sRNA113 regulates Pseudomonas plecoglossicida motility to affect immune response against infection in pearl gentian grouper

Small RNAs(sRNAs) are a class of molecules capable of perceiving environmental changes and exerting posttranscriptional regulation over target gene expression,thereby influencing bacterial virulence and host immune responses. Pseudomonas plecoglossicida is a pathogenic bacterium that poses a significant threat to aquatic animal health. However, the regulatory mechanisms of sRNAs in P. plecoglossicida remain unclear. This study focused on sRNA113, previously identified as a potential regulator of the fliP gene, a key component of the lateral flagellar type III secretion system. To investigate the effects of sRNA113on P. plecoglossicida virulence, as well as its role in regulating pathogenic processes and host immune responses, mutant strains lacking this sRNA were generated and analyzed. Deletion of sRNA113 resulted in the up-regulation of lateral flagellar type III secretion system-related genes in P. plecoglossicida, which enhanced bacterial swarming motility, biofilm formation,and chemotaxis ability in vitro. In vivo infection experiments with pearl gentian grouper revealed that sRNA113 deletion enhanced the pathogenicity of ***. This heightened virulence was attributed to the up-regulation of genes associated with the lateral flagellar type III secretion system, resulting in higher bacterial loads within host tissues. This amplification of pathogenic activity intensified tissue damage, disrupted immune responses, and impaired the ability of the host to clear infection, ultimately leading to mortality. These findings underscore the critical role of sRNA113 in regulating the virulence of P. plecoglossicida and its interaction with host immune defenses. This study provides a foundation for further exploration of sRNAmediated mechanisms in bacterial pathogenesis and hostpathogen interactions, contributing to a deeper understanding of virulence regulation and immune evasion in aquatic pathogens.

Effect of preprocessing on performances of machine learning-based mineral composition analysis on gas hydrate sediments, Ulleung Basin, East sea

Gas hydrate(GH) is an unconventional resource estimated at 1000-120,000 trillion *** on GH is ongoing to determine its geological and flow characteristics for commercial production. After two large-scale drilling expeditions to study the GH-bearing zone in the Ulleung Basin, the mineral composition of 488 sediment samples was analyzed using X-ray diffraction(XRD). Because the analysis is costly and dependent on experts, a machine learning model was developed to predict the mineral composition using XRD intensity profiles as input data. However, the model's performance was limited because of improper preprocessing of the intensity profile. Because preprocessing was applied to each feature, the intensity trend was not preserved even though this factor is the most important when analyzing mineral composition. In this study, the profile was preprocessed for each sample using minmax scaling because relative intensity is critical for mineral analysis. For 49 test data among the 488data, the convolutional neural network(CNN) model improved the average absolute error and coefficient of determination by 41% and 46%, respectively, than those of CNN model with feature-based preprocessing. This study confirms that combining preprocessing for each sample with CNN is the most efficient approach for analyzing XRD data. The developed model can be used for the compositional analysis of sediment samples from the Ulleung Basin and the Korea Plateau. In addition, the overall procedure can be applied to any XRD data of sediments worldwide.

Genetic parameters and response to selection for body weight in turbot(Scophthalmus maximus, Linnaeus)

Genetic parameters and response to selection were estimated for harvest body weight in turbot. The data consisted of 10 952 individuals of 508 full-sib families from three generations（G0, G1, and G2）. The heritability estimates for G0, G1, and G2 were 0.11±0.08, 0.18±0.09, and 0.17±0.07, respectively. Over three generations, the heritability estimate was 0.19±0.04. Maternal and common environmental effects were 0.10±0.04, 0.14±0.04, and0.13±0.03 within each generation and 0.12±0.01 across generations. The selection differential in growth was 18.24 g in G0 and 21.19 g in G1 corresponding to an average of 19.72 g per generation. The genetic gains were also calculated, they were 22.06 g in G1 and 11.93 g in G2, corresponding to 6.36% and 3.52% body weight. The total genetic gain after two generations was 10.10% body weight, which indicated that the selective breeding program for the body weight trait in turbot was successful.

Interaction of temperature and salinity on the expression of immunity factors in different tissues of juvenile turbot Scophthalmus maximus based on response surface methodology

Central Composite Design(CCD) and response surface methodology were used in the experiment to examine the combined effect of temperature(16-28℃) and salinity(18-42) on Hsp70 and IgM genes expression levels in turbot(Scophthalmus maximus) liver and *** results showed that the coefficients of determination(R^2=0.965 2 for liver Hsp70,0.972 9 for kidney Hsp70,0.921 for liver IgM and 0.962 1 for kidney IgM) and probability values(P0.05),while the interactive effect between temperature and salinity on kidney IgM was significant(P<0.01).The model equation could be used in practice for forecasting Hsp70 and IgM genes expression levels in the liver and kidney of juvenile turbot via applying statistical optimization of the response of interest,at which the maximum liver Hsp70,kidney Hsp70,liver IgM and kidney IgM of1.48,1.49,2.48,and 1.38,respectively,were *** present model may be valuable in assessing the feasibility of turbot farming at different geographic locations and,furthermore,could be a useful reference for scientists studying the immunity of turbot.

The applicability of the micellar solubilization spectrophotometry is limited by its own inferior selectivity. The PAN is one of the excellent organic analytical reagents. The micellar solubilization system of metal-PAN-Triton X-100 has been studied for many years. The spectrophotometric scrutiny of the Ni-PAN-Triton X-100 system has been reported by Qi Wen-bin. But, the selectivity problem has not been solved.

Isolation and characterization of 45 polymorphic microsatellite loci of turbot(Scophthalmus maximus)and cross-species amplification

Turbot （Scophthalmus maximus） is a flatfish species commercially important for aquaculture. In this study, we generated a microsatellite-enriched genomic DNA library for Scophthalmus rnaxirnus, and then isolated and characterized 45 microsateIIite loci by genotyping 30 individuals. The observed number of alleles ranged from 2 to 19 with an average of 6.24, while the effective number of alleles ranged from 1.30 to 11.11 with an average of 3.66. The expected heterozygosities varied from 0.235 to 0.9254 and polymorphic information content ranged from 0.204 4 to 0.903 3, with an average of 0.622. Twelve loci deviated significantly from Hardy-Weinberg equilibrium, and no significant linkage disequilibrium was observed between any pair of loci after Bonferroni correction. In cross-species amplification, five flatfish species （Paralichthys lethostigma, Verasper rnoseri, platichthys stellatus, Hippoglossoides dubius and Cynoglossus semilaevis） showed at least one polymorphic locus. These polymorphic microsatellite loci should prove useful for population analysis of turbot and other related species.

Pharmacokinetics and tissue behavior of enrofloxacin and its metabolite ciprofloxacin in turbot Scophthalmus maximus at two water temperatures

Turbot Scophthalmus maximus, an important aquaculture species in China, currently suffers from epizootic diseases because of high density aquaculture. Enrofloxacin has been used to treat various systemic bacterial fish infections. However, studies concerning the pharmacokinetics of enrofloxacin in turbot are limited. In this study, the pharmacokinetics of enrofloxacin and its metabolite ciprofloxacin, were investigated in the turbot following intravenous and oral administration at 10 mg enrofloxacin/kg body weight, at 16℃ and 10℃ water temperatures. The concentrations of enrofloxacin and ciprofloxacin in the main tissues （plasma, muscle, liver and kidney） were detected by HPLC. The results show that the plasma concentration-time data for enrofloxacin were best described as a two-compartment open model after intravenous and oral administration. Three pharmacokinetic equations were established between the concentrations and temperatures. The kinetic profile of enrofloxacin was temperature dependent. The absorption half-life of enrofloxacin was 1.99 h and 2.17 h after oral administration, whereas the elimination half-life of the drug was 98.63 h and 136.59 h at 16℃ and 10℃, respectively. The peak concentration of enrofloxacin in plasma and tissues was higher at 16℃ than that at 10℃, and the peak plasma concentration time in the liver was the shortest at both temperatures among those of other tissues. The plasma ℃/MIC ratio varied between 11.08 and 5 540.00 at 16℃; and between 7.92 and 3 960.00 at 10℃. The AUC/MIC ratio was 467.82-280 690.00 at 16℃, and 359.48-215 690.00 at 10℃. These ratios indicate that it is possible to obtain therapeutic efficacy. Very low levels of ciprofloxacin were detected. The AUC ratios of ciprofloxacin and enrofloxacin in plasma suggest that plasma ciprofloxacin might play a minor role in enrofloxacin treatment for turbot.

Optimization of Protectant, Salinity and Freezing Condition for Freeze-Drying Preservation of Edwardsiella tarda

Novel preservation condition without ultra-low temperature is needed for the study of pathogen in marine fishes. Freeze-drying is such a method usually used for preservation of terrigenous bacteria. However, studies using freeze-drying method to preserving marine microorganisms remain very limited. In this study, we optimized the composition of protectants during the freeze-drying of Edwardsiella tarda, a fish pathogen that causes systemic infection in marine fishes. We found that the optimal composition of protectant mixture contained trehalose(8.0%), skim milk(12.0%), sodium citrate(2.0%), serum(12.0%) and PVP(2.0%). Orthogonal and interaction analyses demonstrated the interaction between serum and skim milk or sodium citrate. The highest survival rate of E. tarda was observed when the concentration of Na Cl was 10.0, 30.0 and between 5.0 and 10.0 g L^(-1) for preparing TSB medium, E. tarda suspension and protectant mixture, respectively. When E. tarda was frozen at-80℃ or-40℃ for 6 h, its survival rate was higher than that under other tested conditions. Under the optimized conditions, when the protectant mixture was used during freeze-drying process, the survival rate(79.63%–82.30%) of E. tarda was significantly higher than that obtained using single protectant. Scanning electron microscopy(SEM) image indicated that E. tarda was embedded in thick matrix with detectable aggregation. In sum, the protectant mixture may be used as a novel cryoprotective additive for E. tarda.

Electroluminescence from the InGaN/GaN Superlattices Interlayer of yellow LEDs with Large V-Pits Grown on Si(111)

A blue emission originated from In GaN/GaN superlattice（SL） interlayer is observed in the yellow LEDs with V-pits embedded in the quantum wells（QWs）, revealing that sufficient holes have penetrated through the QWs into SLs far away from the p-type layer. In the V-pits embedded LEDs, hole transport has two paths： via the flat c-plane region or via the sidewalls of V-pits. It is proved that the holes in SLs are injected from the sidewalls of V-pits, and the transportation process is significantly affected by working temperature, current density, and the size of V-pits. Four motion possibilities are discussed when the holes flow via the sidewalls. All these may contribute to a better understanding of hole transport and device design.

This study presents the complete mitochondrial genome of the hybrid Epinephelus moara♀× Epinephelus lanceolatus♂. The genome is 16886 bp in length, and contains 13 protein-coding genes, 2 r RNA genes, 22 t RNA genes, a light-strand replication origin and a control region. Additionally, phylogenetic analysis based on the nucleotide sequences of 13 conserved protein-coding genes using the maximum likelihood method indicated that the mitochondrial genome is maternally inherited. This study presents genomic data for studying phylogenetic relationships and breeding of hybrid Epinephelinae.
