It is critical to acknowledge that the coexistence of genes related to carbapenem and polymyxin resistance can exacerbate challenges in antimicrobial *** this study,we identified Klebsiella pneumoniae isolate L2425 fromthe feces of an ICU patient in *** isolate belongs to sequence type 11 and carries the antimicrobial resistance genes blaKPC-2,bla_(CTX-M-65) and *** S1-nuclease pulsed-field gel electrophoresis and Southern blotting analysis,we confirmed the presence of a~110 kb plasmid harboring the mcr-8 gene with a conserved structure(inhA-mcr-8-ompR-dgkA-IS903B).Furthermore,another~130 kb plasmid carrying bla_(KPC-2) was detected along with a composite transposon flanked by insertion sequence *** experiments demonstrated that both pL2425-KPC-2 and pL2425-MCR-8 plasmids were ***,this is the first instance of clinical fecal samples from China containing a *** strain carrying both blaKPC-2 and mcr-8 *** findings underscore the significance of surveillance for carbapenemases and mobile colistin–resistance variants.

Effect of a single dose of malathion on spermatogenesisin mice

Aim: To observe the acute effect of the organophosphorous insecticide malathion on testicular function in mice. Methods: The effects of a single dose of malathion [240 mg/kg (1/12 LD50)] on plasma acetylcholinesterase (ACE) activity, spermatozoa (epididymal cauda counts and teratozoospermia), testis and plasma testosterone concentration) were evaluated at day 1,8, 16, 35 and 40 after treatment. Results: The sperm count was decreased significantly 24 h after treatment and teratozoospermia was increased at day 35 and 40. The height of the seminiferous epithelium and the diameter of tubular lumen were decreased at day 8. The percentage of tubular blockade was increased between day 8 and 35. A decrease in testosterone plasma level was observed at day 16 after treatment. Conclusion: Malathion damages male reproduction. The depletion of seminiferous tubules and the increase in teratozoospermia may be a genotoxic damage to the renewing spermatogonia, but the possibility of spermatogenic/ spermiogenic disfunction due to a decrease in the plasma testosterone level can not be ruled out.

Aim: To explore the effect of exposure to commercial Parathion~ (Pc) on the reproductive parameters (sperm and cocoon production and genotoxicity on male germ cells), the survival, the body weight and the gross anatomical changes in Eiseniafoetida. Methods: Three doses of Pc(1478, 739 and 444 mg/kg of soil) and three time intervals of exposure (5, 15 and 30 days) were used. Results: All treated animals were affected. An acute genotoxic effect, revealed by DNA fragmentation (comet assay), was seen by 5 days. Alterations in reproductive parameters were conspicuous in regard to the number of sperm, cocoons and worms born, and the histological observation of the gonads and seminal receptacles. In addition, the body weight and survival rate were decreased. Neuromuscular function was also affected. Conclusion: Earthworms are suitable bioindicators of chemical contamination of the soil, their advantage being their easy and economical handling

Evolution of plant reproduction:From fusion and dispersal to interaction and communication

Based on the existing data concerning the evolution of the sexual reproduction, it is argued that the processes of sex differentiation and interactions play a key role in evolution. From the beginning environment and organism are unified. In a changing dynamic environment life originates and the interaction between life and environment develops from simple to more complex organisms. Sexual reproduction is introduced after the origin of meiosis and is a key process in evolution. The asexual reproduction process prepares to dispersal. Sexual reproduction process adds the genome renewal and the gamete-gamete interaction. Reproduction and dispersal are connected and the process of reproduction has similarities between asexual and sexual reproduction. Unicellular algae develop the physiological and morphological sex differentiation. Sex differentiation is connected with the way of dispersal. The step to multicellular plants introduces cell isolation after meiosis and by the stay on the mother plant within a cell or organ, plant-cell apoplastic interaction originates and by prolonged stay the plant-plant interaction. This stay influences the type of dispersal. A life cycle with alternation of generations and two moments of dispersal permits plants to go on land. In ferns a shift in the moment of sex differentiation to meiospore happens and the stay of the macrospore leads to the seed plants. In water all types of sexual reproduction, interactions and the alternation of generations are prepared and these are used to conquest land. On land the biotic dispersal is realized. The phylogeny of sexual reproduction reveals that the sex differentiation and interaction are the main causes in the evolution of sexual reproduction. Sexual reproduction shows interactions during gamete fusion, between organism and environment and in multicellular plants between organisms. Vitamin B12-induced spermatogenesis recovery in cimetidine-treated rats： effect on the spermatogonia number and sperm concentration

The H2-receptor antagonist cimetidine is an antiulcer drug also used for the treatment of cancer due to its antiangiogenic effect. However, this drug has caused structural changes in the seminiferous tubules. Vitamin B12 has been used as a therapeutic agent for the treatment of male infertility. The supplementation of rats with vitamin B12 during cimetidine treatment has recovered the damaged seminiferous tubules, but how this vitamin restores the seminiferous epithelium has not been clarified. In this study, we evaluated whether vitamin B12 improves the number of spermatogonia, spermatocytes, and sperm concentration in cimetidine-treated rats. Adult male rats were treated for 50 days as follows： cimetidine group received 100 mg kg^-1 b.w. of cimetidine, cimetidine-B12 group received cimetidine and 3 μg of vitamin B12-hydroxocobalamin, B12 group received only 3 μg of vitamin, and control group received saline. Sperm concentration was calculated and historesin-embedded testes sections were used for the quantitative analyses of spermatogonia （A; In/B） and spermatocytes. TUNEL method and PCNA immunofluorescence were performed. Cimetidine caused a significant reduction in sperm concentration. TUNEL-positive spermatogonia and spermatocytes were correlated to a significant reduction in the number of these cells. In cimetidine-B12 group, sperm concentration was higher than cimetidine group and a significant increase in the number of spermatogonia （stages Ⅱ-Ⅵ） was correlated to a high incidence of PCNA-immunolabeled spermatogonia and spermatocytes. The results show that the supplementation of rats with vitamin B12 during cimetidine treatment increases sperm concentration and exerts a potential effect in the recovery of spermatogonia and spermatocytes.

Electron probe microanalysis оf experimentally stimulated osteoarthrosis in dogs

AIM To develop methods of articular cartilage preparation for X-ray-electron probe microanalysis and to study its elements content in experimental *** Twenty dogs aged 2-8 years were divided in research(aged 2 years, induction of osteoarthrosis-IOA) and intact group. Intact group included three subgroups(aged 2, 5 and 8 years). Samples of cartilage after araldite saturation and pouring were partially cut into semithin sections stained with methylene blue and with methylene blue-basic fuchsin. Their smooth surfaces were investigated by X-ray-electron probe microanalysis. Spatial distribution of sulfur, calcium and phosphorus and their concentrations(weight %) were *** X-ray electron probe microanalysis revealed non-uniformsulfur distribution in cartilage of intact animals: Its content increases from superficial zone to deep one, this regularity was preserved in animals with IOA. Differences of IOA with spontaneous chondropathy were revealed. Spontaneous aging was characterized by calcium and phosphorus storage in deep and calcified zones and compensatory increase of sulfated glycosaminoglycans in intermediate and deep cartilage zones as evidenced by the metachromatic reaction and microanalysis data. Unlike spontaneous chondropathy connected with aging in experimentally stimulated osteoarthrosis more intensive storage of calcium but minor phosphorus in intermediate zone were marked. In IOA the calcified cartilage thinning and osteoclastic resorption are apparent with few changes of elements composition; the only difference from control is minority phosphorus *** The obtained results demonstrate specific tricks of X-ray electron probe microanalysis and its possibility in the research of mechanisms of articular cartilage alterations in osteoarthrosis.

Focus on intracytoplasmic morphologically selected sperm injection （IMSI）. a mini-review

Intracytoplasmic sperm injection （ICSI） is the recommended treatment in many cases of male-factor infertility. Several studies have demonstrated a positive correlation between optimal sperm morphology and positive ICSI outcomes. In fact, spermatozoa with severe abnormalities of the head are well documented to be associated with low fertilisation, implantation and pregnancy rates. However, a spermatozoon which is classified as ＇normal＇ by microscopic observation at low magnification could contain ultrastructural defects that impair both the fertilisation process and embryonic development. The intracytoplasmic morphologically selected sperm injection （IMSI） procedure changed the perception of how a spermatozoon suitable for injection should appear. Sperm selection is carried out at x 6000 magnification, allowing improved assessment of the sperm nucleus. Currently, standardized clinical indications for I MSI are lacking and the candidates are selected on the grounds of their medical history or of a careful analysis of the sperm suspension. Further prospective randomized studies are needed to confirm the advantages of IMSI in specific groups of patients. In addition to providing a brief overview of the IMSI procedure, this study aims to review the literature, which explains the theoretical basis and the clinical outcomes of this technique. Several reports show that IMSI is associated with improved implantation and clinical pregnancy rates as well as lower abortion rates when compared to ICSI. Although a possible correlation between the sperm＇s abnormal nucleus shape, increased DNA fragmentation and negative laboratory and clinical outcomes has been long investigated, the results are conflicting.

Benefits of Ilizarov automated bone distraction for nerves and articular cartilage in experimental leg lengthening

AIM To determine peculiarities of tissue responses to manual and automated Ilizarov bone distraction in nerves and articular *** Twenty-nine dogs were divided in two experimental groups: Group M-leg lengthening with manual distraction(1 mm/d in 4 steps), Group A-automated distraction(1 mm/d in 60 steps) and intact group. Animals were euthanized at the end of distraction, at 30 th day of fixation in apparatus and 30 d after the fixator removal. M-responses in gastrocnemius and tibialis anterior muscles were recorded, numerical histology of peronealand tibialis nerves and knee cartilage semi-thin sections, scanning electron microscopy and X-ray electron probe microanalysis were *** Better restoration of M-response amplitudes in leg muscles was noted in A-group. Fibrosis of epineurium with adipocytes loss in peroneal nerve, subperineurial edema and fibrosis of endoneurium in some fascicles of both nerves were noted only in M-group, shares of nerve fibers with atrophic and degenerative changes were bigger in M-group than in A-group. At the end of experiment morphometric parameters of nerve fibers in peroneal nerve were comparable with intact nerve only in A-group. Quantitative parameters of articular cartilage(thickness, volumetric densities of chondrocytes, percentages of isogenic clusters and empty cellular lacunas, contents of sulfur and calcium) were badly changed in M-group and less changed in *** Automated Ilizarov distraction is more safe method of orthopedic leg lengthening than manual distraction in points of nervous fibers survival and articular cartilage arthrotic changes.

Desmodus rotundus and Diphylla ecaudata, both of which are mammals of the order Chiroptera, Desmodontidae family, their diet consisting exclusively of blood. D. rotundus is the main vector and transmitter of the rabies virus, which affects human beings as well as several livestock species so the study of this bat species is of high importance within the fields of animal agriculture and public health. The present study describes and compares the histologic characteristics of the urinary system of two hematophagous bat species. A total of 5 bats from each species were captured in the municipalities of Progreso de Obregón, Hidalgo (D. rotundus), and Huayacocotla, Veracruz (D. ecaudata). Organs belonging to the urinary system were extracted: kidneys, ureters, urinary bladder, and urethra;samples were fixed using 10% formalin and processed by the paraffin embedding technique, obtaining sections of 5 µm thickness, which in turn were stained using hematoxylin-eosin (H-E) and Gomori trichrome (GT) stains. From the obtained histologic preparations, a descriptive and comparative analysis of the structural organography of the urinary system of both species was made, and no noteworthy histological differences between samples were noted. The present research is intended to provide a framework for future studies of these species’ currently understudied microscopic anatomy.
