Maintaining moderate levels of hypochlorous acid promotes neural stem cell proliferation and differentiation in the recovery phase of stroke

It has been shown clinically that continuous removal of ischemia/reperfusion-induced reactive oxygen species is not conducive to the recovery of late ***,previous studies have shown that excessive increases in hypochlorous acid after stroke can cause severe damage to brain *** previous studies have found that a small amount of hypochlorous acid still exists in the later stage of stroke,but its specific role and mechanism are currently *** simulate stroke in vivo,a middle cerebral artery occlusion rat model was established,with an oxygen-glucose deprivation/reoxygenation model established in vitro to mimic *** found that in the early stage(within 24 hours)of ischemic stroke,neutrophils produced a large amount of hypochlorous acid,while in the recovery phase(10 days after stroke),microglia were activated and produced a small amount of hypochlorous ***,in acute stroke in rats,hypochlorous acid production was prevented using a hypochlorous acid scavenger,taurine,or myeloperoxidase inhibitor,4-aminobenzoic acid *** results showed that high levels of hypochlorous acid(200μM)induced neuronal apoptosis after oxygen/glucose deprivation/***,in the recovery phase of the middle cerebral artery occlusion model,a moderate level of hypochlorous acid promoted the proliferation and differentiation of neural stem cells into neurons and *** suggests that hypochlorous acid plays different roles at different phases of cerebral ischemia/reperfusion *** levels of hypochlorous acid(5 and 100μM)promoted nuclear translocation ofβ-*** transfection of single-site mutation plasmids,we found that hypochlorous acid induced chlorination of theβ-catenin tyrosine 30 residue,which promoted nuclear ***,our study indicates that maintaining low levels of hypochlorous acid plays a key role in the recovery of neurological function.

Hydrogen Sulfide Promotes Root Organogenesis in Ipomoea batatas, Salix matsudana and Glycine max

In this report, we demonstrate that sodium hydrosulfide （NariS）, a hydrogen sulfide （H2S） donor, promoted adventitious root formation mediated by auxin and nitric oxide （NO）. Application of the HzS donor to seedling cuttings of sweet potato （Ipomoea batatas L.） promoted the number and length of adventitious roots in a dose-dependent manner. It was also verified that H2S or HS- rather than other sulfur-containing components derived from NariS could be attributed to the stimulation of adventitious root formation. A rapid increase in endogenous H2S, indole acetic acid （IAA） and NO were sequentially observed in shoot tips of sweet potato seedlings treated with HallS. Further investigation showed that H2S-mediated root formation was alleviated by N-1-naphthylphthalamic acid （NPA）, an IAA transport inhibitor, and 2-（4-carboxyphenyl）. 4,4,5,5-tetramethylimidazoline-1-oxyl-3-0xide （cPTIO）, an NO scavenger. Similar phenomena in H2S donor-dependent root organogenesis were observed in both excised willow （Salix matsudana var. tortuosa Vilm） shoots and soybean （Glycine max L.） seedlings. These results indicated that the process of H2S-induced adventitious root formation was likely mediated by IAA and NO, and that H2S acts upstream of IAA and NO signal transduction pathways.

Cylasformicarius is one of the most important pests of sweet potato worldwide, causing considerable ecological and economic *** study improved the effect of comprehensive management and understanding of genetic mechanisms by examining the functional genomics of C. *** Illumina and PacBio sequencing, this study obtained a chromosome-level genome assembly of adult weevils from lines inbred for 15 *** high-quality assembly obtained was 338.84 Mb, with contig and scaffold N50 values of 14.97 and 34.23 Mb, *** total, 157.51 Mb of repeat sequences and 11 907 protein-coding genes were predicted.A total of 337.06 Mb of genomic sequences was located on the 11 chromosomes, accounting for 99.03%of the total length of the associated *** genomic analysis showed that C. formicarius was sister to Dendroctonus ponderosae, and C. formicarius diverged from D. ponderosae approximately 138.89 million years ago (Mya).Many important gene families expanded in the C. formicarius genome were involved in the detoxification of pesticides, tolerance to cold stress and chemosensory *** further study the role of odorant-binding proteins (OBPs) in olfactory recognition of C. formicarius, the binding assay results indicated that Cfor OBP4–6 had strong binding affinities for sex pheromones and other *** high-quality C. formicarius genome provides a valuable resource to reveal the molecular ecological basis, genetic mechanism, and evolutionary process of major agricultural pests;it also offers new ideas and new technologies for ecologically sustainable pest control.

Ipomoea batatas HKT1 transporter homolog mediates K^+ and Na^+ uptake in Saccharomyces cerevisiae

Soil salinity causes the negative effects on the growth and yield of crops. In this study, two sweet potato （Ipomoea batatas L.） cultivars, Xushu 28 （X-28） and Okinawa 100 （O-100）, were examined under 50 and 100 mmol L-1 NaCI stress. X-28 cultivar is relatively high salt tolerant than O-100 cultivar. Interestingly, real-time quantitative polymerase chain reaction （RT-qPCR） results indicated that sweet potato high-affinity K^＋ transporter 1 （IbHKT1） gene expression was highly induced by 50 and 100 mmol L-1 NaCI stress in the stems of X-28 cultivar than in those of O-100 cultivar, but only slightly induced by these stresses in the leaves and fibrous roots in both cultivars. To characterize the function of IbHKT1 transporter, we performed ion-flux analysis in tobacco transient system and yeast complementation. Tobacco transient assay showed that IbHKT1 could uptake sodium （Na^＋）. Yeast complementation assay showed that IbHKT1 could take up K^＋ in 50 mmol L^-1 K^＋ medium without the presence of NaCI. Moreover, Na^＋ uptake significantly increased in yeast overexpressing IbHKTI. These results showed that IbHKT1 transporter could have K^＋-Na^＋ symport function in yeast. Therefore, the modes of action of IbHKT1 in transgenic yeast could differ from the mode of action of the other HKT1 transporters in class I. Potentially, IbHKT1 could be used to improve the salt tolerance nature in sweet potato.

Apolygus lucorum is the dominant pathogenic insect attacking Bacillus thuringiensis(Bt)cotton in ***,20-hydroxyecdysone(20E)has important functions in many biological processes,including insect *** C(PLC),which is an essential enzyme for phosphoinositide metabolism,is involved in 20E signal transduction,but its function in 20E-mediated reproduction in *** remains *** this study,20E increased A/PLCγ transcription as well as the abundance and activity of the encoded protein during molting and *** 20E treatment also induced the considerable accumulation of two second messengers,inositol triphosphate and *** expression levels of genes encoding vitellogenin(AlVg)and soluble trehalase(AlTre-1)were similar to those of AlPLCy,and were upregulated in response to *** silencing of AlPLCγ resulted in downregulated expression o f AITre-γ ***,the silencing of AlTre-1 and AlVg did not affect AlPLCγ ***,the silencing of AlVg did not alter AlTre-1 ***,an examination of the insect specimens indicated that AlPLCy is required for female adult reproduction,and that downregulated expression of this gene is associated with decreases in fecundity,adult longevity,and egg hatching rate as well as delayed oocyte *** propose that 20E regulates AlTre-1 expression via AlPLCy and affects Vg expression as well as ovary development to facilitate the reproductive activities of *** females.

Role of PI3-K/Akt pathway and its effect on glial cell line-derived neurotrophic factor in midbrain dopamine cells

Aim:To explore the intracellular mechanisms underlying the survival/differentia-tion effect of the glial cell line-derived neurotrophic factor(GDNF)on dopamine(DA)***:Midbrain slice culture and primary cell culture wereestablished,and the cultures were divided into 3 groups:control group,GDNFgroup,and the phosphatidylinositol 3-kinase/Akt(PI3-K/Akt)*** the expression of tyrosine hydroxylase(TH)was detected byimmunostaining as well as Western ***:GDNF treatment inducedan increase in the number of TH-immunoreactive(ir)cells and the neurite numberof TH-ir cells,as well as in the level of TH expression in cultures(Number of TH-ir cells in the slice culture:control group,8.76±0.75;GDNF group,18.63±*** of TH-ir cells and neurite number of TH-ir cells in cell culture:controlgroup,3.65±0.88 and 2.49±0.42;GDNF group,6.01±0.43 and 4.89±0.46).Meanwhile,the stimulation of cultured cells with GDNF increased the phosphorylation of Akt,which is a downstream effector of PI3-K/*** effects of GDNF were specifi-cally blocked by the inhibitor of the PI3-K/Akt pathway,wortmannin(Number ofTH-ir cells in slice culture:PI3-K/Akt pathway-inhibited group,6.98±***-ber of TH-ir cells and neurite number of TH-ir cells in cell culture:PI3-K/Aktpathway-inhibited group,3.79±0.62 and 2.50±0.25,respectively).Conclusion:The PI3-K/Akt pathway mediates the survival/differentiation effect of GDNF onDA cells.

Present Situation and Prospect of Grub Control in sweet potato Field

Grub is one of the major pests affecting yield and quality of sweet potato. Although traditional chemical pesticides have quick effect, the disadvantages of high toxicity, strong pest resistance, and heavy pollution hinder the development of modern ecological agriculture, while physical means such as light trapping, sex pheromone method and biological control methods are more safe and lasting compared with chemical means. To provide reference and guidance for grub control and sweet potato production, the control methods against sweet potato grubs are comprehensively explored, and strategy and prospects of using biological technology to control pests are analyzed.

Dissecting the genetic regulation of gene expression is critical for understanding phenotypic variation and species ***,our understanding of the transcriptional variability in sweet potato remains ***,we analyzed two publicly available datasets to explore the landscape of transcriptomic variations and its genetic basis in the storage roots of sweet *** comprehensive analysis identified a total of 724,438 high-confidence single nucleotide polymorphisms(SNPs)and 26,026 expressed *** quantitative trait locus(eQTL)analysis revealed 4408 eQTLs regulating the expression of 3646 genes,including 2261 local eQTLs and 2147 distant *** distant eQTL hotspots were found with target genes significantly enriched in specific functional *** combining the information from regulatory network analyses,eQTLs and association mapping,we found that IbMYB1-2 acts as a master regulator and is the major gene responsible for the activation of anthocyanin biosynthesis in the storage roots of sweet *** study provides the first insight into the genetic architecture of genome-wide expression variation in sweet potato and can be used to investigate the potential effects of genetic variants on key agronomic traits in sweet potato.

Pesticides Could Alleviate the Symptoms of sweet potato Virus Disease(SPVD)

In recent years, sweet potato virus disease （SPVD） has severely affected the production of sweet potato in China. In order to select chemical agents to prevent sweet potato from being infected with SPVD, 14 pesticides were sprayed on the plants of sweet potato infected with SPVD, and the relative mRNA level of the viruses in the leaves and physiological indicators of sweet potato plants were detected. The results showed that after the application of most of the pesticides, the relative mRNA level of the two viruses that caused SPVD decreased, and the chlorophyll content and biomass of sweet potato infected with SPVD increased significantly or extremely significantly. Among the pesticides, BASF virus liquid, 0.1% physcion and Aolike powder for cucumber virus were proved to be the best, as the chlorophyll content and stem length of sweet potato increased significantly. The experiment indicated that the pesticides could reduce the harm of SPVD to sweet potato.
