NSBP1, cyclin B1 and Bcl-2 were analysed in vitro and in vivo by reverse-transcriptase polymerase chain reaction and Western blotting. Knockdown of NSBP1 resulted in a 22.6% decrease in the growth rate of cells compared with the PscNC lentivirus control cells at 96 h, decreased tumour growth in nude mice, and the induction of Gz/M cell cycle arrest （8.78%） and apoptosis （2.19-fold）. Consistent with the cell cycle arrest and apoptosis, the mRNA and protein expression levels of cyclin B 1 and Bcl-2 were decreased. In conclusion, knockdown of NSBP 1 causes a statistically significant inhibition of the in vitro and in vivo growth of the PCa cell line DU145. Growth suppression is at least partially due to NSBP1 knockdown-induced Gz/M cell cycle arrest and apoptosis. The present data provide the evidence that the NSBP1 knockdown-induced G2/M phase arrest and apoptosis may result from negative regulation of cyclin B 1 and Bcl-2 by NSBP1, with the resulting reduced expression of these proteins.

研究目的:探讨碎裂QRS波对急性心肌梗死患者的短期预后评估的价值,及其对ST段抬高和非ST段抬高型心肌梗死(STEMI和NSTEMI)患者不同的预测价值。创新要点:证实了碎裂QRS波对急性心肌梗死尤其是ST段抬高型心肌梗死的短期预后价值,对早期再灌注治疗有指导意义。研究方法:通过对碎裂QRS波发生及持续时间的详细记录(见图1、2),评价出现碎裂QRS波的STEMI及NSTEMI患者住院期间发生恶性心律失常、泵功能衰竭及住院死亡的风险(见图3;表3–5),并观察早期经皮冠状动脉介入治疗(PCI)对预后的不同影响(见表6)。重要结论:1.急性心肌梗死(尤其是STEMI)患者心电图出现碎裂QRS波短期内易发生恶性心律失常、左心室功能不全及死亡;2.对出现碎裂QRS波的急性心肌梗死患者,早期PCI再灌注治疗可改善患者短期预后。

Transient and weak protein–protein interactions are essential to many biochemical reactions, yet are technically challenging to study. Chemical cross-linking of proteins coupled with mass spectrometry analysis(CXMS) provides a powerful tool in the analysis of such interactions. Central to this technology are chemical cross-linkers. Here, using two transient heterodimeric complexes EIN/HPr and EIIAGlc/EIIBGlc as our model systems, we evaluated the effects of two amine-specific homo-bifunctional cross-linkers with different reactivities. We showed previously that DOPA2(di-ortho-phthalaldehyde with a di-ethylene glycol spacer arm) cross-links proteins 60–120 times faster than DSS(disuccinimidyl suberate). We found that though most of the intermolecular cross-links of either cross-linker are consistent with the encounter complexes(ECs), an ensemble of short-lived binding intermediates,more DOPA2 intermolecular cross-links could be assigned to the stereospecific complex(SC), the final lowest-energy conformational state for the two interacting proteins. Our finding suggests that faster cross-linking captures the SC more effectively and cross-linkers of different reactivities potentially probe protein–protein interaction dynamics across multiple timescales.

A new direct method for solving unsymmetrical sparse linear systems（USLS） arising from meshless methods was introduced. Computation of certain meshless methods such as meshless local Petrov-Galerkin （MLPG） method need to solve large USLS. The proposed solution method for unsymmetrical case performs factorization processes symmetrically on the upper and lower triangular portion of matrix, which differs from previous work based on general unsymmetrical process, and attains higher performance. It is shown that the solution algorithm for USLS can be simply derived from the existing approaches for the symmetrical case. The new matrix factorization algorithm in our method can be implemented easily by modifying a standard JKI symmetrical matrix factorization code. Multi-blocked out-of-core strategies were also developed to expand the solution scale. The approach convincingly increases the speed of the solution process, which is demonstrated with the numerical tests.

Intradural spinal seeding and fatal progression of a sacrococcygeal chordoma： a case report

Chordoma as a rare malignant tumor arising from remnants of the fetal notochord accounts for 1%-4%of primary bone tumors. It is usually predominant in males. Local recurrences are common （44%-78%） and distant metastases may occur years after the initial presentation. The reported rates of metastases range from 10% to 48%, which are usually accompanied by a sacrococcygeal chordoma. Cases of surgical seeding and intradural spinal seeding also have been reported climcally. To our knowledge, the present case is the first one concerning cerebrospinal fluid metastases and fatal progression of a sacrococcygeal chordoma.

DelayD roid: an instrumented approach to reducing tail-time energy of Android apps

Mobile devices with 3G/4G networking often waste energy in the so-called "tail time" during which the radio is kept on even though no communication is occurring. Prior work has proposed policies to reduce this energy waste by batching network requests. However, this work is challenging to apply in practice due to a lack of mechanisms. In response, we have developed DelayD roid, a framework that allows a developer to add the needed policy to existing, unmodified Android applications(apps) with no human effort as well as no SDK/OS changes. This allows such prior work(as well as our own policies) to be readily deployed and evaluated. The DelayD roid compile-time uses static analysis and bytecode refactoring to identify method calls that send network requests and modify such calls to detour them to the DelayD roid run-time. The run-time then applies a policy to batch them, avoiding the tail time energy waste. DelayD roid also includes a cross-app communication mechanism that supports policies that optimize across multiple apps running together, and we propose a policy that does so. We evaluated the correctness and universality of the DelayD roid mechanisms on14 popular Android apps chosen from the Google App Store. To evaluate our proposed policy, we studied three DelayD roid-enabled apps(weather forecasting, email client, and news client) running together, finding that the DelayD roid mechanisms combined with our policy can reduce 3G/4G tail time energy waste by 36%.

A reverse headgear is thought to be an effective .treatment approach for skeletal class Ⅲ malocclusion with retruded maxilla. Clinical studies have shown that the changes of occlusion are a combination of skeletal and dental changes： forward movement of the maxilla, proclination of the maxillary incisors, clockwise rotation of the mandible, and retroclination of the mandibular incisors. However, the undesirable dental effects, such as excessive mesial movement and extrusion of maxillary molars and labial tipping of maxillary incisors, have been reported. Most of previous studies have demonstrated that the best time for maxillary protraction is early mixed dentition. The skeletal effect decreases and the dental effect increases with age. So there is a need for an absolute stable anchorage for maxillary orthopedics to produce more skeletal change and less dental movement.

Aberrant Alternative Polyadenylation is Responsible for Survivin Up-regulation in Ovarian Cancer

Background： Survivin is an oncoprotein silenced in normal mature tissues but reactivated in serous ovarian cancer （SOC）. Although transcriptional activation is assumed for its overexpression, the long 3＇-untranslated region （3＇-UTR） in survivin gene, which contains many alternate polyadenylation （APA） sites, implies a propensity for posttranscriptional control and therefore was the aim of our study. Methods： The abundance of the coding region, the proximal and the distal region of survivin mRNA 3＇-UTR, was evaluated by real-time polymerase chain reaction （PCR） in SOC samples, cell lines, and normal fallopian tube （NFT） tissues. The APA sites were confirmed by rapid amplification ofcDNA 3＇ ends and DNA sequencing. Real-time PCR were used to screen survivin-targeting microRNAs （miRNAs） that were inversely correlated with survivin. The expression of an inversely correlated miRNA was restored by pre-miRNA transfection or induction with a genotoxic agent to test its inhibitory effect on survivin overexpression. Results： Varying degrees of APA were observed in SOC by comparing the abundance of the proximal and the distal region of survivin 3＇-UTR, and changes of 3＇-UTR correlated significantly with survivin expression （r = 0.708, P 〈 0.01）. The main APA sites are proved at 1197 and 1673 of survivin 3＇-UTR by DNA sequencing. Higher level of 3＇-UTR proximal region than coding region was observed in NFT, as well as in SOC and cell lines. Among the survivin-targeting miRNAs, only a few highly expressed miRNAs were inversely correlated with survivin levels, and they mainly targeted the distal part of the 3＇-UTR. However, in ovarian cancer cells, restoration of an inversely correlated miRNA （miR-34c） showed little effect on survivin expression. Conclusions： In NFT tissues, survivin is not transcriptionally silenced but regulate posttranscriptionally.
