A transcriptional cascade mediated by two APETALA2 family members orchestrates carotenoid biosynthesis in tomato

Carotenoids are important nutrients for human health that must be obtained from plants since they cannot be biosynthesized by the human *** the regulatory mechanism of carotenoid metabolism in plants represents the first step toward manipulating carotenoid contents in plants by molecular design *** this study,we determined that SlAP2c,an APETALA2(AP2)family member,acts as a transcriptional repressor to regulate carotenoid biosynthesis in tomato(Solanum lycopersicum).Knockout of SlAP2c in both the“Micro Tom”and“Ailsa Craig”backgrounds resulted in greater lycopene accumulation,whereas overexpression of this gene led to orange-ripe fruit with significantly lower lycopene contents than the wild *** established that SlAP2c represses the expression of genes involved in lycopene biosynthesis by directly binding to the cis-elements in their ***,SlAP2c relies on its EAR motif to recruit the co-repressors TOPLESS(TPL)2/4 and forms a complex with histone deacetylase(had)1/3,thereby reducing the histone acetylation levels of lycopene biosynthesis ***,SlAP2a,a homolog of SlAP2c,acts upstream of SlAP2c and alleviates the SlAP2c-induced repression of lycopene biosynthesis genes by inhibiting SlAP2c transcription during fruit ***,we identified a transcriptional cascade mediated by AP2 family members that regulates lycopene biosynthesis during fruit ripening in tomato,laying the foundation for the manipulation of carotenoid metabolism in plants.

A Case Report of a Rare Sarcomatoid Poorly Differentiated Adenocarcinoma Harboring Concurrent Mutations in the ROS1, EGFR, ARID1A, and NFKBIA Genes in the Lung

ROS1 and EGFR are primary oncogenic drivers in non-small cell lung cancer (NSCLC) pathogenesis. However, EGFR mutations and ROS1 fusions are generally mutually exclusive in NSCLC, leading to a negligible probability of their co-occurrence. Consequently, clinical data and treatment strategies for their simultaneous presence are remarkably scarce. This report details the first recorded case of a sarcomatoid, poorly differentiated lung adenocarcinoma harboring both a ROS1 fusion and an EGFR mutation, alongside ARID1A and NFKBIA gene mutations. Moreover, this case study encompasses a review of instances featuring concurrent ROS1 and EGFR mutations. The identified genetic alterations in ROS1, EGFR, ARID1A, and NFKBIA are pivotal in the etiology of NSCLC. These mutations significantly influence disease progression and are essential for the development of personalized therapeutic approaches. Recognizing the unique genetic profiles in patients permits healthcare providers to devise customized treatment regimens that target these specific mutations, thereby enhancing patient outcomes in NSCLC.

Qualitative analysis of aromatic compounds via 1D TOCSY techniques

The aromatic compounds,including o-xylene,m-xylene,p-xylene,and ethylbenzene,primarily originate from the catalytic reforming of crude oil,and have a wide variety of ap***,because of similar physical and chemical properties,these compounds are difficult to be identified by gas chromatography(GC)without standard *** the development of modern nuclear magnetic resonance(NMR)techniques,NMR has emerged as a powerful and efficient tool for the rapid analysis of complex and crude mixtures without *** this study,the parameters of one-dimensional(1D)total correlation spectroscopy(TOCSY)NMR techniques,including 1D selective gradient TOCSY and 1D chemicalshift-selective filtration(CSSF)with TOCSY,were optimized to obtain comprehensive molecular structure *** results indicate that the overlapped signals in NMR spectra of nonpolar aromatic compounds(including o-xylene,m-xylene,p-xylene and ethylbenzene),polar aromatic compounds(benzyl alcohol,benzaldehyde,benzoic acid),and aromatic compounds with additional conjugated bonds(styrene)can be resolved in 1D *** importantly,full molecular structures can be clearly distinguished by setting appropriate mixing time in 1D *** approach simplifies the NMR spectra,provides structural information of entire molecules,and can be applied for the analysis of other structural isomers.

Excessive carbon emissions have resulted in the greenhouse effect, causing considerable global climate change. Marine carbon storage has emerged as a crucial approach to addressing climate change. The Qiantang Sag(QTS) in the East China Sea Shelf Basin, characterized by its extensive area, thick sedimentary strata, and optimal depth, presents distinct geological advantages for carbon dioxide(CO_(2)) storage. Focusing on the lower section of the Shimentan Formation in the Upper Cretaceous of the QTS, this study integrates seismic interpretation and drilling data with core and thin-section analysis. We reveal the vertical variation characteristics of the strata by providing a detailed stratigraphic description. We use petrophysical data to reveal the development characteristics of high-quality carbon-storage layers and favorable reservoircaprock combinations, thereby evaluating the geological conditions for CO_(2) storage in various stratigraphic sections. We identify Layer B of the lower Shimentan Formation as the most advantageous stratum for marine CO_(2) storage. Furthermore, we analyze the carbon emission trends in the adjacent Yangtze River Delta region. Considering the characteristics of the source and sink areas, we suggest a strong correlation between the carbon emission sources of the Yangtze River Delta and the CO_(2) storage area of the QTS, making the latter a priority area for conducting experiments on marine CO_(2) storage.

Phase Evolution of Cubic ZnS Annealed in Mild Oxidizing Atmosphere

ZnS thin films are prepared by thermal evaporation of high-purity ZnS powder on quartz glass *** samples were annealed in floating argon at temperatures from 300℃to 900℃.The effects of annealing temperature on the structural and optical properties were investigated by x-ray diffraction(XRD),scanning electron microscope(SEM)and optical *** results show that annealing below 500℃is beneficial to improve the quality of ZnS *** the annealing temperature exceeds 500℃,ZnS is gradually oxidized into ZnSO4,which has evident influences on the structural and optical properties of ZnS films.

Detection of a mud volcano in the Weitan Banks area of the northern South China Sea

Situated between the petroliferous Cenozoic Zhujiang(Pearl)River Mouth Basin and the mud volcano-rich Mesozoic Dongsha Basin in the middle sector of the northern South China Sea,the Weitan Banks area has been previously mapped as a basement high that is composed of Mesozoic magmatic *** this study,we present several favorable indicators for petroleum geology that were detected from geophysical profiling and benthic sampling in the area.A conspicuous hill was discovered,named“Zhongwei Hill”,~80 m high above the~340 m deep seafloor and~1 km broad,in a depression with more than 7 km thick sedimentary *** Zhongwei Hill was seismically imaged with a mushroom-shaped structure and containing a cake-like crown,fluid flow pipes,and an~10 km broad anticline at ***,the hill represents a source-plumbing-eruption *** gas zones linked to deep fracture were found at or near the *** correlation indicates that the deep strata comprise the Jurassic and Paleogene strata,the major hosts of hydrocarbon source *** addition to the hill,there are number of mounds from which three bottom water samples were collected and the samples are rich in dissolved methane with concentrations high up to~900 nmol/L,much higher than the background level(0.5–2 nmol/L).The benthic samples are rich in coarse sediment clastics,authigenic carbonate nodules,and deep-water habitats likely feeding on methanotrophic *** these observations and the context,we propose that the Zhongwei Hill represents a mud volcano,likely thermally driven,that seeps methane from Jurassic and Paleogene source layers,thus poses a favorable clue for significant hydrocarbon generation capacity in transitional zone of the Zhujiang River Mouth Basin and the Dongsha Basin.

Kinetics and model of gas generation of source rocks in the deepwater area, Qiongdongnan Basin

In order to investigate the hydrocarbon generation process and gas potentials of source rocks in deepwater area of the Qiongdongnan Basin, kinetic parameters of gas generation （activation energy distribution and frequency factor） of the Yacheng Formation source rocks （coal and neritic mudstones） was determined by thermal simulation experiments in the closed system and the specific KINETICS Software. The results show that the activation energy （Ea） distribution of C1–C5 generation ranges from 50 to 74 kcal/mol with a frequency factor of 2.4×1015 s–1 for the neritic mudstone and the Ea distribution of C1–C5 generation ranges from 49 to 73 kcal/mol with a frequency factor of 8.92×1013 s–1 for the coal. On the basis of these kinetic parameters and combined with the data of sedimentary burial and paleothermal histories, the gas generation model of the Yacheng Formation source rocks closer to geological condition was worked out, indicating its main gas generation stage at Ro （vitrinite reflectance） of 1.25%–2.8%. Meanwhile, the gas generation process of the source rocks of different structural locations （central part, southern slope and south low uplift） in the Lingshui Sag was simulated. Among them, the gas generation of the Yacheng Formation source rocks in the central part and the southern slope of the sag entered the main gas window at 10 and 5 Ma respectively and the peak gas generation in the southern slope occurred at 3 Ma. The very late peak gas generation and the relatively large gas potential indices （GPI：20×10^8–60×10^8 m^3/km^2） would provide favorable conditions for the accumulation of large natural gas reserves in the deepwater area.

CO2 geological storage into a lateral aquifer of an offshore gas field in the South China Sea： storage safety and project design

The DFI-1 gas South China Sea, contains thus there is great concern field, located in the western a high concentration of CO2, about the need to reduce the CO2 emissions. Many options have been considered in recent years to dispose of the CO2 separated from the natural gas stream on the Hainan Island. In this study, the feasibility of CO2 storage in the lateral saline aquifer of the DF 1-1 gas field is assessed, including aquifer selection and geological assessment, CO2 migration and storage safety, project design, and economic analysis. Six offshore aquifers have been investigated for CO2 geological storage. The lateral aquifer of the DFI-1 gas field has been selected as the best target for CO2 injection and storage because of its proven sealing ability, and the large storage capacity of the combined aquifer and hydrocarbon reservoir geological structure. The separated CO2 will be dehydrated on the Hainan Island and transported by a long- distance subsea pipeline in supercritical or liquid state to the central platform of the DFI-1 gas field for pressure adjustment. The CO2 will then be injected into the lateral aquifer via a subsea well-head through a horizontal well. Reservoir simulations suggest that the injected CO2 will migrate slowly upwards in the aquifer without disturbing the natural gas production. The scoping economic analysis shows that the unit storage cost of the project is approximately US$26-31/ton CO2 with the subsea pipe- line as the main contributor to capital expenditure （CAPEX）, and the dehydration system as the main factor of operating expenditure （OPEX）.

L1 is one of the largest offshore gas fields currently under *** order to optimize the related design,nodal analysis is applied(including proper consideration of the plant productivity,sensitivity to the tubing size,erosion effects,liquid carrying performance,and tubing string).As a result of such approach,it is shown that 13Cr material should be chosen as the appropriate tubing ***,3-1/2 inches 9.3 lb/ft N80 tubing,4-1/2 inches 12.75 lb/ft N80 tubing,5-1/2 inches 17 lb/ft N80 tubing should be used for a gas production rate under 80×10^(4)m^(3)/d,between 80×10^(4)m^(3)/d and 120×10^(4)m^(3)/d and above 120×10^(4)m^(3)/d,respectively.

Background and Aims:Voriconazole(VRC),a widely used antifungal drug,often causes hepatotoxicity,which presents a significant clinical *** studies demonstrated that Astragalus polysaccharide(APS)can regulate VRC metabolism,thereby potentially mitigating its hepatotoxic *** this study,we aimed to explore the mechanism by which APS regulates VRC ***:First,we assessed the association of abnormal VRC metabolism with hepatotoxicity using the Roussel Uclaf Causality Assessment Method ***,we conducted a series of basic experiments to verify the promotive effect of APS on VRC *** in vitro and in vivo assays,including cytokine profiling,immunohistochemistry,quantitative polymerase chain reaction,metabolite analysis,and drug concentration measurements,were performed using a lipopolysaccharideinduced rat inflammation ***,experiments such as intestinal biodiversity analysis,intestinal clearance assessments,and Bifidobacterium bifidum replenishment were performed to examine the ability of *** to regulate the expression of the VRC-metabolizing enzyme CYP2C19 through the gut–liver ***:The results indicated that APS does not have a direct effect on ***,the assessment of gut microbiota function revealed that APS significantly increases the abundance of ***,which could lead to an anti-inflammatory response in the liver and indirectly enhance VRC *** dual-luciferase reporter gene assay revealed that APS can hinder the secretion of pro-inflammatory mediators and reduce the inhibitory effect on CYP2C19 transcription through the nuclear factor-􀇉B signaling ***:The study offers valuable insights into the mechanism by which APS alleviates VRC-induced liver damage,highlighting its immunomodulatory influence on hepatic tissues and its indirect regulatory control of VRC-metabolizing enzymes within hepatocytes.
