Sanchuan ham is appreciated in yunnan Province,China,for its characteristic flavor and taste,while the microbial community structure and biogenic amines content remain unclear during fermentation *** this study,we explored the physicochemical property,biogenic amines concentration and microbial diversity of external and internal Sanchuan ham by high-throughput sequencing during the processing of Sanchuan *** showed that the nitrite remained at a stable level of 0.15 mg/kg which was significantly lower than the national health standard safety level of 20 mg/*** addition,compared with fresh hams,the content of total free amino acids in ripe Sanchuan ham has grown 14 folds;sour and bitter were the main tastes of Sanchuan ***,the concentration of cadaverine was the highest of all biogenic amines during the entire fermentation *** the bacterial phyla level,Firmicutes and Actinobacteria were the two main phyla,while at the genus level,Staphylococcus was a significant strain throughout the whole ***,the dry stage has a great impact on the succession change of microbial community ***,the change trends and composition of bacteria in the interior have slight discrepancies with those of the exterior of Sanchuan ham.

Variation of microbiological and small molecule metabolite profiles of Nuodeng ham during ripening by high-throughput sequencing and GC-TOF-MS

The internal microbial diversity and small molecular metabolites of Nuodeng ham in different processing years(the first,second and third year sample)were analyzed by high-throughput sequencing technology and gas chromatography-time of flight mass spectrography(GC-TOF-MS)to study the effects of microorganisms and small molecular metabolites on the quality of ham in different processing *** results showed that the dominant bacteria phyla of Nuodeng ham in different processing years were Proteobacteria and Firmicutes,the dominant fungi phyla were Ascomycota and Basidiomycota,while Staphylococcus and Aspergillus were the dominant bacteria and fungi of Nuodeng ham,***,252 kinds of small molecular metabolites were identified from Nuodeng ham in different processing years,and 12 different metabolites were screened through multivariate statistical *** metabolic pathway analysis showed that 23 metabolic pathways were related to ham fermentation,of which 8 metabolic pathways had significant effects on ham fermentation(Impact>0.01,P<0.05).The content of L-proline,phenyllactic acid,L-lysine,carnosine,taurine,D-proline,betaine and creatine were significantly positively correlated with the relative abundance of Staphylococcus and Serratia,but negatively correlated with the relative abundance of Halomonas,Aspergillus and Yamadazyma.

The application of a male-sterile line is an ideal approach for hybrid seed production in non-heading Chinese cabbage(Brassica rapa ***).However,the molecular mechanisms underlying male sterility in *** are still largely *** previously obtained the natural male sterile line WS24-3 of non-heading Chinese cabbage and located the male sterile locus,Bra2Ms,on the A2 *** observations revealed that the male sterility of WS24-3 resulted from disruption of the meiosis process during pollen *** mapping of Bra2Ms delimited the locus within a physical distance of about 129 kb on the A2 chromosome of *** Bra039753 gene encodes a plant homeodomain(PHD)-finger protein and is considered a potential candidate gene for ***039753 was significantly downregulated in sterile line WS24-3 compared to the fertile line at the meiotic anther *** analysis of Bra039753 identified a 369 bp fragment insertion in the first exon in male sterile plants,which led to an amino acid insertion in the Bra039753 *** addition,the 369 bp fragment insertion was found to cosegregate with the male sterility *** study identified a novel locus related to male sterility in non-heading Chinese cabbage,and the molecular marker obtained in this study will be beneficial for the marker-assisted selection of excellent sterile lines in non-heading Chinese cabbage and other Brassica crops.

Plant height(PH)is associated with lodging resistance and planting density,which is regulated by a complicated gene *** this study,we identified a spontaneous dwarfing mutation in maize,m30,with decreased internode number and length but increased internode diameter.A candidate gene,ZmCYP90D1,which encodes a member of the cytochrome P450 family,was isolated by map-based ***90D1 was constitutively expressed and showed highest expression in basal internodes,and its protein was targeted to the nucleus.A G-to-A substitution was identified to be the causal mutation,which resulted in a truncated protein in *** of function of ZmCYP90D1 changed expression of hormoneresponsive genes,in particular brassinosteroid(BR)-responsive genes which is mainly involved in cell cycle regulation and cell wall extension and modification in *** concentration of typhasterol(TY),a downstream intermediate of ZmCYP90D1 in the BR pathway,was reduced.A haplotype conferring dwarfing without reducing yield was ***90D1 was inferred to influence plant height and stalk diameter via hormone-mediated cell division and cell growth via the BR pathway.

Background C16:0 and cis-9 C18:1 may have different effects on animal growth and health due to unique metabolism in *** study was investigated to explore the different effects of altering the ratio of C16:0 and cis-9 C18:1 in fat supplements on growth performance,lipid metabolism,intestinal barrier,cecal microbiota,and inflammation in fattening *** finishing Angus bulls(626±69 kg,21±0.5 months)were divided into 3 treatments according to the randomized block design:(1)control diet without additional fat(CON),(2)CON+2.5%palmitic acid calcium salt(PA,90%C16:0),and(3)CON+2.5%mixed fatty acid calcium salt(MA,60%C16:0+30%cis-9 C18:1).The experiment lasted for 104 d,after which all the bulls were slaughtered and sampled for *** MA tended to reduce 0–52 d dry matter intake compared to PA(DMI,P=0.052).Compared with CON and MA,PA significantly increased 0–52 d average daily gain(ADG,P=0.027).PA tended to improve the 0–52 d feed conversion rate compared with CON(FCR,P=0.088).Both PA and MA had no significant effect on 52–104 days of DMI,ADG and FCR(P>0.05).PA tended to improve plasma triglycerides compared with MA(P=0.077),significantly increased plasma cholesterol(P=0.002)and tended to improve subcutaneous adipose weight(P=0.066)when compared with CON and *** PA and MA increased visceral adipose weight compared with CON(P=0.021).Only PA increased the colonization of Rikenellaceae,Ruminococcus and Proteobacteria in the cecum,and MA increased Akkermansia abundance(P<0.05).Compared with CON,both PA and MA down-regulated the m RNA expression of Claudin-1 in the jejunum(P<0.001),increased plasma diamine oxidase(DAO,P<0.001)and lipopolysaccharide(LPS,P=0.045).Compared with CON and MA,PA down-regulated the ZO-1 in the jejunum(P<0.001)and increased plasma LPS-binding protein(LBP,P<0.001).Compared with CON,only PA down-regulated the Occludin in the jejunum(P=0.013).Compared with CON,PA and MA significantly up-regulated the expression of TLR-4 and NF-κB

Tumor necrosis factorα(TNFα)exhibits diverse biological functions;however,its regulatory roles in myogenesis are not fully *** the present study,we explored the function of TNFαin myoblast proliferation,differentiation,migration,and myotube fusion in primary myoblasts and C2C12 *** this end,we constructed TNFαmuscle-conditional knockout(TNFα-CKO)mice and compared them with flox mice to assess the effects of TNFαknockout on skeletal *** indicated that TNFα-CKO mice displayed phenotypes such as accelerated muscle development,enhanced regenerative capacity,and improved exercise endurance compared to flox mice,with no significant differences observed in major visceral organs or skeletal *** label-free proteomic analysis,we found that TNFα-CKO altered the distribution of several muscle development-related proteins,such as Hira,Casz1,Casp7,Arhgap10,Gas1,Diaph1,Map3k20,Cfl2,and Igf2,in the nucleus and *** set enrichment analysis(GSEA)further revealed that TNFαdeficiency resulted in positive enrichment in oxidative phosphorylation and MyoD targets and negative enrichment in JAK-STAT *** findings suggest that TNFα-CKO positively regulates muscle growth and development,possibly via these newly identified targets and pathways.

Walnut dreg protein hydrolysates(WDPHs)exhibit a variety of biological activities,however,the cyclooxygenase-2(COX-2)inhibitory peptide of WDPHs remain *** aim of this study was to rapidly screen for such peptides in WDPHs through a combination of in silico and in vitro *** total,1262 peptide sequences were observed by nano liquid chromatography/tandem mass spectrometry(nano LC-MS/MS)and 4 novel COX-2 inhibitory peptides(AGFP,FPGA,LFPD,and VGFP)were *** kinetic data indicated that AGFP,FPGA,and LFPD displayed mixed-type COX-2 inhibition,whereas VGFP was a non-competitive *** is mainly because the peptides form hydrogen bonds and hydrophobic interactions with residues in the COX-2 active *** results demonstrate that computer analysis combined with in vitro evaluation allows for rapid screening of COX-2 inhibitory peptides in walnut protein dregs.

Rhizosphere microbes influence wogonoside accumulation in perennial Scutellaria baicalensis

Dried roots of Scutellaria baicalensis Georgi are an acclaimed traditional Chinese medicine,and wogonoside content is a key indicator used to evaluate *** *** than rising linearly with cultivation years,*** quality initially increases and then ***,little is known regarding the dynamic variations in *** rhizosphere microorganisms under long-term cultivation and underlying mechanisms of their effects on wogonoside formation and *** aim of this study was to investigate the roles of soil nutrients and rhizosphere microbes on *** quality across different cultivation years(1-4 years).The wogonoside content(25.14 mg g^(-1))was highest in the biennial *** and then decreased following long-term *** soil nutrients(available nitrogen,available phosphorus,available iron,available manganese,and available zinc)were reduced significantly as cultivation years *** also affected rhizosphere bacterial community structure significantly,driving it toward deterministic process(i.e.,β-nearest taxon index<-2).Available manganese and exchangeable calcium indirectly affected wogonoside formation and *** inhibition was driven by the reduction in rhizosphere bacterial diversity,which significantly increased the relative abundance of beta-glucosidase and decreased the relative abundance of phenylalanine *** rhizosphere microenvironment was altered under long-term cultivation,thereby shaping rhizosphere bacterial community,reducing the community diversity,and ultimately inhibiting wogonoside formation and *** findings may aid in understanding of the mechanisms and reasons for the reduction in *** quality under long-term cultivation from the perspective of soil nutrients and microorganisms,which may theoretically support the future artificial cultivation and management of Chinese medicinal plants.

The near-infrared imaging channel constitutes a crucial component of the multichannel high-resolution imaging system of the New Vacuum Solar Telescope(NVST). We have successfully achieved high-resolution, narrowband imaging of the chromosphere using He I 10830 A triplet within this channel, which significantly enhances the imaging observation capabilities of NVST. This paper provides a concise overview of the optical system associated with the near-infrared imaging channel, detailing data processing procedures and presenting several observed images. Leveraging a high-resolution image reconstruction algorithm, we were able to generate a narrowband image near the diffraction limit at 10830 A with a temporal resolution of less than 10 s.

Prediction plays an important role in data *** averaging method generally provides better prediction than using any of its *** though model averaging has been extensively investigated under independent errors,few authors have considered model averaging for semiparametric models with correlated *** this paper,the authors offer an optimal model averaging method to improve the prediction in partially linear model for longitudinal *** model averaging weights are obtained by minimizing criterion,which is an unbiased estimator of the expected in-sample squared error loss plus a *** properties,including asymptotic optimality and consistency of averaging weights,are established under two scenarios:(i)All candidate models are misspecified;(ii)Correct models are available in the candidate *** studies and an empirical example show that the promise of the proposed procedure over other competitive methods.
