The phycocyanin-chlorophyll-protein complexes isolated from Chroomonas placoidea

An active photosystem(PS)Ⅱparticle and two light-harvesting complexes,as well as their subcomplexes that have not been reported previously,were isolated from a cryptophyte Chroomonas placoidea by Triton X-100 sucrose density gradient *** fluorescence spectra revealed that there were efficient energy couplings between phycocyanin(PC645)and chlorophyll(Chl)within both zonesⅢandⅣof the gradient,which were designated respectively as light-harvesting complex and PSⅡparticles whose size was 15-20 nm according to negative staining in electron *** the two complexes were further resolved into sub-complexes,the energy coupling was retained in the core PSⅡcomplex(named as zoneⅣ-2 of the sucrose gradient),which contained almost no outer antenna pigment Chl *** dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)showed that the PC645 components appeared in Chl-containing protein complexes were mainly the β subunit with molecular weight of 20 *** results demonstrate that PC645 in this cryptophyte was structurally but preferentially combined with the light-harvesting complex and PSⅡ*** excitation energy absorbed by PC645 could be directly transferred to Chl a(especially the long wavelength of Chl a)in the PSⅡreaction center or via the Chl a/c-protein *** β subunit corresponded to the terminal fl uorescence emission and might play an important role in transmitting energy from PC645 to the Chl-protein *** results will help in elucidating the architecture and function of the energy transfer system comprising phycobiliproteins and Chl-protein complexes in cryptophytes.

Typical adaptive neural control for hypersonic vehicle based on higher-order filters

A typical adaptive neural control methodology is used for the rigid body model of the hypersonic vehicle. The rigid body model is divided into the altitude subsystem and the velocity subsystem. The proportional integral differential(PID) controller is introduced to control the velocity track. The backstepping design is applied for constructing the controllers for the altitude *** avoid the explosion of differentiation from backstepping, the higher-order filter dynamic is used for replacing the virtual controller in the backstepping design steps. In the design procedure,the radial basis function(RBF) neural network is investigated to approximate the unknown nonlinear functions in the system dynamic of the hypersonic vehicle. The simulations show the effectiveness of the design method.

Finite Element Prediction of Residual Stress in Rhombic Dodecahedron Ti‑6Al‑4V Titanium Alloy Additively Manufactured by Electron Beam Melting

In this work,a three-dimensional nonlinear transient thermo-mechanically coupled finite element model(FEM)is established to investigate the variation in temperature and stress fields during electron beam melting(EBM)of rhombic dodecahedron Ti-6Al-4V *** influence of the processing parameters on the temperature and residual stress evolutions was predicted and verified against existing literature *** calculated results indicate that the interlayer cooling time has very little effect on both the temperature and stress evolutions,indicating that the interlayer cooling time can be set up as short as possible to reduce manufacturing *** is presented that the residual stress of the intersection is higher than that of *** increasing preheating temperature,the residual stress decreases continuously,which is about 20%–30%for every 50℃rise in *** temperature and stress fields repeated every four layers with the complex periodic scanning *** x and y-component residual stresses are tensile stresses,while z-component stress is weak compressive or tensile stress in typical *** is proposed that the interlayer cooling is necessary to obtain a rhombic dodecahedron with low residual *** results can bring insights into the understanding of the residual stress during EBM.

Investigating the Impact of Ocean Acidification on Anti-Stress Mechanisms in Sepia esculenta Larvae Based on Transcriptome Profiling

With the rapid development of oil,energy,power and other industries,CO_(2) emissions rise sharply,which will cause a large amount of CO_(2) in the air be absorbed by the ocean and lead to ocean *** growth and development of organisms can be seriously affected by acidified *** esculenta is a mollusk with high nutritional and economic value and is widely cultured in offshore waters of *** are the early life forms of the organism and are more vulnerable to changes in the external *** low pH will lead to some adverse reactions in larvae,which will affect metabolism,immune response and other life *** this study,we sequenced the transcriptome of *** subjected to acidified seawater stress and identified 1072differentially expressed genes(DEGs).The detected atypical expression of DEGs substantiates cellular malformation and translocation in *** under low pH ***,this also substantiates the notable impact of ocean acidification on *** DEGs were used for functional enrichment analysis of GO and KEGG,and the top twenty items of the biological process classification in GO terms and 11 KEGG signaling pathways were significantly ***,the constructed proteinprotein interaction network(PPI)was used to analyze protein-protein interactions,and 12 key DEGs and 3 hub genes were *** reliability of 12 genes was verified by quantitative RT-PCR.A comprehensive analysis of the KEGG signaling pathway and PPI revealed that ocean acidification leads to abnormalities in lipid metabolism in *** larvae,which can lead to cancer development and metastasis,accompanied by some degree of *** results of the study will help to further investigate the physiological processes of *** when stimulated by ocean acidification,and provide a reference to cope with the captive breeding of *** affected by acidification.

High-throughput sequencing of 16S r RNA amplicons characterizes gut microbiota shift of juvenile sea cucumber Apostichopus japonicus feeding with three antibiotics

Sea cucumber Apostichopus japonicus is an important marine economic species in Asian countries due to its profound nutritional and medicinal value. So far, with the rapid development of intensifi ed artifi cial aquaculture of sea cucumbers, the use of antibiotics is still an inexpensive and dispensable way to treat pathogenic infections, especially during the nursery phase. However, there is little information on the eff ects of antibiotics on the intestinal microbiota of sea cucumber. Therefore an Illumina based sequencing method was used to examine the intestinal bacterial composition of juvenile A . japonicas following diets with three typical antibiotics (tetracycline, erythromycin, and norfl oxacin) under 15, 30, and 45 d. The fi ndings reveal that diff erent antibiotics have distinct eff ects on the growth performance of juvenile sea cucumbers. However, the richness and diversity of microbiota were barely aff ected by antibiotics but the community composition alterations indicated that the three antibiotics exhibited their respective patterns of reshaping the intestinal bacteria of juvenile sea cucumbers. In common, the abundance of some sensitive genera with helpful functions, such as Thalassotalea , Shewanella , Sulfi tobacter , and Halomonas decreased signifi cantly with exposure to antibiotics and the abundance of multiple potential pathogenic- and suspected antibiotic-resistant microorganisms like Arcobacter , Leucothrix , and Clostridium_sensu_stricto_1 was found increased signifi cantly in the antibiotic groups. These results suggest that low doses of antibiotics could aff ect the composition of the intestinal microbiota of sea cucumbers and might increase the risk of infection of the hosts. This study could help us to explore how antibacterial compounds modify the gut microbiota of sea cucumbers and provide theoretical guidance in hatchery management by scientifi c antibiotic use in sea cucumber mariculture.

In this work, the amination of sulfonated polystyrene resin with alkyl secondary amine is investigated. The catalytic activities of the modified resins are determined through the hydration of l-butene. The optimum chain length and the best range of amination rate are determined. It is found that the single-pass conversion of 1 -butene was raised 2% on average, and the relative activity was increased over 30% after modification. A hypothesis about the enhancement of catalytic activities by the inclusion of alkyl chain to wrap up the butene molecule is proposed.

Hydrogen Storage Capacity Study of a Li+Graphene Composite System with Different Charge States

Based on first-principles plane-wave calculations,we firstly reconfirm that the Li+graphene complex can be taken as a hydrogen storage medium with capacity of 12.8wt%.Then metal adsorption properties of this Li+graphene system with different charge states are ***,the hydrogen storage ability of the charging system is *** calculations show that adding positive charge on a Li+graphene composite results in a conspicuous reduction of Li2𝑡and Li2𝑞orbital occupation with respect to the C2𝑞*** a result,a stronger bonding between Li and graphene is formed,and a special double-layer hydrogen adsorption structure has been *** to the neutral system,utilizing the positive charged Li+graphene to store hydrogen molecules can solve the issue of clustering of metal atoms after releasing hydrogen,and can improve hydrogen storage capacity up to a gravimetric density of 20.4wt%,correspondingly one adsorbed Li atom can effectively absorb up to seven H 2 molecules.

Effect of Lanthanum on Expression of Recombinant Allophycocyanin Gene in Pichia Pastoris

The enhancing effect of lanthanum on gene expression of recombinant allophycocyanin （rAPC）, a potential antitumor medicine, in Pichia pastoris was studied. PCR and sequence analysis were used in order to prove whether the APC gene had integrated into the yeast genome. The expression level of the recombinant allophycocyanin （rAPC） in BMMY medium containing LaCl3 was detected by ELISA method. The recombinant allophycocyanin was determined by Western blot. The results show that the recombinant Pichia pastoris chromosome contained allophycocyanin gene. Expression efficiency of rAPC gene in Pichia pastoris was promoted by proper LaCl3 concentration like 2, 5, 10 mmol·L-1, among which 5 (mmol·L-1) was the most effective. The highest expression yield of rAPC in the BMMY medium containing 5 mmol·L-1 LaCl3 was 44 mg·L-1 at 48 h, that was increased by 110% compared with 2.1 mg·L-1 of control, in the meantime, the optimum culture time is shortened from 72 to 48 h. The result of western blot analysis indicates that the rAPC consisted of two kinds of subunits with molecular weight of 19 and 21 kDa respectively.
