Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Genome sequencing and phylogenetic analysis of Banna virus(genus Seadornavirus,family Reoviridae)isolated from Culicoides

In an investigation of blood-sucking insects and arboviruses, a virus(YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of yunnan Province, China. The virus caused cytopathic effect(CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA(dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region(1.12 segments) were17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 protein(RdRp) revealed that YN12243 belonged to genus Seadornavirus of family Reoviridae, and further analysis indicated that YN12243 belongs to the Banna virus(BAV) genotype A2. Additionally, YN12243 was located in the same evolutionary cluster as BAV strains isolated from different mosquito species, suggesting that the BAV isolated from Culicoides does not have species barriers. These results indicate that Culicoides can also be a vector for BAV. In view of the hematophagous habits of Culicoides on cattle, horses, deer, and other large animals, as well as the possibility of spreading and causing a variety of animal arboviral diseases, it is important to improve infection detection and monitor the BAV in large livestock.

Introduction:The epidemic of human anthrax is at a low level in China in recent years,but the reported incidence increased in *** order to understand the current landscape of research and knowledge about anthrax in China,the epidemiological characteristics of anthrax in humans from 2018 to 2021 were analyzed and the prevention and control suggestions were ***:Surveillance data of anthrax in humans and livestock,together with human outbreaks data during 2018-2021,were collected and analyzed by descriptive statistics *** number and proportion of outbreaks,cases and deaths by provincial-level administrative divisions(PLADs),clinical types,and contributing factors were ***:A total of 1,244 cases of human anthrax and 53 outbreaks were reported from 2018 to 2021 in *** the incidence of anthrax declined from 2018 to 2020,it increased in *** regions of anthrax were mainly located in the west and the northeast PLADs of China,though cases were reported in some central and eastern PLADs in *** and middle-aged men involved in animal husbandry were found to be at a higher risk of *** the reported outbreaks were associated with the exposure of infected livestock.A total of 296 livestock anthrax cases were ***:The increased incidence and wider geographical distribution of human anthrax in 2021 were found to be the result of inadequate supervision of diseased animals as well as updated diagnostic *** such,the monitoring of risk factors and emergency preparation procedures should be strengthened at the national *** addition,it is also critical to strengthen health education for high-risk occupational groups and strengthen professional training for local ***,more measures should be carried out to strengthen anthrax surveillance in livestock husbandry.

Kaschin-Beck disease (KBD) is an endemic and chronic osteoarthropathy characterized by pathological aspects including chondrocyte degeneration, necrosis, progressive loss of articular cartilage, and secondary degenerative osteoarthrosis of epiphyseal cartilage, epiphyseal plate cartilage, and articular cartilage, during puberty[1]. The main clinical symptoms are limb joint pain, thickening, deformation, limited movement, muscle atrophy, and in case of more severely affected patients, short fingers (toes), short limbs, and even short stature[1].

The first indigenous incidence of Mpox(previously known as monkeypox)within Chinese mainland was documented in May 2023,with subsequent local and imported cases identified.A comprehensive understanding of the Mpox virus’s(MPXV)characteristics within Beijing remains *** this study,84 MPXV genomes from 82 local incidents and two imported instances,detected between May and July 2023,were *** MPXV strains fell within lineage C.1 of the West African clade,displaying limited genetic heterogeneity,encompassing 76–87 nucleotide substitutions and holding nucleotide identities between 99.996%and 100%.Phylogenetic exploration indicated that all genomes exhibited high homology to those presently prevalent in neighboring East Asian and Southeast Asian ***-six distinct haplotypes were identified among the strains,with 36.90%of genomes corresponding to four common haplotypes,suggesting repeated cross-regional introductions and restrained distribution via recurrent local *** findings elucidate the genetic diversity and phylogenesis of MPXVs during their nascent transmission within Beijing and provide vital information to enhance future Mpox containment strategies.

Hepatitis E virus(HEV)is the prototype of the family Hepeviridae and the causative agent of common acute viral *** diverse HEV-related viruses have been detected in a variety of mammals and some of them may have zoonotic *** this study,we tested 278 specimens collected from seven wild small mammal species in yunnan province,China,for the presence and prevalence of orthohepevirus by broad-spectrum reverse transcription(RT)-***-related sequences were detected in two rodent species,including Chevrier’s field mouse(Apodemus chevrieri,family Muridae)and Père David’s vole(Eothenomys melanogaster,family Cricetidae),with the infection rates of 29.20%(59/202)and 7.27%(4/55),*** four representative full-length genomes were generated:two each from Chevrier’s field mouse(named Rd HEVAc14 and Rd HEVAc86)and Père David’s vole(Rd HEVEm40 and Rd HEVEm67).Phylogenetic analyses and pairwise distance comparisons of whole genome sequences and amino acid sequences of the gene coding regions showed that orthohepeviruses identified in Chinese Chevrier’s field mouse and Père David’s vole belonged to the species Orthohepevirus C but were highly divergent from the two assigned genotypes:HEV-C1 derived from rat and shrew,and HEV-C2 derived from ferret and possibly *** real-time RT-PCR demonstrated that these newly discovered orthohepeviruses had hepatic *** summary,our work discovered two putative novel genotypes orthohepeviruses preliminarily named HEVC3 and HEV-C4 within the species Orthohepevirus C,which expands our understanding of orthohepevirus infection in the order Rodentia and gives new insights into the origin,evolution,and host range of orthohepevirus.

Effect of Reduction in Iodine Content of Edible Salt on the Iodine Status of the Chinese Population

Objective The aim of this study was to evaluate the impact of the revised Chinese National Standard GB26878-2011 ＇Iodine Content in Edible Salt＇ on the iodine status among the Chinese population. Methods In 2011 and 2014, the probability proportionate to size sampling（PPS） was used in each Chinese province to obtain the representative data. In each sampling unit, school children aged 8-10 years and pregnant women were selected. Key indicators included urinary iodine concentration（UIC）, thyroid volume（TV）, and the iodine content in edible household salt. Results The median urinary iodine concentration（MUIC） decreased between 2011 and 2014 from 238.6 to 197.9 μg/L in school-age children. The number of provinces with iodine excess decreased to zero. The proportion of children whose UIC was 〉 300 μg/L was 18.8% and decreased to 11% compared with 29.8% in 2011. There was no significant difference in UIC 〈 50 μg/L between 2014（4.3%） and 2011（3.7%）（P 〉 0.05）. The MUIC among pregnant women in 2014 was more concentrated between 110 and 230 μg/L. The goiter rate among children aged 8-10 years was unchanged, both the goiter rate of 2011 and 2014 remaining below 5%, in view of the sustainable elimination of iodine deficiency disorders. Conclusion The National Standard GB26878-2011 ＇Iodine Content in Edible Salt＇ that was introduced in March 2012 resulted in an overall improvement in iodine status, reducing the risk of excessive iodine intake in the Chinese population.
