Antitumor effects of different Ganoderma lucidum spore powder in cell-and zebrafish-based bioassays

Objective:Ganoderma lucidum spore(GLS)is gaining reco.nition as a medicinal part of *** and has been reported to possess various pharmaco.ogical properties,such as antitumor *** this work,wall-broken GLS powder(BGLSP)and wall-removed GLS powder(RGLSP),two kinds of GLS powder with different manufacturing techniques,were co.pared in terms of co.tents of active co.stituents and in vivo and in vitro antitumor ***:The ultraviolet and visible spectrophotometry method was used to determine the co.tents of polysaccharides and total triterpenoids in BGLSP and *** individual triterpenoids were further quantified using ultra-high-performance liquid chromatography and quantitative analysis of multico.ponents by single *** antitumor effects of BGLSP and RGLSP were evaluated using in vitro cell viability assay against human gastric carcinoma SGC-7901,lung carcinoma A549 and lymphoma Ramos and further validated by in vivo zebrafish xenograft models with transplanted SGC-7901,A549 and Ramos,Results:The results showed that the co.tents of polysaccharides,total triterpenoids and individual triterpenoids of RGLSP were significantly higher than those of *** both BGLSP and RGLSP inhibited the three tumor cell lines in vitro in a dose-dependent manner,the inhibitory effects of RGLSP were much better than those of *** the in vivo zebra fish assay,RGLSP exhibited more potent inhibitory activities against tumors transpla nted into the zebra fish co.pared with BGLSP,and the inhibition rates of RGLSP reached approximately 78%,31%and 83%on SGC-7901,A549 and Ramos,***:The results indicated that the antitumor effects of GLS were positively co.related with the co.tents of the polysaccha rides and triterpenoids and demonstrated that the wall-removing manufacturing technique co.ld significantly improve the levels of active co.stituents,and thereby enhance the antitumor activity.

Traditionally,hyaluronic acid has been widely used for drug delivery,but the current application bottleneck is that hyaluronic acid is hydrophilic and electronegative,which makes it difficult to carry hydrophobic drugs and small interfering RNA(siRNA)with the same *** on previous studies,we designed and synthesized hyaluronic acid nanocarriers HA-spermine/N,N,N-trimethylcystamine/DOX-TPP(HSTD)for loading siRNA to overco.e the problem of siRNA release caused by strong electrostatic ***,N,N,Ntrimethylcystamine in the carrier can be degraded by intracellular glutathione to co.pletely and rapidly release siRNA,thus promoting ***,when co.elivered with the chemotherapy drug doxorubicin(DOX),this novel nanocarrier showed promising synergy in inhibiting tumor growth.

Salvia miltiorrhiza and *** are rich in diterpenoids and have therapeutic effects on cardiovascular *** this study,the spatial distribution of diterpenoids in both species was analyzed by a co.bination of metabolomics and mass spectrometry imaging *** results indicated that diterpenoids in *** were mainly abietane-type norditerpenoid quinones with a furan or dihydrofuran D-ring and were mainly distributed in the periderm of the roots,*** and tanshinone *** co.pounds in *** were mainly phenolic abietane-type tricyclic diterpenoids with six-or seven-membered C-rings,and were widely distributed in the periderm,phloem,and xylem of the roots,e.g.11-hydroxy-sugiol,11,20-dihydroxy-sugiol,and 11,*** addition,the leaves of *** were rich in tanshinone biosynthesis precursors,such as 11-hydroxy-sugiol,while those of *** were rich in phenolic *** in the upstream pathway of tanshinone biosynthesis were highly expressed in the root of ***,and genes in the downstream pathway were highly expressed in the root of ***,we describe the specific tissue distributions and mechanisms of diterpenoids in two Salvia species,which will facilitate further investigations of the biosynthesis of diterpenoids in plant synthetic biology.

Chromopeptide A, a highly cytotoxic depsipeptide from the marine sediment-derived bacterium Chromobacterium sp. HS-13-94

A bicyclic depsipeptide, chromopeptide A(1), was isolated from a deep-sea-derived bacterium Chromobacterium sp. HS-13-94. Its structure was determined by extensive spectrosco.ic analysis and by co.parison with a related known co.pound. The absolute co.figuration of chromopeptide A was established by X-ray diffraction analysis employing graphite monochromated Mo K_α radiation(λ ? 0.71073 ?) with small Flack parameter 0.03. Chromopeptide A suppressed the proliferation of HL-60, K-562, and Ramos cells with average IC_(50) values of 7.7, 7.0, and 16.5 nmol/L, respectively.

Global gene expression analysis in liver of db/db mice treated with catalpol

Catalpol,a major bioactive co.ponent from Rehmannia glutinosa,which has been used to treat *** present study was designed to elucidate the anti-diabetic effect and mechanism of action for catalpol in db/db *** db/db mice were randomly divided into six groups(10/group) acco.ding to their blood gluco.e levels:db/db co.trol,metformin(positive co.trol),and four dose levels of catalpol treatment(25,50,100,and 200 mg·kg^(-1)),and 10 db/m mice were used as the normal *** the groups were administered orally for 8 *** levels of fasting blood gluco.e(FBG),random blood gluco.e(RBG),gluco.e tolerance,insulin tolerance,and glycated serum protein(GSP) and the globe gene expression in liver tissues were *** results showed that catalpol treatment obviously reduced water intake and food intake in a dose-dependent *** treatment also remarkably reduce fasting blood gluco.e(FBG) and random blood gluco.e(RBG) in a dose-dependent *** RBG-lowering effect of catalpol was better than that of ***,catalpol significantly improved gluco.e tolerance and insulin tolerance via increasing insulin *** treatment significantly decreased GSP *** co.parisons of gene expression in liver tissues among normal co.trol mice,db/db mice and catalpol treated mice(200 and 100 mg·kg^(-1)) indicated that there were significant increases in the expressions of 287 genes,whichwere mainly involved in lipid metabolism,response to stress,energy metabolism,and cellular processes,and significant decreases in the expressions of 520 genes,which were mainly involved in cell growth,death,immune system,and response to *** genes expressed differentially were linked to gluco.e metabolism or insulin signaling pathways,including Irs1(insulin receptor substrate 1),Idh2(isocitrate dehydrogenase 2(NADP+),mitochondrial),G6pd2(gluco.e-6-phosphate dehydrogenase 2),and SOCS3(suppressor of cytokine signaling 3).In co.clusion,catalpol e

A novel AgNPs/MOF substrate-based SERS sensor for high-sensitive on-site detection of wheat gluten

Gluten,known as the major allergen in wheat,has gained increasing co.cerns in industrialized co.ntries,resulting in an urgent need for accurate,high-sensitive,and on-site detection of wheat gluten in co.plex food ***,we proposed a silver nanoparticles(AgNPs)/metal-organic framework(MOF)substrate-based surface-enhanced Raman scattering(SERS)sensor for the high-sensitive on-site detection of wheat *** detection occurred on the newly in-situ synthesized AgNPs/MOF-modified SERS substrate,providing an enhancement factor(EF)of 1.89×10^(5).Benefitting from the signal amplification function of AgNPs/MOF and the superiority of SERS,this sensor represented high sensitivity performance and a wide detection range from 1×10^(-15)mol/L to 2×10^(-6)mol/L with a detection limit of 1.16×10^(-16)mol/L,which allowed monitoring the trace of wheat gluten in co.plex food system without matrix *** reliable sandwich SERS sensor may provide a promising platform for high-sensitive,accurate,and on-site detection of allergens in the field of food safety.

Gastric ulcer is a widespread disease caused by various *** offi cinale flowers exert several health benefits owing to their rich flavonoid *** this study,protective effects and possible action mechanisms of *** cinale flowers’flavonoid enriched extract(DOFF)were assessed against gastric *** result of sodium nitrite-aluminum nitrate co.orimetry showed that 52.34%of the total extractive was flavonoid,and ultra-high performance liquid chromatography time of flight mass spectrometer(UPLC-QTOF/MS)revealed the presence of 28 co.ponents in DOFF of which 14 belonged to *** addition,in vivo assay revealed DOFF potential in reducing the formation of ethanol-induced gastric muco.al lesions,with drop-in ulcer index from 64.33±8.76 to 32.00±*** results were revealed in human gastric muco.al epithelia cells,with cells viability to increase from 27.2%to 61.6%post DOFF *** analyzed the protect effect of DOFF,we used Western blotting and immunofluorometric assay to revealed the expression levels of key proteins in cell *** results showed that DOFF(320μg/mL)co.ld increase the level of oxidation marker protein(HO-1),apoptosis regulatory protein(Bcl-2)and autophagy marker(LC3β)by 50.84%,43.85%,and 59.21%co.pared with ethanol-treated group *** analyzed of the mitochondrial activity and apoptosis pathway,we found that DOFF appeared to mitigate against ethanolinduced gastric muco.al injury via AMPK/mTOR/ULK1 and PI3K/AKT autophagy signaling pathways.
