BACKGROUND Prior studies have noted great variability in the plasma levels of risperidone(RIS).Plasma concentrations of RIS and its active moiety are highly variable and depend on absorption,metabolism,and other predictors of metabolic dysregulation;however,these factors are poorly understood and the association between metabolic change and change in psychopathology is *** To ascertain the characteristics of chronic schizophrenic patients treated with RIS,and to assess their relationship with plasma RIS *** This was a descriptive cross-sectional study of 50 patients with a diagnosis of schizophrenic psychosis treated with RIS in a psychiatric *** plasma concentrations of RIS and its metabolite 9-hydroxyrisperidone were determined by high performance liquid *** patients’demographic and clinical characteristics,and psychopathologies were assessed,and the associations between clinical variables and plasma levels of RIS were *** Male patients received higher doses of RIS than female ones,but plasma concentrations of RIS and risperidone+9-hydroxyrisperidone(active moiety)were higher in female *** and the mean scores of the general psychopathology subscale of the Positive and Negative Syndrome Scale(PANSS)were significantly positively correlated with plasma concentrations of risperidone+9-hydroxyrisperidone adjusted for weight and dose in all 50 *** male subjects,we found a statistically significant positive correlation between the concentrations of risperidone+9-hydroxyrisperidone in plasma/(dose×kg)and age,mean PANSS negative subscale scores,mean PANSS general psychopathology subscale scores,and mean PANSS total *** Long-term use of RIS should be closely monitored in older patients and females to minimize the risk of high concentrations which could induce side effects.

Higher levels of the lipophilic antioxidants coenzyme Q_(10) and vitamin E in long-lived termite queens than in short-lived workers

Pyrimethamine upregulates BNIP3 to interfere SNARE-mediated autophagosome-lysosomal fusion in hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is one of the most common tumor types and remains a major clinical challenge. Increasing evidence has revealed that mitophagy inhibitors can enhance the effect of chemotherapy on HCC. However, few mitophagy inhibitors have been approved for clinical use in humans. Pyrimethamine (Pyr) is used to treat infections caused by protozoan parasites. Recent studies have reported that Pyr may be beneficial in the treatment of various tumors. However, its mechanism of action is still not clearly defined. Here, we found that blocking mitophagy sensitized cells to Pyr-induced apoptosis. Mechanistically, Pyr potently induced the accumulation of autophagosomes by inhibiting autophagosome-lysosome fusion in human HCC cells. In vitro and in vivo studies revealed that Pyr blocked autophagosome-lysosome fusion by upregulating BNIP3 to inhibit synaptosomal-associated protein 29 (SNAP29)-vesicle-associated membrane protein 8 (VAMP8) interaction. Moreover, Pyr acted synergistically with sorafenib (Sora) to induce apoptosis and inhibit HCC proliferation in vitro and in vivo. Pyr enhances the sensitivity of HCC cells to Sora, a common chemotherapeutic, by inhibiting mitophagy. Thus, these results provide new insights into the mechanism of action of Pyr and imply that Pyr could potentially be further developed as a novel mitophagy inhibitor. Notably, Pyr and Sora combination therapy could be a promising treatment for malignant HCC.

Several autonomous navigation methods have been studied to reduce reliance on the ground stations. Geomagnetic navigation, gravity gradient navigation, and celestial navigation using optical sensors are typical autonomous navigation methods. Geomagnetic and gravity gradient navigation have low navigation accuracy (1301 m [1] and 235 m [2], respectively) due to the poor accuracy of the geomagnetic and gravity gradient maps.

Named Entity Recognition(NER)stands as a fundamental task within the field of biomedical text mining,aiming to extract specific types of entities such as genes,proteins,and diseases from complex biomedical texts and categorize them into predefined entity *** process can provide basic support for the automatic construction of knowledge *** contrast to general texts,biomedical texts frequently contain numerous nested entities and local dependencies among these entities,presenting significant challenges to prevailing NER *** address these issues,we propose a novel Chinese nested biomedical NER model based on RoBERTa and Global Pointer(RoBGP).Our model initially utilizes the RoBERTa-wwm-ext-large pretrained language model to dynamically generate word-level initial *** then incorporates a Bidirectional Long Short-Term Memory network for capturing bidirectional semantic information,effectively addressing the issue of long-distance ***,the Global Pointer model is employed to comprehensively recognize all nested entities in the *** conduct extensive experiments on the Chinese medical dataset CMeEE and the results demonstrate the superior performance of RoBGP over several baseline *** research confirms the effectiveness of RoBGP in Chinese biomedical NER,providing reliable technical support for biomedical information extraction and knowledge base construction.

The Pearl River Delta(PRD),a tornado hotspot,forms a distinct trumpet-shaped coastline that concaves toward the South China *** the summer monsoon season,low-level southwesterlies over the PRD’s sea surface tend to be turned toward the west coast,constituting a convergent wind field along with the landward-side southwesterlies,which influences regional convective *** two-part study explores the roles of this unique land–sea contrast of the trumpet-shaped coastline in the formation of a tornadic mesovortex within monsoonal flows in this *** I primarily presents observational analyses of pre-storm environments and storm *** rotating storm developed in a lowshear environment(not ideal for a supercell)under the interactions of three air masses under the influence of the land–sea contrast,monsoon,and storm cold *** intersection zone(or“triple point”)is typically characterized by local enhancements of ambient vertical vorticity and *** on a rapid-scan X-band phased-array radar,finger-like echoes were recognized shortly after the gust front intruded on the triple *** over the triple point,they rapidly wrapped up with a well-defined low-level *** is thus presumed that the triple point may have played roles in the mesovortex genesis,which will be demonstrated in Part II with multiple sensitivity numerical *** findings also suggest that when storms pass over the boundary intersection zone in the PRD,the expected possibility of a rotating storm occurring is relatively high,even in a low-shear *** knowledge of such environments provides additional guidance to assess the regional tornado risk.

To the Editor:Assessment of data appropriateness is a process to answer whether electronic health records(EHRs)from routine healthcare practices couldt intended study purposes,for example,available to be proceeded,with enough individual records,with relevant information able to be extracted from records,etc.[1-3]This has been increasingly underscored as a prerequisite when using EHRs(one important type of real-world data[RWD])for scientic purposes.[4-6]Although controversies remain on the denitions,types and assessment methods of the dimensions of RWD appropriateness,the latest ofcial guidelines(i.e.,from the *** and Drug administration[FDA][4]and China National Medical Products administration[NMPA][6])suggest that the assessment start from a preliminary stage on variable existence,and subsequently deepen into the issue of data value(such as missing value,outliers,etc.),as the preliminary assessment is the foundation of the overall RWD appropriateness.[7]Little is known about the appropriateness of EHRs in developing regions,which feature high visit volumes and a great number of ***,this study aimed to preliminarily investigate RWD appropriateness from hospitals in developing regions from the perspective of core variables’comparison.

Spatial and Temporal Distribution Characteristics of Solar Energy Resources in Tibet

The Tibet Plateau is one of the regions with the richest solar energy resources in the *** the process of achieving carbon neutrality in China,the development and utilization of solar energy resources in the region will play an important *** this study,the gridded solar resource data with 1km resolution in Tibet were obtained by spatial correction and downscaling of SMARTS *** this basis,the spatial and temporal distribution characteristics of solar energy resources in the region in the past 30 years(1991–2020)are finely evaluated,and the annual global horizontal radiation resource is *** results show that:1)The average annual global horizontal radiation amount in Tibet is 1816 kWh/m^(2).More than 60%of the area belongs to the“Most abundant”(GHI≥1750 kWh/m^(2))area of China’s solar energy resources category A,and nearly 40%belongs to the“Quite abundant”(1400≤GHI<1750)area of China’s solar energy resource category B.2)In space,the solar energy resources in Tibet increased gradually from north to south and from east to ***,Central and Eastern Shigatse,Shannan,and Southwestern Ali are the most abundant cities,with a maximum annual radiation level of 2189 kWh/m2.3)In terms of time,the total horizontal radiation in Tibet was the highest in May and the lowest in December.74%of the total area belongs to the“Very stable”(R_(w)≥0.47)area of solar resource stability category A,and 26%belongs to the“stable”(0.36≤R_(w)<0.47)area of solar resource stability category *** energy resources in the region show the characteristics of both strong and *** solar energy resources in the region have shown a fluctuating downward trend over the past 30 years,with an average decline of about 12.86(kWh/m2)per decade.4)In terms of solar radiation resources reaching the earth’s surface,the theoretical total amount of annual horizontal radiation in Tibet is about 240.07 billion tons of standard coal or 222.91 billion kilowatts on average.

Spotlight on eltrombopag concentration in pediatric immune thrombocytopenia: A single-center observational study in China

Importance: Eltrombopag has been recommended for pediatric immune thrombocytopenia (ITP). Response and adverse drug reactions (ADRs) varied widely between individuals, even at the same dose of eltrombopag. The appropriate eltrombopag concentration in ITP has not been ***: This study aims to explore the appropriate eltrombopag concentration in pediatric ***: This was a single-center, prospective cohort study. Children diagnosed with refractory persistent/chronic ITP and platelet count < 30×109/L were treated with eltrombopag and followed up for at least 2 months. Concentration was detected by high-performance liquid chromatography-mass spectrometry at least 2 weeks after eltrombopag. The clinical characteristics-concentration, concentration-response, and concentration-ADRs were ***: A total of 30 patients were enrolled, comprising 13 males and 17 females, with a median age of 72 (45-94) months. The median dose and concentration were 1.39 (1.09-1.56) mg/kg and 2.70 (2.25-4.13) mg/L, respectively. Of the enrolled patients, 14 responded to treatment, whereas 16 did not. Additionally, five experienced adverse drug reactions. No linear correlation was observed between eltrombopag concentration and clinical characteristics. The concentration was lower in the response group than in the nonresponse group, but there was no significant difference (t = 0.755,P = 0.457). Patients who experienced ADRs had a higher concentration than those without ADRs (t = 2.538,P = 0.017). The area under the receiver operating characteristic curve of ADRs was 0.78 (95% confidence interval: 0.56-1.00). Youden’s index identified the cutoff point as 4.33 mg/L, with a sensitivity of 88% and a specificity of 60%. Logistic regression analysis demonstrated that a higher platelet count before eltrombopag predicted a favorable ***: Eltrombopag proves efficacious and well-tolerated for treating pediatric ITP.
