Neuroprotective and antidiabetic lanostane-type triterpenoids from the fruiting bodies of Ganoderma theaecolum

Eight previously undescribed lanostane triterpenoids,including five nortriterpenoids with 2 6carbons,ganothenoids A-E（1-5）,and three lanostanoids,ganothenoids F-H（6-8）,along with 24 known ones（9-32）,were isolated from the fruiting bodies of Ganodrma *** structures of the novel compounds were elucidated using comprehensive spectroscopic methods,including electronic circular dichroism（ECD） and nuclear magnetic resonance（NMR） *** 1-32 were assessed for their neuroprotective effects against H2O2-induced damage in human neuroblastoma SH-SY5Y cells,as well as their inhibitory activities against protein tyrosine phosphatase 1B（PTP1B） and α-*** 4demonstrated the most potent neuroprotective activity against H2O2-induced oxidative stress by suppressing G0/G1phase cell cycle arrest,reducing reactive oxygen species（ROS） levels,and inhibiting cell apoptosis through modulation of B-cell lymphoma 2 protein（Bcl-2） and Bcl-2 associated X-protein（Bax） protein *** 26,12,and 28 exhibited PTP1B inhibitory activities with IC50values ranging from 13.92 to 56.94 μmol·L-1,while compound12 alone displayed significant inhibitory effects on α-glucosidase with an IC50value of 43.56μmol·***,enzyme kinetic analyses and molecular docking simulations were conducted for compounds 26 and 12 with PTP1B and α-glucosidase,respectively.

Osteoporosis is a prevalent skeletal condition characterized by reduced bone mass and strength, leading to increased fragility. Buqi-Tongluo(BQTL) decoction, a traditional Chinese medicine(TCM) prescription, has yet to be fully evaluated for its potential in treating bone diseases such as osteoporosis. To investigate the mechanism by which BQTL decoction inhibits osteoclast differentiation in vitro and validate these findings through in vivo *** employed MTS assays to assess the potential proliferative or toxic effects of BQTL on bone marrow macrophages(BMMs) at various concentrations. TRAc P experiments were conducted to examine BQTL's impact on osteoclast differentiation. RT-PCR and Western blot analyses were utilized to evaluate the relative expression levels of osteoclast-specific genes and proteins under BQTL stimulation. Finally, in vivo experiments were performed using an osteoporosis model to further validate the in vitro findings. This study revealed that BQTL suppressed receptor activator of NF-κB ligand(RANKL)-induced osteoclastogenesis and osteoclast resorption activity in vitro in a dose-dependent manner without observable *** inhibitory effects of BQTL on osteoclast formation and function were attributed to the downregulation of NFATc1 and c-fos activity, primarily through attenuation of the MAPK, NF-κB, and Calcineurin signaling pathways. BQTL's inhibitory capacity was further examined in vivo using an ovariectomized(OVX) rat model, demonstrating a strong protective effect against bone loss. BQTL may serve as an effective therapeutic TCM for the treatment of postmenopausal osteoporosis and the alleviation of bone loss induced by estrogen deficiency and related conditions.

Drought is one of the most severe environmental stresses affecting soybean growth and development,especially in arid and semi-arid *** aim of this experiment is to evaluate the effect of regulated deficit irrigation during the vegetative stages on soybean plants and determine the amount irrigation water can be reduced without affecting the physiological parameters,the crop phenology,and the yield of the soybean *** field experiments were conducted during two irrigation crop seasons(2021 and 2022)in Louata,*** results showed that regulated deficit irrigation regimes during the vegetative stages was combined with high temperatures and low air humidities during the beginning of flowering and the pod filling stage during 2021 in comparison with 2022,especially for 25%CWR(crop water requirements).Regulated deficit irrigation regimes reduced the stomatal conductance by 46%and 52%respectively during the first and second growing seasons by limiting CO2 intake for the Calvin *** stomata closure increased the leaf temperature and affected the functioning of the photosynthetic apparatus by damaging the chlorophyll pigments and impairment of electron transport chains in *** transition from regulated deficit irrigation to 100%CWR at the beginning of flowering(R1)compensated for the photosynthetic loss,improved the growth and development of soybean plants and enhanced the yield and its components for 50%and 75%*** adaptative mechanism such as the remobilization of the carbon reserved in the stems and leaves(vegetative tissues)to the grains improved the grain yield by 36.7%during 2021 and by 32.2%during 2022 *** consequently improved the water use efficiency,the water productivity of soybean for 50%and 75%CWR and contributed to water saving with an average of 60 mm per growing season.

Seed priming is an effective seed pretreatment technology that enhances germination and overall crop performance by optimizing seed hydration and metabolic processes before *** quality is a critical determinant of cotton(Gossypium hirsutum)crop performance,influencing germination,plant vigor,and *** study evaluates the effects of seed priming with potassium salts(1%and 2%KCl and K2SO4)on germination,morphological traits,and Cry1Ac gene expression in three Bt cotton cultivars(IUB-2013,NIAB-878B,FH-142)as Cry1Ac enhance the pest resistance in Bt cotton and reduce the plant’s dependence on chemical *** were primed for six hours,air-dried,and sown in the *** rates,plant height,number of bolls per plant,boll weight,seed cotton yield,and ginning outturn(GOT)were assessed at crop ***1Ac gene expression was quantified to explore the influence of priming treatments on transgene *** demonstrated that 1%K2SO4 priming significantly enhanced germination and yield-related traits,with Cry1Ac expression peaking in the IUB-2013 cultivar under 1%K2SO4 *** findings suggest that potassium-based halopriming improves cotton seedling establishment and Bt gene *** study addresses the critical gaps in understanding the effects of seed halopriming on morphological traits,germination,and expression of the Cry1Ac gene in Bt cotton while providing a novel eco-friendly and cost-effective halopriming approach,offering the potential to improve cotton production.

Background: Despite considerable advancements in identifying factors contributing to the development of hepatocellular carcinoma(HCC), the pathogenesis of HCC remains unclear. In many cases, HCC is a consequence of prolonged liver fibrosis, resulting in the formation of an intricate premalignant microenvironment. The accumulation of extracellular matrix(ECM) is a hallmark of premalignant microenvironment. Given the critical role of different matrix components in regulating cell phenotype and function, this study aimed to elucidate the interplay between the fibrotic matrix and malignant features in HCC. Methods: Liver tissues from both control(normal) and carbon tetrachloride(CCl_(4))-induced fibrotic rats were decellularized using sodium dodecyl sulfate(SDS) and Triton X-100. The resulting hydrogel from decellularized ECM was processed into micro-particles via the water-in-oil emulsion method. Microparticles were subsequently incorporated into three-dimensional liver biomimetic micro-tissues(MTs) comprising Huh-7 cells, human umbilical vein endothelial cells(HUVECs), and LX-2 cells. The MTs were evaluated using the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS) assay at day 11, immunofluorescence staining, immunoblotting, and spheroid migration assay at day 14 after co-culture. Results: Fibrotic matrix from CCl4-treated rat livers significantly enhanced the growth rate of the MTs and their expression of CCND1 as compared to the normal one. Fibrotic matrix, also induced the expression of epithelial-to-mesenchymal transition(EMT)-associated genes such as TWIST1, ACTA2, MMP9, CDH2, and VIMENTIN in the MTs as compared to the normal matrix. Conversely, the expression of CDH1 and hepatic maturation genes HNF4A, ALB, CYP3A4 was decreased in the MTs when the fibrotic matrix was used. Furthermore, the fibrotic matrix increased the migration of the MTs and their secretion of alpha-fetoprotein. Conclusions: Our findings suggest a

Pseudoalteromonas is a group of marine bacteria widespread in diverse marine sediments,producing a wide range of bioactive ***,only a limited number of Pseudoalteromonas phages have been isolated and *** this study,a novel lytic Pseudoalteromonas phage,denoted as vB_PalP_Y7,was isolated from sewage samples collected at the Seafood Market in Qingdao,***_PalP_Y7 remained stable across a wide range of temperatures(-20–50℃)and a wide pH range(3–12).The vB_PalP_Y7 phage harbors a linear double-stranded DNA molecule of 57699 base pairs(bp)with a G+C content of 45.90%.Furthermore,it is predicted to contain 58 open reading frames(ORFs).Phylogenetic analysis and protein network relationship analysis revealed low similarity between vB_PalP_Y7 and viruses in the ICTV and IMG/VR4 database,suggesting that vB_PalP_Y7 may be a potential new genus,*** study contributed valuable insights to comprehend the relationship between Pseudoalteromonas phages and their host organisms.

Gastrointestinal cancer has been one of the five most commonly diagnosed and leading causes of cancer mortality over the past few decades. Great progress in traditional therapies has been made, which prolonged survival in patients with early cancer, yet tumor relapse and drug resistance still occurred, which is explained by the cancer stem cell(CSC) theory. Oncolytic virotherapy has attracted increasing interest in cancer because of its ability to infect and lyse CSCs. This paper reviews the basic knowledge, CSC markers and therapeutics of gastrointestinal cancer(liver, gastric, colon and pancreatic cancer), as well as research advances and possible molecular mechanisms of various oncolytic viruses against gastrointestinal CSCs. This paper also summarizes the existing obstacles to oncolytic virotherapy and proposes several alternative suggestions to overcome the therapeutic limitations.

The intricate interplay between natural compounds like curcumin and the gut microbiome has gained significant attention in recent years due to their potential therapeutic implications in various health ***,a polyphenolic compound derived from turmeric,exhibits diverse pharmacological properties,including anti-inflammatory,antioxidant,and anticancer *** how curcumin modulates gut microbiota composition and function is crucial for elucidating its therapeutic *** review examines the current literature on the interactions between curcumin and the gut microbiome.A systematic search of relevant databases was conducted to identify studies investigating the effects of curcumin on gut microbial diversity and *** findings from studies exploring curcumin's efficacy in neurological disorders,gastrointestinal diseases,and metabolic dysfunction are synthesized and *** have demonstrated that curcumin supplementation can modulate gut microbiota composition and function,leading to beneficial effects on gut health and *** underlying curcumin's therapeutic effects include immune modulation,neuroprotection,and inflammation ***,challenges such as poor bioavailability and safety concerns remain significant hurdles to *** interactions between curcumin and the gut microbiome hold promise for therapeutic interventions in a diverse range of health *** research is needed to optimize curcumin formulations,improve bioavailability,and address safety concerns.
