Binary asymmetric nanocrystals （BNCs）, composed of a photoactive TiO2 nanorod joined with a superparamagnetic γ-Fe203 spherical domain, were embedded in polyethylene glycol modified phospholipid micelle and successfully bioconjugated to a suitably designed peptide containing an RGD motif. BNCs represent a relevant multifunctional nanomaterial, owing to the coexistence of two distinct domains in one particle, characterized by high photoactivity and magnetic properties, that is particularly suited for use as a phototherapy and hyperthermia agent as well as a magnetic probe in biological imaging. We selected the RGD motif in order to target integrin expressed on activated endothelial cells and several types of cancer cells. The prepared RGD-peptide/BNC conjugates, comprehensively monitored by using complementary optical and structural techniques, demon- strated a high stability and uniform dispersibility in biological media. The cytotoxicity of the RGD-peptide/BNC conjugates was studied in vitro. The cellular uptake of RGD-peptide conjugates in the cells, assessed by means of two distinct approaches, namely confocal microscopy analysis and emission spectroscopy determination in cell lysates, displayed selectivity of the RGD-peptide-BNC conjugate for the cw]33 integrin. These RGD-peptide-BNC conjugates have a high potential for theranostic treatment of cancer.

Confocal-based fluorescence fluctuation spectroscopy with a SPAD array detector

The combination of confocal laser-scanning microscopy(CLSM)and fluorescence fluctuation spectroscopy(FFS)is a powerful tool in studying fast,sub-resolution biomolecular processes in living cells.A detector array can further enhance CLSM-based FFS techniques,as it allows the simultaneous acquisition of several samples-essentially images-of the CLSM detection volume.However,the detector arrays that have previously been proposed for this purpose require tedious data corrections and preclude the combination of FFS with single-photon techniques,such as fluorescence lifetime imaging.Here,we solve these limitations by integrating a novel singie-photon-avalanche-diode(SPAD)array detector in a CLSM system.We validate this new implementation on a series of FFS analyses:spot-variation fluorescence correlation spectroscopy,pair-correlation function analysis,and image-derived mean squared displacement analysis.We predict that the unique combination of spatial and temporal information provided by our detector will make the proposed architecture the method of choice for CLSM-based FFS.

Resin foraging dynamics in Varroa destructor-infested hives: a case of medication of kin?

t Social insects have evolved colony behavioral, physiological, and organiza. tional adaptations (social immunity) to reduce the risks of parasitization and/or disease transmission. The collection of resin from various plants and its use in the hive as propolis is a clear example of behavioral defense. For Apis mellifera, an increased propolis content in the hive may correspond to variations in the microbial load of the colony and to a downregulation of an individual bee's immune response. However, many aspects of such antimicrobial mechanism still need to be clarified. Assuming that bacterial and fungal infection mechanisms differ from the action of a parasite, we studied the resin collection dynamics in Varroa destructor-infested honeybee colonies. Comparative experiments involving hives with different mite infestation levels were conducted in order to assess the amount of resin collected and propolis quality within the hive, over a 2-year period (2014 and 2015). Our study demonstrates that when A. mellifera colonies are under stress because of Varroa infestation, an increase in the number of resin foragers is recorded, even if a general intensification of the foraging activity is not observed. A reduction in the total polyphenolic content in propolis produced in infested versus uninfested hives was also noticed. Considering that different propolis types show varying levels of inhibition against a variety of honey bee pathogens in vitro, it would be very important to study the effects against Varroa of two diverse types of propolis: from Varroa-free and from Varroa-infested hives.

Overview of Gait Synthesis for the Humanoid COMAN

This paper focuses on the developments of a generic gait synthesis for the humanoid robot COMAN. Relying on the essential Gait Pattern Generator （GPG）, the proposed synthesis offers enhanced versatilities for the locomotion under different purposes, and also provides the data storage and communication mechanisms among different modules. As an outcome, we are able to augment new abilities for COMAN by integrating new control modules and software tools at a cost of very few modifications. Moreover, foot placement optimization is introduced to the GPG to optimize the gait parameter references in order to meet the robot＇s natural dynamics and kinematics, which enhances the synthesis＇s robustness while it＇s being implemented on real robots. We have also presented a practical approach to generate pelvis motion from CoM references using a simplified three-point-mass model, as well as a straightforward but effective idea for the state estimation using the sensory feedback. Three physical experiments were studied in an increasing complexity to demonstrate the effectiveness and successful implementation of the proposed gait synthesis on a real humanoid system.

3D （Three-dimensional） Caco-2 Spheroids： Optimized in vitro Protocols to Favor Their differentiation Process and to Analyze Their Cell Growth Behavior

3D (Three-dimensional) Caco-2 spheroids closely recapitulating in vivo physiological organization of intestinal epithelial cells, provide an excellent in vitro model system to study their pathophysiology and their response to stressful stimuli. The objective of this technical note is to provide optimized in vitro experimental protocols for culturing 3D Caco-2 spheroids and for analyzing their cell growth features. An optimized 3D Caco-2 spheroid culturing technique based on a new configuration of the culture medium is provided. A methodological approach to determine the distribution of the cell cycle phases in disaggregated Caco-2 spheroids by using cytofluorimetric analysis is also described. The optimized culturing protocol favors 3D Caco-2 spheroid differentiation process, as evaluated by the number of well-differentiated spheroids with a single hollow lumen. The cytofluorimetric analysis allows rapid collection of cell cycle phase data from high numbers of spheroid samples, thus, permitting to estimate their growth dynamics in a relatively short time. The optimized technical approaches described here can be applied in systematic manner to a variety of research activities utilizing 3D Caco-2 spheroids. Ease of use, time and economic saving advantages deriving from these protocols further highlight their potential.

There are essentially six sources for obsidians in the Mediterranean Basin: Mount Arci in Sardinia, the islands of Lipari and Pantelleria in Sicily and Palmarola in Latium, as well as the islands of Melos and Gyali in the south-eastern part of Greece. Identification and quantification of principal and trace elements which compose this volcanic glass allow source tracking. The present study proposes a investigation methodology, which was developed on a sample set composed by archaeological obsidian findings from Neolithic sites in Latium; three different analytical techniques are compared: SEM-EDXS, ED-XRF and LA-ICP-MS, in order to gather a wide data set to be compared with data in literature and to be processed by Principal Component Analysis (PCA) for assigning a provenance to each find.

This work concerns the Middle Lias Ammonites of Brescia province depo-sited in the paleontological Collection of Civic Museum of Natural History(Brescia).Thefossils,collected by museum personnel or by private collectors come from Medolo andCorso Formations.47 species have been identified.They belong to the following families:Phyllocerati-dae,Juraphyllitidae,Lytoceratidae,Nannolytoceratidae,Dactylioceratidae and Hildocerati-dae.The,biostratigrafic distribution of the examined species ranges from the base of theCarixian to the lower Toarcian.
