Identification and Metabolic Mechanism of Non-fermentative Short-cut Denitrifying Phosphorus-removing Bacteria

To investigate the characteristics and metabolic mechanism of short-cut denitrifying phosphorus-removing bacteria (SDPB) that are capable of enhanced biological phosphorus removal (EBPR) using nitrite as an electron acceptor, an aerobic/anoxic sequencing batch reactor was operated under three phases. An SDPB-strain YC was screened after the sludge enrichment and was identified by morphological, physiological, biochemical properties and 16S rDNA gene sequence analysis. Denitrifying phosphorus-removing experiments were conducted to study anaerobic and anoxic metabolic mechanisms by analyzing the changes of chemical oxygen demand (COD), phosphate, nitrite, poly-β-hydroxybutyrate (PHB), and glycogen. The results show that strain YC is a non-fermentative SDPB similar to Paracoccus denitrificans. As a kind of non-fermentative bacteria, the energy of strain YC was mainly generated from phosphorus release (96.2%) under anaerobic conditions with 0.32 mg P per mg synthesized PHB. Under anoxic conditions, strain YC accumulated 0.45 mg P per mg degraded PHB, which produced most of energy for phosphate accumulation (91.3%) and a little for glycogen synthesis (8.7%). This metabolic mechanism of strain YC is different from that of traditional phosphorus-accumulating organisms (PAOs). It is also found that PHB, a kind of intracellular polymer, plays a very important role in denitrifying and accumulating phosphorus by supplying sufficient energy for phosphorous accumulation and carbon sources for denitrification. Therefore, monitoring ΔP/ΔPHB and ΔNO2--N/ΔPHB is more necessary than monitoring ΔP/ΔCOD,ΔNO2--N/ΔCOD, or ΔP/ΔNO2--N.

Single-step genomic best linear unbiased prediction(ss GBLUP) is now intensively investigated and widely used in livestock breeding due to its beneficial feature of combining information from both genotyped and ungenotyped individuals in the single model. With the increasing accessibility of whole-genome sequence(WGS) data at the population level, more attention is being paid to the usage of WGS data in ss GBLUP. The predictive ability of ss GBLUP using WGS data might be improved by incorporating biological knowledge from public databases. Thus, we extended ss GBLUP, incorporated genomic annotation information into the model, and evaluated them using a yellow-feathered chicken population as the examples. The chicken population consisted of 1 338 birds with 23 traits, where imputed WGS data including 5 127 612 single nucleotide polymorphisms(SNPs) are available for 895 birds. Considering different combinations of annotation information and models, original ss GBLUP, haplotype-based ss GHBLUP, and four extended ss GBLUP incorporating genomic annotation models were evaluated. Based on the genomic annotation(GRCg6a) of chickens, 3 155 524 and 94 837 SNPs were mapped to genic and exonic regions, respectively. Extended ss GBLUP using genic/exonic SNPs outperformed other models with respect to predictive ability in 15 out of 23 traits, and their advantages ranged from 2.5 to 6.1% compared with original ss GBLUP. In addition, to further enhance the performance of genomic prediction with imputed WGS data, we investigated the genotyping strategies of reference population on ss GBLUP in the chicken population. Comparing two strategies of individual selection for genotyping in the reference population, the strategy of evenly selection by family(SBF) performed slightly better than random selection in most situations. Nanoscaled spin-dependent tunnelling lines were patterned on doped Si and studied for tunnelling from the SDT ferromagnetic layer through an insulating barrier into Si. The injection contacts have the form of long strips with width and separation, ranging from 100 nm to 2 μm, and are patterned using e-beam lithography. The measured I-V characteristics versus temperature （80 to 300K） on the 100 nm scaled devices between the layered-magnetic metals and the semiconductor clearly showed ballistic tunnelling, with weak dependence on the temperature. This is qualitatively different, at elevated temperatures, from 2-μm-wide scaled-up spin-dependent tunnelling structures, where thermal-ionic emission was observed to dominate carrier transport.

Several photorespiratory bypasses have been introduced into plants and shown to improve photosynthesis by increasing chloroplastic C02 concentrations or optimizing energy *** recently reported that an engineered GOC bypass could increase photosynthesis and productivity in ***,the grain yield of GOC plants was unstable,fluctuating in different cultivation seasons because of varying seed setting *** this study,we designed a synthetic photorespiratory shortcut(the GCGT bypass)consisting of genes en-coding Oryza sativa glycolate oxidase and Escherichia coli catalase,glyoxylate carboligase,and tartronic semialdehyde *** GCGT bypass was guided by an optimized chloroplast transit peptide that targeted rice chloroplasts and redirected 75% of carbon from glycolate metabolism to the Calvin cycle,identical to the native photorespiration *** transgenic plants exhibited significantly increased biomass production and grain yield,which were mainly attributed to enhanced photosynthesis due to increased chloroplastic C02 *** the increases in biomass production and grain yield,GCGT transgenic plants showed a reduced seed setting rate,a phenotype previously reported for the GOC *** transcriptomic,physiological,and biochemical assays revealed that photosynthetic car-bohydrates were not transported to grains in an efficient manner,thereby reducing the seed setting *** together,our results demonstrate that the GCGT photorespiratory shortcut confers higher yield by promoting photosynthesis in rice,mainly through increasing chloroplastic C02 concentrations.

Sedum alfredii Hance is a newly reported zinc （Zn） and cadmium （Cd） hyperaccumulator native to China. In this study, four populations of S. alfredii were collected from Yejiwei （YJW）, Jinchuantang （JCT） and Qiaokou （QK） lead （Pb）lZn mines located in Hunan Province as well as Quzhou （QZ） PblZn mine located in Zhejiang Province for exploring the intraspecies difference of this plant in metal accumulation. Although they grew in the PblZn spoils with relatively similar levels of Zn, Cd and Pb, remarkable differences among the four populations in tissue heavy metal concentrations were observed. The shoot Zn concentration of QZ population （11 116mglkg） was highest and nearly five times higher than that of the JCT population （1 930 mg/kg）. Furthermore, the shoot Cd concentration observed in the QZ population （1 090 mg/kg） was also highest and 144 times higher than that found in the JCT population （7.5 mg/kg）. As for Pb concentrations in the shoot of different populations, a fourfold difference between the highest and the lowest was also found. Such difference on metal accumulation was population-specific and may be significantly explained by differences in the soil properties such as pH, organic matter （OM）, and electrical conductivity （EC）. Taking biomass and metal concentration in plants into consideration, the QZ, YJW and QK populations may have high potential for Zn phytoremediation, the QZ population may have the highest potential in Cd phytoremediation, and the QK population may be the most useful in Pb phytoremediation.
