Hemorrhagic activity and mechanism of FIIa,a fibrinolytic enzyme from Agkistrodon acutus venom

AIM: To study the local hemorrhagic activity of a fibrinolytic enzyme (FIIa) from Agkistrodon acutus venom and its mechanism. METHODS: The local hemorrhagic activity was determined by subcutaneous injection on the back of mouse. The effects of FIIa on factor X, prothrombin, gelatin, and collagen were visualized by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Platelet aggregation assays were performed in rat platelet- rich plasma (PRP). Human umbilical vein endothelial cells (HUVEC) were cultured and passaged in complete M199 medium. Cell viability and nuclear morphology change were determined by fluorescein diacetate (FDA) staining and Hoechst 33258 staining, respectively. RESULTS: The minimum hemorrhagic dose (MHD) of FIIa was 89 μg. In vitro, FIIa (0.25 g/L) degraded factor X, prothrombin, collagen, and gelatin, and dose-dependently (0.25, 0.50, 0.75, and 1.00 g/L) inhibited the platelet aggregation induced by ADP in rat PRP. When HUVEC in culture treated with FIIa, HUVEC showed detachment in a dose-dependent manner, but no apoptosis sign was observed. CONCLUSION: FIIahad local hemorrhagic activity, and the mechanism was related to the degradation of factor X, prothrombin, gelatin, and collagen, the inhibition of ADP-induced platelet aggregation, and inducement of HUVEC detachment.2Now in Beijing Universityof Chinese Medicine,Beijing 100029,China.3Correspondence to Dr Xiu-xia LIANG.

Efficiency of adenoviral vector mediated CTLA4Ig gene delivery into mesenchymal stem cells

Objective To prevent Graft-versus-host disease (GVHD) in rat model, we evaluated the feasibility of mesenchymal stem cells (MSCs) as a gene transfer target and studied the efficiency of recombinant adenovirus mediated gene therapy.Methods We constructed the recombinant adenovirus containing CTLA4lg gene. Rat MSCs of passages 3-5 were infected by the adenovirus, and the transfection efficiency was monitored by GFP markers. We performed flow cytometric analysis, immunohistochemical and Western blotting analysis to identify the CTLA4lg expression. The gene transferred MSCs were tested for their ability to inhibit the allogeneic lymphocyte response in vitro and to prevent GVHD in a rat model.Results Recombinant adenovirus pAd-CTLA4lg was correctly constructed and confirmed. After MSCs were infected by the adenovirus, the CTLA4lg protein was detected not only in transgenic MSCs, but also in the culture medium. In a mixed lymphocytes response (MLR) test, the transgenic MSCs could significantly inhibit the allogeneic lymphocyte response compared with the control groups (P < 0. 05) . A model of GVHD was developed by transplanting bone marrow cells and spleen lymphocytes of F344 rats to lethally irradiated SD rats. The onset of GVHD could be ameliorated or prevented by co-administration of transgenic MSCs. All the rats in the control groups suffered severe acute GVHD. CTLA4lg expression was observed in the liver, intestine, kidney and spleen 30 days post- transplantation.Conclusions Our results indicate that adenoviral vectors could efficiently transfer CTLA4lg gene into MSCs and sustain long-term stable expression in vitro and in vivo.

A New Processing Method for the Nonlinear Signals Produced by Electromagnetic Flowmeters in Conditions of Pipe Partial Filling

Aim:To study the enzymological characterization of a fibrinolytic enzyme(FⅡa)from Agkistrodon acutus ***:The fibrinogenolytic effect and theinfluences of several protease inhibitors,chelating agents,and metal ions onfibrinogenolytic activity were visualized by sodium dodecyl sulfate-polyacryla-mide gel *** metal content of FⅡawas determined by atomicabsorption ***:After incubation with FⅡa(0.25 g/L),Aα-,Bβ-and γ-chains of fibrinogen disappeared within 5 min,30 min,and 8 h,*** molecular weights of major degradation products were 45 000 and 41 000,which were different from those bands produced by *** fibrinogenolyticactivity of FⅡawas strongly inhibited by ethylenediamine tetraacetic acid(EDTA),ethyleneglycol tetraacetic acid(EGTA),dithiothreitol and cysteine,but not byphenyhnethyl-sulfonyl fluoride and soybean trypsin ***(3171±25rag&g),potassium(489±17 mg/kg)and calcium(319±13 mg/kg)were found in FⅡ***2+,Ca2+and Mg2+could recover the fibrinogenolytic activity of FⅡa,which wasinhibited by *** Ca2+ could recover the fibrinogenolytic activity inhibitedby ***:FⅡacan degrade the Aα-,Bβ-and γ-chains of fibrinogen.FⅡais a metalloproteinase,and Zn2+,Ca2+,and disulfide bonds are necessary forits fibrinogenolytic activity.

Stability of TiO_(2) and Al_(2)O_(3) Nanofluids

Aiming at the dispersion stability of nanofluids,we investigate the absorbency and the zeta potential of TiO_(2) and Al2O3 nanofluids under different pH values and different dispersant *** results show that in the mass fraction 0.05%alumina and 0.01%titanium dioxide nanosuspensions,the absolute value of the zeta potential and the absorbency of the two nanofluids with sodium dodecyl sulfate(SDS)dispersant are the highest at an optimal pH(pHAl_(2)O_(3)≈6.0,pHTiO2≈9.5)and that there is a good correlation between absorbency and zeta potential:the higher the absolute value of the zeta potential is,the greater the absorbency is,and the better the stability of the system *** optimizing concentrations for SDS are 0.10%and 0.14%,respectively,at which the two nanofluids have the best dispersion *** calculated DLVO interparticle interaction potentials verify the experimental results of the pH effect on the stability behavior.

The crystal structure of the title compound with good acetylcholinesterase inhibitory activity （C21H19NO8, Mr = 413.37） has been determined by single-crystal X-ray diffraction analysis. The crystal is of monoclinic, space group P21 with a = 10.257（3）, b = 8.954（3）, c = 11.541（4） A°, β= 109.772（6）°, V= 997.5（6）A°^3, Dc= 1.376 g/cm^3, μ = 0.107 mm^-1, Z= 2, F（000） = 432, S = 1.041, the final R = 0.0356 and wR = 0.0884. In the molecular structure, the ring of N（1）-C（5）-C（6）-C（7）-C（8）-C（13） is of twisting-boat conformation, and the ring of N（1 ）-C（1 ）- C（2）-C（3）-C（4）-C（5） is nearly a plane.

Novel mutation in a Chinese patient with progressive familial intrahepatic cholestasis type 3

Genotyping is conclusive for the diagnosis of progressive familial intrahepatic cholestasis type 3(PFIC3). Here we report a Chinese patient of PFIC3 with compound mutations in the ABCB4 gene. Liver biopsy was performed on a 17-year-old male patient with intrahepatic cholestasis of unknown etiology. Liver histology findings are indicative of intrahepatic cholestasis with extensive fibrosis. Genotyping revealed c.175C>T(p.L59L) mutation in exon 4, c.504C>T(p.N168N) mutation in exon 6, c.711A>T(p.I237I) mutation in exon 8, c.874A>T(p.K292X) in exon 9 and a novel mutation, c.1804G>T(p.G602W) in exon 15. Based on these findings, the patient was diagnosed with PFIC3. The novel mutation p.G602 W in exon 15 was predicted as probably damaging by Poly Phen-2 with a score of 0.986(sensitivity: 0.54; specificity: 0.94) and was predicted to affect protein function with a SIFT score of 0.01.
