A rapid approach for isolating a single fungal spore from rice blast diseased leaves

Single spore isolation is a fundamental approach in plant pathology and mycology to isolate and identify plant fungal pathogens from diseased samples.However,routine single spore isolation procedure is time-consuming and has a high risk of contamination by other microorganisms.In this study,we developed a rapid approach for isolating a single spore of the fungal pathogen,Pyricularia oryzae,from rice blast diseased leaves in the paddy field with low potential of contamination.First,rice blast leaves with single lesions were selected in the paddy field,and a single lesion was cut out and pressed and dragged gently across the surface of water agar.Next,a germinated single spore with a barely visible piece of agar was cut out of water agar with a dissecting needle under a stereomicroscope at approximately 120-fold magnification.Last,the germinated single spore with water agar was transferred onto oatmeal tomato agar for growth and preservation.Based on our experience,a skilled technician or student can successfully isolate single spore from over 150 independent diseased samples with nearly no contaminations in a single working day.This approach is also suitable for isolating single spore from other fungal disease samples that produce abundant aerial conidia.

In order to study the effect of post simulation in Equipment Maintenance teaching, in recent teaching, we made some try. Actual situations of teaching, analog troops and equipment maintenance, under the guidance of a teacher, students group and allow them to deal with failure, for failure analysis equipment maintenance technician role, troubleshooting. Practice has proved that the post simulation teaching methods helping to train the students’ interest in learning and the spirit of innovation, and improve the ability of students to solve practical problems, and to lay the foundation for future work.

A novel Bacterial Foraging Algorithm (BFA) based neural network is presented for image compression. To improve the quality of the decompressed images, the concepts of reproduction, elimination and dispersal in BFA are firstly introduced into neural network in the proposed algorithm. Extensive experiments are conducted on standard testing images and the results show that the pro- posed method can improve the quality of the reconstructed images significantly.

As an intensive and efficient way of transportation,drop and pull transport has become the mainstream transportation organization of foreign developed countries and regions.Based on the freight demand vector and aging of a given initial state,this paper established a goal planning and scheduling model based on the maximization of time window revenue to get the most efficient deployment plan under the condition of high customer satisfaction.The genetic algorithm based on task urgency was used to solve this model to obtain the optimal allocation scheme of total income.Finally,the number of algorithm iterations was analyzed,which proved the effectiveness of the algorithm performance.Through the sensitivity analysis of the number of tractors,this paper gave suggestions for more efficient tractor configuration.

近年来,经自然腔道取标本手术(NOSES)治疗结直肠癌已引起了广泛关注。NOSES应用于结直肠手术的潜在优势包括减轻术后疼痛,减少伤口并发症,减少对术后镇痛的需求,加快肠道功能的恢复,缩短住院时间,更好的美容效果,以及给患者带来了更佳的心理状态。尽管NOSES的手术创伤显著减轻,然而该技术也存在一些潜在问题。尤其随着这项新技术的兴起,人们对其无菌的把控、肿瘤学结果和病例选择等问题表示出关切。因此,迫切需要一部操作指南来规范NOSES手术在结直肠癌中的开展。经过国际NOSES联盟全体成员的三轮讨论,最终形成了该部共识。这一共识有助于NOSES在全球范围内的长远发展。

Pseudopteroxazole(1) and ileabethoxazole(2) are two marine benzoxazole alkaloids isolated from the Caribbean sea whip Pseudopterogorgia elisabethae by Rodriguez and co-workers. Notably, both 1 and 2 show significant antimicrobial activity against M. tuberculosis. The first total synthesis and structural revision of 1 were achieved by Corey and coworkers. Harmata and co-workers reported a synthetic route to 1 through benzothiazine chemistry.

To date there is virtually no information available concerning the fungi associated with Tectona grandis(teak)(Lamiaceae)in Thailand.In this study,samples of microfungi were collected from both asymptomatic stems and dead wood,and symptomatic branches,stem and leaves of T.grandisfrom 27 sites in six provinces(Chiang Mai,Chiang Rai,Phayao,Phitsanulok,Phrae and Uttaradit Provinces).Morphology and combined multi-gene phylogeny(CAL,GAPDH,ITS,LSU,RPB2,SSU,TEF1 and TUB)were used to identify taxa.A total of 270 collections,representing 28 fungal species residing in 12 families,7 orders and 21 genera,with three species of uncertain taxonomic placement were identified.Of these,one family,three genera and 14 species are new to science.The new family,Pseudocoleodictyosporaceae is introduced based on its distinct lineage in the Dothideomycetes and its unique morphology as compared to Roussoellaceae and Torulaceae.The new genera are Neooccultibambusa,Pseudocoleodictyospora and Subglobosporium.The newly described species are Diaporthe neoraonikayaporum,D.tectonendophytica,D.tectonae,D.tectonigena,Hermatomyces tectonae,H.thailandica,Manoharachariella tectonae,Neooccultibambusa chiangraiensis,Pseudocoleodictyospora sukhothaiensis,Ps.tectonae,Ps.thailandica,Rhytidhysteron tectonae,Subglobosporium tectonae and Tubeufia tectonae.Fourteen species are known published taxa including Alternaria tillandsiae,Berkleasmium talaumae,Boerlagiomyces macrospora,Ceratocladium purpureogriseum,Fusarium solani,Helicoma siamense,Lasiodiplodia theobromae,Macrovalsaria megalospora,Paradictyoarthrinium diffractum,Phaeoacremonium italicum,Sphaeropsis eucalypticola,Stachybotrys levispora,St.renispora and Thaxteriellopsis lignicola.Epitypifications or reference specimens are designated for Boerlagiomyces macrospora and Macrovalsaria megalospora.Macrovalsaria megalospora is transferred from Botryosphaeriaceae to Dothideomycetes genus,incertae sedis based on taxonomy and phylogenetic analysis,which indicate it is distinct from Botryosphaeriaceae.All fungal
