Sphingolipid inhibitor response gene GhMYB86 controls fiber elongation by regulating microtubule arrangement

Although the cell membrane and cytoskeleton play essential roles in cellular morphogenesis, the interaction between the membrane and cytoskeleton is poorly understood. Cotton fibers are extremely elongated single cells, which makes them an ideal model for studying cell development. Here, we used the sphingolipid biosynthesis inhibitor, fumonisin B1(FB1), and found that it effectively suppressed the myeloblastosis(MYB) transcription factor Gh MYB86,thereby negatively affecting fiber elongation. A direct target of Gh MYB86 is Gh TUB7, which encodes the tubulin protein, the major component of the microtubule cytoskeleton. Interestingly, both the overexpression of Gh MYB86 and Gh TUB7 caused an ectopic microtubule arrangement at the fiber tips,and then leading to shortened fibers. Moreover, we found that Gh MBE2 interacted with Gh MYB86 and that FB1 and reactive oxygen species induced its transport into the nucleus, thereby enhancing the promotion of Gh TUB7 by Gh MYB86. Overall, we established a Gh MBE2-Gh MYB86-Gh TUB7 regulation module for fiber elongation and revealed that membrane sphingolipids affect fiber elongation by altering microtubule arrangement.

Plant-based fermentations provide an untapped source for novel biotechnological *** this study,a probiotic named Lactobacillus fermentum 21828 was introduced to ferment Lentinus *** were extracted from fermented and non-fermented *** and purified via DEAE-52 and Sephadex *** components designated F-LEP-2a and NF-LEP-2a were analyzed by FT-IR,HPGPC,HPAEC,SEM,GC-MS and *** results revealed that probiotic fermentation increased the molecular weight from 1.16×10^(4) Da to 1.87×10^(4) Da and altered the proportions of glucose,galactose and mannose,in which glucose increased from 45.94%to 48.16%.Methylation analysis and NMR spectra indicated that F-LEP-2a and NF-LEP-2a had similar linkage ***,their immunomodulatory activities were evaluated with immunosuppressive ***-LEP and F-LEP improved immune organ indices,immunoglobulin(Ig G and Ig M)and cytokines concentrations;restored the antioxidation capacity of liver;and maintained the balance of gut microbiota.F-LEP displayed better moderating effects on the spleen index,immunoglobulin,cytokines and the diversity of gut microbiota than NF-LEP(200,400 mg/kg).Our study provides an efficient and environment-friendly way for the structural modification of polysaccharides,which helps to enhance their biological activity and promote their wide application in food,medicine and other fields.

In this paper, we give improved error estimates for linearized and nonlinear CrankNicolson type finite difference schemes of Ginzburg-Landau equation in two dimensions. For linearized Crank-Nicolson scheme, we use mathematical induction to get unconditional error estimates in discrete L2and H1norm. However, it is not applicable for the nonlinear ***, based on a ‘cut-off ’ function and energy analysis method, we get unconditional L2and H1error estimates for the nonlinear scheme, as well as boundedness of numerical solutions. In addition, if the assumption for exact solutions is improved compared to before, unconditional and optimal pointwise error estimates can be obtained by energy analysis method and several Sobolev inequalities. Finally, some numerical examples are given to verify our theoretical analysis.

Cost-Effective Method of Gene Synthesis by Sequencing from Microchip-Derived Oligos for Droplet Cloning

Gene synthesis has provided important contributions in various fields including genomics and medicine. Current genes are 7 - 30 cents depending on the assembly and sequencing methods performed. Demand for gene synthesis has been increasing for the past few decades, yet available methods remain expensive. A solution to this problem involves microchip-derived oligonucleotides (oligos), an oligo pool with a substantial number of oligo fragments. Microchips have been proposed as a tool for gene synthesis, but this approach has been criticized for its high error rate during sequencing. This study tests a possible cost-effective method for gene synthesis utilizing fragment assembly and golden gate assembly, which can be employed for quicker manufacturing and efficient execution of genes in the near future. The droplet method was tested in two trials to determine the viability of the method through the accuracy of the oligos sequenced. A preliminary research experiment was performed to determine the efficacy of oligo lengths ranging from two to four overlapping oligos through Gibson assembly. Of the three oligo lengths tested, only two fragment oligos were correctly sequenced. Two fragment oligos were used for the second experiment, which determined the efficacy of the droplet method in reducing gene synthesis cost and speed. The first trial utilized a high-fidelity polymerase and resulted in 3% correctly sequenced oligos, so the second trial utilized a non-high-fidelity polymerase, resulting in 8% correctly sequenced oligos. After calculating, the cost of gene synthesis lowers down to 0.8 cents/base. The final calculated cost of 0.8 cents/base is significantly cheaper than other manufacturing costs of 7 - 30 cents/base. Reducing the cost of gene synthesis provides new insight into the cost-effectiveness of present technologies and protocols and has the potential to benefit the fields of bioengineering and gene therapy.

In recent years, stem cells have been a focal point in research designed to evaluate the efficacy of ophthalmologic therapies, specifically those for corneal conditions. The corneal epithelium is one of the few regions of the body that maintains itself using a residual stem cell population within the adjacent limbus. Stem cell movement has additionally captivated the minds of researchers due to its potential application in different body regions. The cornea is a viable model for varying methods to track stem cell migratory patterns, such as lineage tracing and live imaging from the limbus. These developments have the potential to pave the way for future therapies designed to ensure the continuous regeneration of the corneal epithelium following injury via the limbal stem cell niche. This literature review aims to analyze the various methods of imaging used to understand the limbal stem cell niche and possible future directions that might be useful to consider for the better treatment and prevention of disorders of the cornea and corneal epithelium. .

Solid-state atomic hydrogen as a broad-spectrum RONS scavenger for accelerated diabetic wound healing

Hydrogen therapy shows great promise as a versatile treatment method for diseases associated with the overexpression of reactive oxygen and nitrogen species（RONS）. However, developing an advanced hydrogen therapy platform that integrates controllable hydrogen release, efficient RONS elimination, and biodegradability remains a giant technical challenge. In this study, we demonstrate for the first time that the tungsten bronze phase H0.53WO3（HWO） is an exceptionally ideal hydrogen carrier, with salient features including temperature-dependent highly-reductive atomic hydrogen release and broad-spectrum RONS scavenging capability distinct from that of molecular hydrogen. Moreover, its unique p H-responsive biodegradability ensures post-therapeutic clearance at pathological sites. Treatment with HWO of diabetic wounds in an animal model indicates that the solid-state atomic H promotes vascular formation by activating M2-type macrophage polarization and anti-inflammatory cytokine production, resulting in acceleration of chronic wound healing. Our findings significantly expand the basic categories of hydrogen therapeutic materials and pave the way for investigating more physical forms of hydrogen species as efficient RONS scavengers for clinical disease treatment.

The Influential Mechanisms of Theory of Mind on Prosocial Behavior and the Effect of Mindfulness Intervention

Background Theory of Mind (ToM) and empathy are crucial cognitive and emotional capacities that influence social interactions. While their role in promoting prosocial behavior has been established, the potential moderating effect of mindfulness on this relationship remains unexplored. Understanding these complex interactions is vital for developing effective interventions to foster prosocial behavior among college students. This study examines the influence of ToM on college students’ prosocial behavior and explores the moderating role of mindfulness in this relationship. Methods A mixed-methods approach combining questionnaires and experimental design was employed. Study 1: A survey of 759 college students (mean age 22.03 years; 477 females) was conducted using the ToM Scale, Interpersonal Reactivity Index, Prosocial Behavior Tendency Scale, and Mindfulness Awareness Scale. Data were analyzed using correlation and moderated mediation analyses. Study 2: An 8-week mindfulness attention training program was implemented for the intervention group and compared with a control group. Mindfulness training served as a moderating variable to validate Model 59 from Study 1. Results 1. Study 1 found: (a) ToM was significantly positively correlated with prosocial behavior (r = 0.31, p 1). (b) Empathy partially mediated the relationship between ToM and prosocial behavior (β = 0.10, p 1, 95% CI [0.06, 0.14]). (c) Mindfulness negatively moderated the direct path between ToM and three dimensions of prosocial behavior, as well as the indirect path between empathy and kin altruism and reciprocal altruism. Specifically, high levels of mindfulness weakened the direct impact of ToM on prosocial behavior. high levels of mindfulness also weakened the indirect influence of ToM on prosocial behavior through empathy. 2. First-principles calculations were conducted to investigate the structural,electronic,and magnetic properties of single Fe atoms and Fe dimers on Cu_(2)N/Cu(100).Upon adsorption of an Fe atom onto Cu_(2)N/Cu(100),robust Fe-N bonds form,resulting in the incorporation of both single Fe atoms and Fe dimers within the surface Cu_(2)N *** partial occupancy of Fe-3d orbitals lead to large spin moments on the Fe ***,both single Fe atoms and Fe dimers exhibit in-plane magnetic anisotropy,with the magnetic anisotropy energy(MAE)of an Fe dimer exceeding twice that of a single Fe *** magnetic anisotropy can be attributed to the predominant contribution of the component along the x direction of the spin-orbital coupling ***,the formation of Fe-Cu dimers may further boost the magnetic anisotropy,as the energy levels of the Fe-3d orbitals are remarkably influenced by the presence of Cu *** study manifests the significance of uncovering the origin of magnetic anisotropy in engineering the magnetic properties of magnetic nanostructures.
