Comparison of the effects of recombinant human endostatin and docetaxel on human umbilical vein endothelial cells in different growth states

Background Recombinant human endostatin （rh-endostatin, Endostar） has been proved to be an inhibitor of angiogenesis. Docetaxel has been also considered as a common chemotherapeutic agent with inhibition of angiogenesis of malignancies. However, their function has been seldom compared and a best synergism protocol is not determined. This study aimed to compare the effects of two drugs, investigate their combined impact on human umbilical vein endothelial cells （HUVECs）, a molecular basis and find ideal protocols to inhibit endothelial cell proliferation. Methods HUVECs on confluent growth or activated by vascular endothelial growth factor （VEGF） were treated by rh-endostatin or/and docetaxel at respective gradient concentration in following operations as cell proliferation determined by MTT assay, cell cycle distribution, apoptosis and markers of CD146, CD62E and CD105 detected by flow cytometery, the structure of the channel formed by HUVECs measured by tube formation count. Results Rh-endostatin exhibited time dependent inhibition of proliferation while docetaxel showed both time and dose dependent inhibition. HUVECs accumulated in G0-G1 with decreased numbers of cells in G2 after a single treatment of rh-endostatin or that followed by docetaxel treatment. Cells accumulated in G2 after both a single docetaxel and simultaneous administration. Both the number of cells in Go-G1 and apoptotic cells were increased by docetaxel followed by rh-endostatin treatment. The number of non-apoptotic cells at Go-G1 was increased by first administering rh-endostatin then docetaxet. Sequential treatment of docetaxel followed by rh-endostatin resulted in the greatest increase in apoptosis （34.7%） and the second highest apoptosis was seen with simultaneous administration （18.2%）. Expression of CD146 and CD105 on confluent HUVECs was reduced at certain doses of rh-endostatin and/or docetaxel. Effect of ginseng polysaccharides and dendritic cells on the balance of Th1/Th2 T helper cells in patients with non-small cell lung cancer

OBJECTIVE: To investigate the effect of thorascopic administration of ginseng polysaccharides(GPS)plus dendritic cells(DC) on T helper cell type 1/T helper cell type 2(Th1/Th2) balance in patients with non-small cell lung cancer(NSCLC).METHODS: A total of 96 NSCLC patients were divided evenly into two groups. The control group was treated with DCs alone and the treatment group was treated with DCs plus GPS. After DCs and GPS were administered thoracoscopically, once a week,4 times for 30 days, the patients' quality of life was measured with the Functional Assessment of Can-cer treatment-Lung(FACT-L) questionnaire before and after treatment. Serum interferon-γ(INF-γ), interleukin-4(IL-4), IL-2 and IL-5 were examined before and after ***: The level of Th1 cytokines(INF-γ, IL-2)and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) increased in both treatment groups, while Th2cytokines(IL-4, IL-5) and FACT-L scores decreased(Ptreatment Th1 cytokines(INF-γ, IL-2) and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) were higher in the DCs +GPS group than in the control group(Ptreatment regime of DCs plus GPS had a greater effect on NSCLC patients' immune function as compared with DCs alone. This was evident by increased expression of Th1 cytokines(INF-γ, IL-2) and the ratio of Th1/Th2(INF-γ/IL-4, IL-2/IL-5), as well as by decreased FACT-L scores and the expression of Th2 cytokines(IL-4,IL-5).

Immune checkpoint inhibitors targeting the programmed cell death-1(PD-1)protein significantly improve survival in patients with advanced non-small-cell lung cancer(NSCLC),but its impact on early-stage ground-glass opacity(GGO)lesions remains *** is a single-arm,phase II trial(NCT04026841)using Simon’s optimal two-stage design,of which 4 doses of sintilimab(200 mg per 3 weeks)were administrated in 36 enrolled multiple primary lung cancer(MPLC)patients with persistent high-risk(Lung-RADS category 4 or had progressed within 6 months)*** primary endpoint was objective response rate(ORR).T/B/NK-cell subpopulations,TCR-seq,cytokines,exosomal RNA,and multiplexed immunohistochemistry(mIHC)were monitored and compared between responders and ***,two intent-to-treat(ITT)lesions(pure-GGO or GGO-predominant)showed responses(ORR:5.6%,2/36),and no patients had progressive disease(PD).No grade 3-5 TRAEs *** total response rate considering two ITT lesions and three non-intent-to-treat(NITT)lesions(pure-solid or solid-predominant)was 13.9%(5/36).The proportion of CD8^(+)T cells,the ratio of CD8^(+)/CD4^(+),and the TCR clonality value were significantly higher in the peripheral blood of responders before treatment and decreased over ***,the mIHC analysis showed more CD8^(+)T cells infiltrated in ***,responders’cytokine concentrations of EGF and CTLA-4 increased during *** exosomal expression of fatty acid metabolism and oxidative phosphorylation gene signatures were down-regulated among ***,PD-1 inhibitor showed certain activity on high-risk pulmonary GGO lesions without safety *** effects were associated with specific T-cell re-distribution,EGF/CTLA-4 cytokine compensation,and regulation of metabolism pathways.

Dear Editor,In the first-line setting for advanced gastric cancer(AGC),a combination of nivolumab and fluoropyrimidine/platinum has become the standard treatment[1].However,treatment options are limited after failure of first-line therapy,particularly in ***,a monoclonal antibody against vascular endothelial growth factor receptor(VEGFR)-2,was associated with significant improvements in progression-free survival(PFS)when combined with paclitaxel in the second-line setting[2,3].Further,ramucirumab has not yet been approved by the National Medical ProductsAdministration;hence,it is still inaccessible in ***,single-agent paclitaxel is still a standard-of-care second-line treatment for AGC in China.

Objective: Patients with radioactive iodine-refractory differentiated thyroid cancer(RAIR-DTC) are often diagnosed with delay and constrained to limited treatment options. The correlation between RAI refractoriness and the underlying genetic characteristics has not been extensively ***: Adult patients with distant metastatic DTC were enrolled and assigned to undergo next-generation sequencing of a customized 26-gene panel(Thyro Lead). Patients were classified into RAIR-DTC or non-RAIR groups to determine the differences in clinicopathological and molecular characteristics. Molecular risk stratification(MRS) was constructed based on the association between molecular alterations identified and RAI refractoriness, and the results were classified as high, intermediate or low ***: A total of 220 patients with distant metastases were included, 63.2% of whom were identified as RAIRDTC. Genetic alterations were identified in 90% of all the patients, with BRAF(59.7% vs. 17.3%), TERT promoter(43.9% vs. 7.4%), and TP53 mutations(11.5% vs. 3.7%) being more prevalent in the RAIR-DTC group than in the non-RAIR group, except for RET fusions(15.8% vs. 39.5%), which had the opposite pattern. BRAF and TERT promoter are independent predictors of RAIR-DTC, accounting for 67.6% of patients with RAIR-DTC. MRS was strongly associated with RAI refractoriness(P<0.001), with an odds ratio(OR) of high to low MRS of 7.52 [95%confidence interval(95% CI), 3.96-14.28;P<0.001] and an OR of intermediate to low MRS of 3.20(95% CI,1.01-10.14;P=0.041).Conclusions: Molecular alterations were associated with RAI refractoriness, with BRAF and TERT promoter mutations being the predominant contributors, followed by TP53 and DICER1 mutations. MRS might serve as a valuable tool for both prognosticating clinical outcomes and directing precision-based therapeutic interventions.

Smart nanoparticles,which can respond to biological cues or be guided by them,are emerging as a promising drug delivery platform for precise cancer *** field of oncology,nanotechnology,and biomedicine has witnessed rapid progress,leading to innovative developments in smart nanoparticles for safer and more effective cancer *** this review,we will highlight recent advancements in smart nanoparticles,including polymeric nanoparticles,dendrimers,micelles,liposomes,protein nanoparticles,cell membrane nanoparticles,mesoporous silica nanoparticles,gold nanoparticles,iron oxide nanoparticles,quantum dots,carbon nanotubes,black phosphorus,MOF nanoparticles,and *** will focus on their classification,structures,synthesis,and intelligent *** smart nanoparticles possess the ability to respond to various external and internal stimuli,such as enzymes,pH,temperature,optics,and magnetism,making them intelligent ***,this review will explore the latest studies on tumor targeting by functionalizing the surfaces of smart nanoparticles with tumor-specific ligands like antibodies,peptides,transferrin,and folic *** will also summarize different types of drug delivery options,including small molecules,peptides,proteins,nucleic acids,and even living cells,for their potential use in cancer *** the potential of smart nanoparticles is promising,we will also acknowledge the challenges and clinical prospects associated with their ***,we will propose a blueprint that involves the use of artificial intelligence-powered nanoparticles in cancer treatment *** harnessing the potential of smart nanoparticles,this review aims to usher in a new era of precise and personalized cancer therapy,providing patients with individualized treatment options.

Lung cancer is the leading cause of cancer-related deaths *** is a common metastatic site of lung cancer,about 50%of bone metastatic patients will experience skeletal related events(SREs).SREs not only seriously impact the quality of life of patients,but also shorten their survival *** treatment of bone metastasis requires multi-disciplinary therapy(MDT)and development of individualized treatment *** order to standardize the diagnosis and treatment of bone metastasis in lung cancer,the expert group of the MDT Committee of the Chinese Medical Doctor Association has developed the expert consensus on the diagnosis and treatment of lung cancer bone metastasis.
