Cloning of genes encoding nonhost hypersensitive response-inducing elicitors from Phytophthora boehmeriae

We have devised a high-throughput functional cloning method to isolate cDNAs from Phytophthora boehmeriae of which the products elicit a hypersensitive response (HR) in tobacco. The cDNAs were cloned into a binary potato virus X (PVX)-based expression vector and transformed into Agrobacterium tumefeciens (Mog101). 4100 colonies were individually toothpick-inoculated onto leaflets of Nicotiana benthamiana. 12 cDNAs were identified whose expression induced formation of a necrotic lesion around the inoculation site. 7 of these clones have different sequences. One of these clones PBC43 encodes specific elicitin. Clone PBC163 encodes a protein highly homologous to Rab; PBC241 en-codes a prohibitin protein; PBN62 encodes a Heat Shock Protein 60 (HSP60). The other five cDNAs reveal no homology to known protein and are thus considered novel. These observations suggest that this functional screening method is a versatile strategy to identify cDNAs of pathogens that encode elicitors and other HR-inducing proteins.

Evaluation of the antioxidant activity of blueberry ethanol extracts under microwave extraction

Blueberry extract contains diversiform monomers of anthocyanins, whose antioxidant activity determines the functionality of anthocyanidins and the efficacy of extraction technology. The evaluation of the antioxidant activity of anthocyanidins of blueberry ethanol extracts (BEE) was investigated by considering the monomers' identification and thermal degradation in microwave-assisted extraction (MAE). Thirteen monomers of anthocyanins were identified from the extracts of blueberry under MAE, where the highest content was for cyanidin glycosides, and the lowest content was for the malvidin glycosides. The onset temperatures of degradation for anthocyanidins monomers in BEE were 41.07°C-48.50°C, and the glycoside contents of delphinidin, cyanidin, petunidin, peonidin, peonidin, and malvidin, in turn, decreased obviously. The degradation of anthocyanidins with extraction temperature improved, other than weakened, its antioxidant activities in BEE under MAE. The research results may provide valuable guidance for the antioxidant application of extracts from blueberries.

The major viral diseases that occur on rice plants in zhejiang Province, eastern China, are stripe and rice black-streaked dwarf diseases. Rice stripe disease is only caused by rice stripe tenuivirus （RSV）, while rice black-streaked dwarf disease can be caused by rice black-streaked dwarf fijivirus （RBSDV） and/or southem rice black-streaked dwarf fijivirus （SRBSDV）. Here we review the characterization of these viruses, methods for their detection, and extensive surveys showing their occurrence and spread in the province.

In this study,an untargeted metabolomics method based on LC-MS was used to investigate the differential metabolites of coix seed after fermentation by Monascus *** compounds with highly correlated variable importance in projection(VIP)values of orthogonal partial least squares discriminant analysis(OPLS-DA)were selected as the important metabolites of coix seed *** seed was fermented by *** for 9 *** color value units(CVU)of pigment in fermented coix seed reached 157.47 CVU/g,and the contents of total phenols and total flavonoids were 0.32 mg/g and 1.2 mg/g,respectively.A total of 270 differential metabolites were detected,including amino acids,peptides,fatty acids,phenols,steroids,and their derivatives,of which the relative contents of 187 metabolites showed a significant upward trend(p<0.05).Amino acids,peptides and analogues accounted for the largest proportion at 26%,among which most metabolites were *** example,N-alpha-acetyllysine,l-tryptophan and l-arginine exhibited a fold change(FC)of 6200.87,330.29,and 35.59,*** particular,the detected steroid metabolites accounted for 3%of the total metabolites,and they all participated in the same pathway,in which androsterone,2-methoxyestrone and etiocholanolone were *** synthesis of these steroids was related to the precursors and enzymes in the pathway,such as dehydroepiandrosterone,androstenedione,and 3beta-hydroxy-delta5-steroid dehydrogenase(HSD3B).The results of this study reveal *** fermentation can successfully transform coix seed,improving the functional activities of ***,this process has high development value with important applications in the food industry.

In recent years,the occurrence of rice Chilo suppressalis( Walker) in Longyou County of zhejiang Province has increased year by year and has become aggravated. This poses a great threat to the safety of rice production. In order to curb the recurrence momentum of rice C. suppressalis,according to the dynamic monitoring and field system survey of rice C. suppressalis in Longyou County since 2015,the causes of the annual increase of rice C. suppressalis in recent years were analyzed,and the recurrence area of C. suppressalis was introduced,and finally some prevention and control measures were put forward.

In this paper, the first generation and second generation of Chilo suppressalis Walker were conducted as the test objects to investigate control of a new pesticide(chlorantraniliprole) with preparation of chlorantraniliprole 20% SC,abamectin 1.8% EC and chlorpyrifos 40% EC to Chilo suppressalis Walker. The results showed that chlorantraniliprole 20% SC, abamectin 1.8% EC and chlorpyrifos40% EC had effective control to Chilo suppressalis Walker for its excellent control efficacy and long persistence period. Seedling protection efficacies were between84.2%-100%, 90.1% and 92.1%, and insecticidal efficacies were between 84.6%-100%,90.7% and 93.8% 36 days after application of chlorantraniliprole 20% SC in treatments with abamectin 1.8% EC and chlorpyrifos 40% EC for first generation Chilo suppressalis Walker control. The dead panicles were between 0.2%-1.4%, 0.8%and 0.7%, pest plant rate control in 0.4%-2.2%, 1.1% and 0.9%, and the rates of damaged rice plants were between 82.4%-97.6%, 90.2%-91.2% and 91.4%-92.8%,30 days after application of chlorantraniliprole 20% SC, abamectin 1.8% EC and chlorpyrifos 40% EC for the second generation Chilo suppressalis Walker control. It can effectively protect seedlings, protect spike, increase grain number and grain weight, which promoting the increase production income effect. The real estate increased by 9.7%-12.3%, 10.9% and 11.2% than water treatment(CK) with a very significant level, while the incomes were higher than 1 719.19-1 998.22, 1 956.52 and 2 057.83 yuan.

After systematic monitoring and field survey in orchard for three years from 2009 to 2011, it was found that Bactrocera dorsalis occurred for more than three generations each year on the north shore of the middle and lower reaches of Yangtze River, with overlapping generations; the activity peak of adults was from mid August to late September; the occurrence in citrus orchard in hilly and mountainous area was severe; the average insect-fruit rate in October reached 0.20% - 15.88% ; B. dorsalis had sporadic occurrence in other orchards. The integrated control measures of plowing in winter to kill pupae, removing insect-fruits, boycott- ing on the ground, trapping by sex attraetants and spraying pesticides could basically control the damage of B. dorsalis.

Occurrence and Epidemic Regularity of Wheat Stripe Rust in Nanchong City of Sichuan Province

In order to realize monitoring and early warning and comprehensive management of wheat stripe rust and to reduce its occurrence in Nanchong City, the occurrence and epidemic regularity of wheat stripe rust in Nanchong was studied by system monitoring and general survey, resistance identification, physiological race monitoring and meteorological data analysis. The initial occurrence location and spreading pathway of Puccinia striiformis f. sp. tritici （Pst） were first verified; there were two infection peaks of wheat stripe rust in Nanchong and one to three epidemic peaks in fields, in which the occurrence area of the first epidemic peak played a pivotal role in disease prevalence in that year; the cumulative occurrence area in late January was positively correlated with annual occurrence area, with the correlation coefficient of 0.769 ; the prediction model for infected field rate, diseased plant rate and annual occurrence area was established. The internal reason for heavy occurrence and prevalence of wheat stripe rust in Nanchong was the decline or loss of wheat resistance against stripe rust, as well as the appearance of physiological races of Pst, which later became dominant races. Large fluctuation of temperature in warm winter and spring and more fog and dew days were external reasons responsible for prevalence of stripe rust. From 2002 to 2014 ,the accuracy rate of short-term prediction of wheat stripe rust reached 100%, while that of me- dium-term and long-term prediction reached over 98% and 95%, respectively, 5% -15% higher than that of the years before 1998.
