Switch of phosphorylation to O-GlcNAcylation of AhR contributes to vascular oxidative stress induced by benzo[a]pyrene

Benzo[a]pyrene(B[a]P)is a food contaminant toxic for cardiovascular *** nuclear translocation of Arylhydrocarbon receptor(AhR)plays an important role in B[a]P-induced oxidative stress and vascular *** confi rmed that B[a]P promoted ROS production in vascular smooth muscle cells(VSMCs)in vitro and in vivo,associated with the nuclear translocation of *** is known that phosphorylation inhibits while dephosphorylation of AhR promotes nuclear translocation of ***,from the posttranslational modifi cation level,the mechanism by which B[a]P activates and regulates the nuclear translocation of AhR is ***-immunoprecipitation results showed that cytoplasmic AhR was phosphorylated before B[a]P stimulation,and switched to O-GlcNAcylation upon B[a]P 1-h stimulation in VSMCs,suggesting there may be a competitively inhibitory relationship between O-GlcNAcylation and phosphorylation of ***,siRNAs of O-linked N-acetylglucosamine transferase(OGT),O-GlcNAcase(OGA)and OGA inhibitor PUGNAc were *** blocks but siOGA and PUGNAc promote B[a]P-dependent AhR nuclear translocation and oxidative ***11 may be the competitive binding site for phosphorylation and O-GlcNAcylation of ***-mimic variant inhibits but O-GlcNAcylation of AhR promotes AhR nuclear translocation and oxidative *** fi ndings highlight a new perspective for AhR nuclear translocation regulated by the competitive modifi cation between phosphorylation and O-GlcNAcylation.

Brassinosteroids regulate outer ovule integument growth in part via the control of INNER NO OUTER by BRASSINOZOLE‐RESISTANT family transcription factors

Brassinosteroids(BRs)play important roles in regulating plant reproductive *** signaling or BR biosynthesis null mutants do not produce seeds under natural conditions,but the molecular mechanism underlying this infertility is poorly *** this study,we report that outer integument growth and embryo sac development were impaired in the ovules of the Arabidopsis thaliana BR receptor null mutant *** expression and RNA-seq analyses showed that the expression of INNER NO OUTER(INO),an essential regulator of outer integument growth,was significantly reduced in the bri1-116 *** INO expression due to overexpression or increased transcriptional activity of BRASSINAZOLE-RESISTANT 1(BZR1)in the mutant alleviated the outer integument growth defect in bri1-116 ovules,suggesting that BRs regulate outer integument growth partially via BZR1-mediated transcriptional regulation of ***,INO expression in bzr-h,a null mutant for all BZR1 family genes,was barely detectable;and the outer integument of bzr-h ovules had much more severe growth defects than those of the bri1-116 ***,our findings establish a new role for BRs in regulating ovule development and suggest that BZR1 family transcription factors might regulate outer integument growth through both BRI1-dependent and BRI1-independent pathways.

Autophagy inhibits PDGF-BB-induced calcification in vascular smooth muscle cells

AIM: To investigate the relationship between autophagy and calcification in vascular smooth muscle cells( VSMCs) after plateletderived growth factor( PDGF)-BB stimulation. METHODS: Cultured VSMCs were stimulated with PDGF-BB for different time,the expression of vascular calcification-related proteins and autophagy-related proteins were detected by Western blot. The interaction between Beclin1 and PI3KC3 was detected by co-immunoprecipitation. RESULTS: The expression of BMP2 and ALP showed a trend from decline to rise. ALP slumped at 12 h,and BMP2 slumped at 6 h. Moreover,the expression of Beclin-1 showed a trend from rise to decline,and peaked at 12 h. The conversion of LC3-Ⅰto Ⅱ increased in a time-dependent manner,and peaked at 24 h. The expression of BMP2 and ALP was increased in VSMCs incubated with PDGF-BB and autophagy inhibitor 3-MA,compared with PDGFBB-stimulated VSMCs. Furthermore,the interaction between Beclin1 and PI3KC3 was enhanced at 6 h after PDGF-BB stimulated,peaked at 12 h,and kept in high level at 24 h. Moreover,the phosphorylation level of Beclin1 was enhanced by PDGF-BB stimulation,and peaked at 6 h. CONCLUSION: Our findings demonstrate that PDGF-BB-induced autophagy inhibits VSMC calcification by enhancing Beclin1 phosphorylation and interaction between Beclin1 and PI3KC3.

Breast cancer ranks second in the list of most common cancers among women and brings the double burden of economy and health to women. Therefore, it is an urgent and necessary task to study the pathogenic mechanism and the treatment of breast cancer. Glycoprotein hormone is a kind of hormones to promote the growth and the development of cell and stanniocalcin 2(STC2) is one of *** has shown us a various expression of SCT2 in organs and tissues and it can regulate many different pathological and physiological processes. In addition, there are a lot of previous studies that indicated a close correlation between STC2 and the development and metastasis of many cancers, which infers STC2 can serve as biomarker of certain cancers. Until now, the effects of STC2 on breast cancer have been studied widely, but research findings demonstrated two different views, one view is that STC2 plays an oncogenic role and the other is the opposite. In this paper, it will summarize and evaluate the research data and results about mammalian STC2 on breast cancer.

Offspring Sex Is Not Determined by Gestation Temperature in a Viviparous Lizard(Eremias multiocellata) from the Desert Steppe of Inner Mongolia

Sex-determining systems show a striking diversity not only among species, but also among popula tions. In reptiles, sex-determina tion is a continuum, from tempera ture-dependent sex determination(TSD) to genetic sex determination(GSD).The multi-ocellated racerunner(Eremias multiocellata)is reported to be a cryptic ZZ/ZW chromosomal TSD species, with male-biased sex ratios at high temperatures in two Gansu populations. However, the generality of the sex-determining pattern in different populations of this species remains unclear. To investigate the mode of sex determina tion in a popula tion of *** from the desert steppe of Inner Mongolia,we first identified sex chromosomes via comparative genomic hybridization(CGH). We then conducted a thermal manipulation experiment to determine the effect of gestation temperature on offspring sex *** the CGH studies we found that lizards from the Inner Mongolia population possessed ZZ/ZW sex chromosomes. However, our thermal manipulation experiment showed that gestation temperature did not affect the sex ratio of neonates in this population. In combination, these results rule out TSD in the Inner Mongolia population of E. multiocellata, and suggest that there is widespread geographic variation in the sexdetermining system of this species.

DICER1 regulates antibacterial function of epididymis by modulating transcription of p-defensins

DICER1 is a key enzyme responsible for the maturation of microRNAs. Recent evidences suggested that DICER1 and microRNAs expressed in epididymis were involved in the control of male fertility. However, the exact mechanism remains to be elucidated. Here, we created a mouse line by targeted disruption of Dicerl gene in the principal cells of distal caput epididymis. Our data indicated that a set of β-defensin genes were downregulated by DICER1 rather than by microRNAs. Moreover, DICER1 was significantly enriched in the promoter of β-defensin gene and controlled transcription. Besides, the antibacterial ability of the adult epididymis significantly declined upon Dicerl deletion both in vitro and in vivo. And a higher incidence of reproductive defect was observed in middle-aged Dicerl^-|- males. These results suggest that DICER1 plays an important role in transcription of β-defensin genes, which are associated with the natural antibacterial properties in a microRNA-independent manner, and further impacts the male fertility.

Effects of Er^(3+) on the proliferation,differentiation and mineralization function of primary mouse osteoblasts in vitro

A series of experimental methods including 3-（4,5-dimethyl-2-thiazolyl）-2,5-diphenyl-2H tetrazolium bromide （MTT） test,alkaline phosphatase （ALP） activity measurement,oil red O stain and measurement,mineralized function and quantitive real time RT-PCR （qRT-PCR） were employed to assess the effects of Er3＋ on the proliferation,differentiation and mineralization function of primary osteoblasts （OBs） in vitro at cell and molecular levels. The results indicated that Er3＋ inhibited the proliferation of OBs at a concentration of 1×10–7 mol/L,but had no effect at other concentrations. Er3＋ inhibited the differentiation of OBs at concentrations of 1×10–8,1×10–7,and 1×10–6 mol/L,but had no effect at a higher concentration of 1×10–5 mol/L. Er3＋ had no effect on the transdifferentiation of OBs at tested concentrations. Er3＋ inhibited the mineralization function of OBs at concentrations of 1×10–7,1×10–6,and 1×10–5 mol/L,but had no effect at a lower concentration of 1×10–8 mol/L. The expression of the mRNA for runt-related transcription factor 2 （RUNX-2） and peroxisome proliferators activated receptor γ （PPAR-γ） was down-regulated in the presence of 1×10–6 mol/L Er3＋. These findings suggested that Er3＋ might have negative effect on bone metabolism.

COR27 and COR28 encode nighttime repressors integrating Arabidopsis circadian clock and cold response

Summary It was noted that circadian components function in plant adaptation to diurnal temperature cycles and freezing tolerance. Our genome-wide transcriptome analysis revealed that evening-phased COR27 and COR28 mainly repress the transcription of clockassociated evening genes PRRS, ELF4 and cold-responsive genes. Chromatin immunoprecipitation indicated that CCAI is recruited to the site containing EE elements of COR27 and COR28 promoters in a temperaturedependent way. Further genetic analysis shows COR28 is essential for the circadian function of PRR9 and PRRT. Together, our results support a role of COR27 and COR28 as nighttime repressors integrating circadian clock and plant cold stress responses.

In this paper, the role of the plasma membrane (PM) H+-ATPase in extracellular calmodulin (CaM)-promoted pollen germination and in tube growth of Arabidopsis thaliana was investigated. Pollen germination, pollen tube growth rate, free cytosolic Ca2+ concentration ([Ca2+]cyt) and Ca2+ channel activity in the PM of pollen cells were measured. In response to fusicoccin or CaM treatment, in vitro pollen germination and pollen tube growth rate, [Ca2+]cyt and activity of a hyperpolarization-activated Ca2+-permeable channel increased. Sodium vanadate inhibited the promotion of these parameters by extracellular CaM. The results suggest that H+-ATPase may be involved in extracellular CaM-regulated pollen germination and in tube growth by modulation of the hyperpolarization-activated Ca2+ channel in the PM of A. thaliana pollen cells.
