Spatio-temporal variation of water salinity in mangroves revealed by continuous monitoring and its relationship to floristic diversity

Salinity is among the most critical factors limiting the growth and species distribution of coastal plants.Water salinity in estuarine ecosystems varies temporally and spatially,but the variation patterns across different time scales and salinity fluctuation have rarely been quantified.The effects of salinity on floristic diversity in mangroves are not fully understood due to the temporal and spatial heterogeneity of salinity.In this study,we monitored water salinity at an interval of 10-min over one year in three mangrove catchment areas representing the outer part,middle part,and inner part respectively of Dongzhai Bay,Hainan,China.The number of mangrove community types and dominant mangrove species of the three catchment areas were also investigated.We found that the diurnal variation and dry-season intra-month variation in water salinity were driven by tidal cycles.The seasonal variation in water salinity was mainly driven by rainfall with higher salinity occurring in the dry season and lower salinity occurring in the wet season.Spatially,water salinity was highest at the outer part,intermediate at the middle part,and lowest at the inner part of the bay.The intra-month and annual fluctuations of water salinity were highest at the middle part and lowest at the outer part of the bay.The number of mangrove community types and dominant species were lowest at the outer part,intermediate at the middle part,and highest at the inner part of the bay.These results suggest that the temporal variation of water salinity in mangroves is driven by different factors at different time scales and therefore it is necessary to measure water salinity at different time scales to get a complete picture of the saline environment that mangroves experience.Spatially,lower salinity levels benefit mangrove species richness within a bay landscape,however,further research is needed to distinguish the effects of salinity fluctuation and salinity level in affecting mangrove species richness.

Brain Transcriptome Analysis Reveals Metabolic Changes Adapting to Hyperhaline or Hypohaline Environments in Spotted Scat(Scatophagus argus)

The fish brain is crucial for adjusting to environmental changes.Metabolic changes play a vital role in the adaptation to salinity change in aquatic animals.However,few studies have evaluated the responses of the fish brain to salinity changes.To evaluate the response to various salinities,spotted scat(Scatophagus argus)was cultured in water with salinity levels of 5(low salinity:LS),25(control group:Ctrl),and 35(high salinity group:HS)for 22 days.The brain transcriptome was analyzed.In total,1698 differentially expressed genes(DEGs)were identified between the HS and Ctrl groups,and 841 DEGs were identified between the LS and Ctrl groups.KEGG analysis showed that the DEGs in the HS vs.Ctrl comparison were involved in steroid biosynthesis,terpenoid backbone biosynthesis,fatty acid biosynthesis,ascorbate and aldarate metabolism,other types of O-glycan biosynthesis,and fatty acid metabolism.Glyoxylate and dicarboxylate metabolism,one carbon pool by folate,steroid biosynthesis,and cysteine and methionine metabolism were significantly enriched in the LS vs.Ctrl comparison.Additionally,the genes related to metabolism(acc,fas,hmgcr,hmgcs1,mvd,soat1,nsdhl,sqle,cel,fdft1,dnmt3a and mtr)were significantly up-regulated in the HS vs.Ctrl comparison.The genes related to metabolism(lipa,sqle,acc,fas,bhmt,mpst,dnmt3a,mtr,hao2,LOC111225351 and hmgcs1)were significantly up-regulated,while hmgcr and soat1 were significantly down-regulated in the LS vs.Ctrl compparison.These results suggest that salinity stress affects signaling pathways and genes’expressions involved in metabolic processes in the brain,and the differences in metabolism play an important role in adaptation to hyperhaline or hypohaline environments in spotted scat.This research provides a comprehensive overview of transcriptional changes in the brain under hyperhaline or hypohaline conditions,which is helpful to understand the mechanisms underlying salinity adaptation in euryhaline fishes.

Estimating the total allowable catch and management of Threadfin porgy(Evynnis cardinalis)fisheries in the northern South China Sea based on sampling surveys conducted at fishing ports

In order to develop a method that apply sampling survey data randomly obtained at fishing ports to fish stock assessments,based on fish landing surveys conducted at fishing ports in the northern South China Sea(SCS),19 Threadfin porgy(Evynnis cardinalis)catch per unit effort(CPUE)datasets were collected for fishing vessels with different gear types and engine powers and incorporated into surplus production models.Considering only the fitting performance,the Schaefer model had the best overall goodness of fit,followed by the Fox,CYP,W-H,and Schnute models.Among fishing vessels with different gears and engine powers,the data were best fitted for single-trawl vessels powered by 301-400-kW engines and for gillnet vessels powered by>200-kW engines.Eight model expressions were superior and selected for subsequent analyses based on their goodness of fit and relative residuals.The Kobe plot analysis results showed an optimistic fish stock status when using the four model expressions,required more caution when using three model expressions and output pessimistic estimations using one model expression.Considering the incomplete information acquired,a compromising decision-making method was used to derive a 2017 northern SCS E.cardinalis total allowed catch(TAC)of 44,691.21 t.The different conclusions drawn from estimations using CPUEs reflect variable exploitation and utilization fish stock statuses among fishing vessels with different gears and engine powers.Hence,the fishing operations were grouped according to their CPUE relationship,and recommendations regarding optimum fishing efforts were assigned to the groups following a fundamental principle:to improve fishery TAC management,fishing efforts should be reduced if the fish stock assessment is pessimistic and maintained if the assessment is optimistic.This study providing a feasible technical method for the TAC management of China’s offshore fisheries.

Risk Assessment of Deep-Water Horizontal X-Tree Installation

Due to the high potential risk and many influencing factors of subsea horizontal X-tree installation,to guarantee the successful completion of sea trials of domestic subsea horizontal X-trees,this paper established a modular risk evaluation model based on a fuzzy fault tree.First,through the analysis of the main process oftree down and combining the Offshore&Onshore Reliability Data(OREDA)failure statistics and the operation procedure and the data provided by the job,the fault tree model of risk analysis of the tree down installation was established.Then,by introducing the natural language of expert comprehensive evaluation and combining fuzzy principles,quantitative analysis was carried out,and the fuzzy number was used to calculate the failure probability of a basic event and the occurrence probability of a top event.Finally,through a sensitivity analysis of basic events,the basic events of top events significantly affected were determined,and risk control and prevention measures for the corresponding high-risk factors were proposed for subsea horizontal X-tree down installation.

A Transcriptome Analysis of Neural Tissue of Litopenaeus vannamei After Acute Exposure to Alexandrium pacificum

Alexandrium pacificum(A.pacificum)is a typical paralytic shellfish poisonous dinoflagellate.Harmful algal blooms(HABs)caused by this species can bring serious environmental problems and economic losses to the aquaculture industry.In this study,transcriptome sequencing and analyses were performed on the neural tissue of Litopenaeus vannamei(L.vannamei)after acute exposure to A.pacificum disrupted solution for 72 h,and differentially expressed genes(DEGs)were identified.The results showed that,compared with the control samples,300 DEGs were identified in the experimental group,of which 194 were up-regulated,and 106 down-regulated.The gene ontology(GO)functional enrichment analysis showed that DEGs were significantly enriched in the cortical cytoskeleton organization,troponin complex,amylo-alpha-1,6-glucosidase and thymidine phosphorylase.Kyoto encyclopedia of genes and genomes(KEGG)enrichment analysis found that DEGs were mainly enriched in the oxidative phosphorylation process,intercellular tight junctions and mitophagy.The results showed that the proteoglycans,signaling pathways,and various metabolic processes that regulate cell proliferation,differentiation,and apoptosis all played an essential role in the response of L.vannamei to A.pacificum toxins.

This comprehensive review elucidates the complex interplay between gut microbiota and constipation in Parkinson’s disease(PD),a prevalent non-motor symptom contributing significantly to patients’morbidity.A marked alteration in the gut microbiota,predominantly an increase in the abundance of Proteobacteria and Bacteroidetes,is observed in PD-related constipation.Conventional treatments,although safe,have failed to effectively alleviate symptoms,thereby necessitating the development of novel therapeutic strategies.Microbiological interventions such as prebiotics,probiotics,and fecal microbiota transplantation(FMT)hold therapeutic potential.While prebiotics improve bowel movements,probiotics are effective in enhancing stool consistency and alleviating abdominal discomfort.FMT shows potential for significantly alleviating constipation symptoms by restoring gut microbiota balance in patients with PD.Despite promising developments,the causal relationship between changes in gut microbiota and PD-related constipation remains elusive,highlighting the need for further research in this expanding field.

Endogenous Trypsin Triggers Free Fluoride Release from Antarctic Krill(Euphausia superba)Cuticle

During postmortem storage,fluoride in Antarctic krill can be enriched in the muscle.Trypsin,as the most important digestive enzyme in Antarctic krill with a high activity in low temperature,plays a potential role in this process.In this study,endogenous trypsin was purified and its properties were investigated.The involvement of trypsin in the generation of free fluoride from Antarctic krill cuticle was explored.Cuticle microstructure before and after hydrolysis was compared with scanning electron microscopy,and the ash samples of the hydrolyzed Antarctic krill cuticle were analyzed with X-ray diffraction,Fourier transform infrared spectroscopy,and electron dispersive spectroscopy,respectively.Mass spectrometry analysis and inhibition tests confirmed that the purified enzyme was endogenous trypsin.Results of the present study indicated that trypsin digestion caused the increases of the concentrations of both fluoride ions and free amino N simultaneously,while the protein coated on the cuticle surface was dissolved too.However,no compositional change was detected in the cuticle inorganic salts.These findings suggest that trypsin triggered free fluoride release from Antarctic krill cuticle.In addition,the kinetics of free fluoride release could be described by the equation C_(W)=(1-0.97^(-0.006t)-0.03e^(0.0558t))x337.53+10.50.The present study improved the understanding of the role of trypsin in free fluoride release from Antarctic krill cuticle,facilitating future studies aimed at reducing the fluoride content in krill protein during Antarctic krill processing.

Silencing of the long non-coding RNA LINC00265 triggers autophagy and apoptosis in lung cancer by reducing protein stability of SIN3A oncogene

Background:Long non-coding RNAs are important regulators in cancer biology and function either as tumor suppressors or as oncogenes.Their dysregulation has been closely associated with tumorigenesis.LINC00265 is upregulated in lung adenocarcinoma and is a prognostic biomarker of this cancer.However,the mechanism underlying its function in cancer progression remains poorly understood.Methods:Here,the regulatory role of LINC00265 in lung adenocarcinoma was examined using lung cancer cell lines,clinical samples,and xenografts.Results:We found that high levels of LINC00265 expression were associated with shorter overall survival rate of patients,whereas knockdown of LINC00265 inhibited proliferation of cancer cell lines and tumor growth in xenografts.Western blot andﬂow cytometry analyses indicated that silencing of LINC00265 induced autophagy and apoptosis.Moreover,we showed that LINC00265 interacted with and stabilized the transcriptional co-repressor Switch-independent 3a(SIN3A),which is a scaffold protein functioning either as a tumor repressor or as an oncogene in a context-dependent manner.Silencing of SIN3A also reduced proliferation of lung cancer cells,which was correlated with the induction of autophagy.These observations raise the possibility that LINC00265 functions to promote the oncogenic activity of SIN3A in lung adenocarcinoma.Conclusions:Ourﬁndings thus identify SIN3A as a LINC00265-associated protein and should help to understand the mechanism underlying LINC00265-mediated oncogenesis.

Interplay between microglia and environmental risk factors in Alzheimer's disease

Alzheimer s disease,among the most common neurodegenerative disorders,is chara cterized by progressive cognitive impairment.At present,the Alzheimer’s disease main risk remains genetic ris ks,but major environmental fa ctors are increasingly shown to impact Alzheimer’s disease development and progression.Microglia,the most important brain immune cells,play a central role in Alzheimer’s disease pathogenesis and are considered environmental and lifestyle"sensors."Factors like environmental pollution and modern lifestyles(e.g.,chronic stress,poor dietary habits,sleep,and circadian rhythm disorde rs)can cause neuroinflammato ry responses that lead to cognitive impairment via microglial functioning and phenotypic regulation.However,the specific mechanisms underlying interactions among these facto rs and microglia in Alzheimer’s disease are unclear.Herein,we:discuss the biological effects of air pollution,chronic stress,gut micro biota,sleep patterns,physical exercise,cigarette smoking,and caffeine consumption on microglia;consider how unhealthy lifestyle factors influence individual susceptibility to Alzheimer’s disease;and present the neuroprotective effects of a healthy lifestyle.Toward intervening and controlling these environmental risk fa ctors at an early Alzheimer’s disease stage,understanding the role of microglia in Alzheimer’s disease development,and to rgeting strategies to to rget microglia,co uld be essential to future Alzheimer’s disease treatments.

Role of targeting ferroptosis as a component of combination therapy in combating drug resistance in colorectal cancer

Colorectal cancer(CRC)is a form of cancer that is often resistant to chemotherapy,targeted therapy,radiotherapy,and immunotherapy due to its genomic instability and inflammatory tumor microenvironment.Ferroptosis,a type of non-apoptotic cell death,is characterized by the accumulation of iron and the oxidation of lipids.Studies have revealed that the levels of reactive oxygen species and glutathione in CRC cells are significantly lower than those in healthy colon cells.Erastin has emerged as a promising candidate for CRC treatment by diminishing stemness and chemoresistance.Moreover,the gut,responsible for regulating iron absorption and release,could influence CRC susceptibility through iron metabolism modulation.Investigation into ferroptosis offers new insights into CRC pathogenesis and clinical management,potentially revolutionizing treatment approaches for therapy-resistant cancers.
