The prefrontal neocortex is involved in many high cognitive functions in humans.Deficits in neuronal and neurocircuitry development in this part of the cerebrum have been associated with various neuropsychiatric disorders in adolescents and adults.There are currently little available data regarding prenatal dendrite and spine formation on projecting neurons in the human prefrontal neocortex.Previous studies have demonstrated that Golgi silver staining can identify neurons in the frontal lobe and visual cortex in human embryos.In the present study,five fetal brains,at 19,20,26,35,and 38 gestational weeks,were obtained via the body donation program at Xiangya School of medicine,Central South University,China.Golgi-stained pyramidal neurons in layer V of Brodmann area 46 in fetuses were quantitatively analyzed using the Neurolucida morphometry system.Results revealed that somal size,total dendritic length,and branching points of these neurons increased from 26 to 38 gestational weeks.There was also a large increase in dendritic spines from 35 to 38 gestational weeks.These findings indicate that,in the human prefrontal neocortex,dendritic growth in layer V pyramidal neurons occurs rapidly during the third trimester of gestation.The use of human fetal brain tissue was approved by the Animal Ethics Committee of Xiangya School of medicine,Central South University,China(approval No.2011-045)on April 5,2011.

Objective: To investigate the distribution and contents of vimentin(Vim) and glial fibrillary acidic protein(GFAP) immunoreactivities in the central nervous system(CNS)of normal newborn, adult and aged rats.Methods: In this study, thirty healthy and normal Sprague–Dawley rats were simply classified into three groups: Newborn(7 days aged), adult(5 months aged) and aged(24 months aged) rats. Brains and spinal cord were dissected and cut into frozen sections. The expression of Vim and GFAP in CNS were detected by confocal immunofluorescence.Results: In each group, Vim was expressed in all the regions of CNS including the hippocampal, cerebral cortex, the third ventricle and spinal cord, and the expression was highest in neuron-like cell of newborn rats, while Vim was mainly expressed in cell bodies in adult and aged rats. GFAP was expressed in all the regions of CNS including the hippocampal, cerebral cortex, the third ventricle and spinal cord, and the expression was in astrocytes of aged rats. In the third ventricle, Vim was detected in all groups, and only observed in neuron-like cells of newborn. Meanwhile, the GFAP expression showed no significant differences between adult and aged rats in this region. The co-localization of Vim and GFAP were mainly observed in hippocampus and cerebral cortex of newborn,but this co-localization was found in the third ventricle of the rats in all groups.Conclusion: Our data demonstrate for the first time that the expression of Vim and GFAP in the rat's CNS during development. This data may provide a foundation for the further mechanistic studies of these two main intermediate filaments during development of CNS.

In this paper, we study the optimal procurement management by reverse auctions for a price-setting newsvendor(retailer) in a single period setting. The retailer facing price-dependent stochastic demand first designs a procurement contract and then invites the suppliers to bid for this contract in the reverse auction. The winning supplier produces and delivers the demanded quantity.The retailer obtains the procurement quantity and simultaneously determines the retail price. By using the price elasticity of the lost-sales rate, we show that the retailer’s expected profit(excluding the procurement cost) is a concave function of the purchased quantity, which can be used to obtain the optimal procurement and retail pricing decisions for the retailer. Further, when the underlying random term of demand function is normally distributed under left-truncation(at 0), we get the analytical expressions of the purchased quantity and expected profit function for the retailer. Moreover, some numerical examples are given.

Stock market trading is an activity in which investors need fast and accurate information to make effective decisions.But the fact is that forecasting stock prices by using various models has been suffering from low accuracy,slow convergence,and complex parameters.This study aims to employ a mixed model to improve the accuracy of stock price prediction.We present how to use a random walk based on jump-diffusion,to obtain stock predictions with a good-fitting degree by adjusting different parameters.Aimed at getting better parameters and then using the time series model to predict the data,we employed the time series model to smooth the sequence utilizing logarithm and difference,which successfully resulted in drawing the auto-correlation figure and partial the auto-correlation figure.According to the comparative analysis,which focuses on checking the mean absolute error,including root mean square error and R square evaluation index,we have drawn a clear conclusion that our mixed model prediction effect is relatively good.In the context of Chinese stocks,the hybrid random walk model is very suitable for predicting stocks.It can“interpret”the randomness of stocks very well,and it also has an unparalleled prediction effect compared with other models.Based on the time series model’s application in this paper,the abovementioned series is more suitable for predicting trends.

Most prostate cancers initially respond to androgen deprivation therapy(ADT).With the long-term application of ADT,localized prostate cancer will progress to castration-resistant prostate cancer(CRPC),metastatic CRPC(mCRPC),and neuroendocrine prostate cancer(NEPC),and the transcriptional network shifted.Forkhead box protein A1(FOXA1)may play a key role in this process through multiple mechanisms.To better understand the role of FOXA1 in prostate cancer,we review the interplay among FOXA1-targeted genes,modulators of FOXA1,and FOXA1 with a particular emphasis on androgen receptor(AR)function.Furthermore,we discuss the distinct role of FOXA1 mutations in prostate cancer and clinical significance of FOXA1.We summarize possible regulation pathways of FOXA1 in different stages of prostate cancer.We focus on links between FOXA1 and AR,which may play different roles in various types of prostate cancer.Finally,we discuss FOXA1 mutation and its clinical significance in prostate cancer.FOXA1 regulates the development of prostate cancer through various pathways,and it could be a biomarker for mCRPC and NEPC.Future efforts need to focus on mechanisms underlying mutation of FOXA1 in advanced prostate cancer.We believe that FOXA1 would be a prognostic marker and therapeutic target in prostate cancer.

Effect and mechanism of Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia through connexin43(cx43)

Objective: To explore the effect and mechanism of angiotensin Ⅱ receptor blockers-Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia. Methods: Rats with embryonic cardiomyocytes-H9c2 were randomly divided into control group, ischemia group, Irbesartan group and Irbesartan+ischemia group. The cell viability of rats in each group was tested using MTT. Real-time PCR was employed to detect the expression of connexin43(Cx43) mR NA and western blot to detect the expression of Cx43 and phosphorylated Cx43. SD rats were randomly divided into the sham-operation group(SO), myocardial infarction group(MI), Irbesartan group and MI+ Irbesartan group, with 10 rats in each group. HE staining was employed to observe the change in the pathomorphology of left ventricular tissue and TUNEL method to analyze the cell apoptosis in the tissue. The immunofluorescence was adopted to observe the expression and distribution of Cx43 in the left ventricular myocardium and study the change in the expression of Cx43 in the cardiac muscular tissue at mR NA and protein level. Results: The intervention of Irbesartan in the condition of ischemia indicated the significant decrease in the number of necrotic cells. The expression of Cx43 was significantly decreased under the culture of ischemia(P<0.05), but in the presence of Irbesartan, the expression of Cx43 was increased compared with the ischemia group(P<0.01). The results of WB assay showed the similar trend of change at mRNA level. There was the significant difference in the score of ventricular arerythmia between MI group and SO group(P<0.01). The incidence of ventricular tachycardia or ventricular fibrillation was significantly increased compared with the one in SO group(P<0.05). There was the significant difference in the overall score between MI+Irbesartan group and MI group(P<0.05). The expression of Cx43 in the cardiac muscular tissue in MI group was significantly decreased(P<0.01 vs SO group). But the expression of Cx43 was increased after the treatment

Role of depressive symptoms in cardiometabolic diseases and subsequent transitions to all-cause mortality:an application of multistate models in a prospective cohort study

Background and purpose The role of depression in the development and outcome of cardiometabolic diseases remains to be clarified.We aimed to examine the extent to which depressive symptoms affect the transitions from healthy to diabetes,stroke,heart disease and subsequent all-cause mortality in a middle-aged and elderly European population.Methods A total of 78212 individuals aged≥50 years from the Survey of health Ageing and Retirement in Europe were included.Participants with any baseline cardiometabolic diseases including diabetes,stroke and heart disease were excluded.Depressive symptoms were measured by the Euro-Depression scale at baseline.Participants were followed up to determine the occurrence of cardiometabolic diseases and all-cause mortality.We used multistate models to estimate the transition-specific HRs and 95%CIs after adjustment of confounders.Results During 500711 person-years of follow-up,4742 participants developed diabetes,2173 had stroke,5487 developed heart disease and 7182 died.Depressive symptoms were significantly associated with transitions from healthy to diabetes(HR:1.12,95%CI:1.05 to 1.20),stroke(HR:1.31,95%CI:1.18 to 1.44),heart disease(HR:1.26,95%CI:1.18 to 1.34)and all-cause mortality(HR:1.41,95%CI:1.34 to 1.49).After cardiometabolic diseases,depressive symptoms were associated with the increased risk of all-cause mortality in patients with diabetes(HR:1.54,95%CI:1.25 to 1.89),patients who had stroke(HR:1.29,95%CI:1.03 to 1.61)and patients with heart disease(HR:1.21,95%CI:1.02 to 1.44).Conclusions Depressive symptoms increase the risk of diabetes,stroke and heart disease,and affect the risk of mortality after the onset of these cardiometabolic conditions.Screening and treatment of depressive symptoms may have profound implications for the prevention and prognosis of cardiometabolic diseases.

Sangguayin preparation(SGY-P) is refined from the traditional Chinese medicinal compound Sangguayin, which"clears heat and promotes fluid" and "tonifies kidney and spleen" for "Xiaoke", commonly known as ‘Diabetes mellitus’ in clinics.Previous studies have shown that SGY-P could reduce insulin resistance and repair damaged pancreas in db/db mice, but the underlying mechanisms were unclear. Here, we investigated whether treatment with SGY-P could protect pancreatic β-cells from apoptosis and uncovered the underlying mechanisms. db/db mice were used to observe the hypoglycemic and islet protective effect in vivo. Apoptosis was induced in mouse insulinoma 6(MIN6) cells by palmitate, following which the cells were treated with SGY-P for elucidating the anti-apoptotic mechanism in vitro. Cell viability and nuclear morphology were detected by CCK-8 assay and Hoechst 33258 staining. The expression levels of apoptosis-, endoplasmic reticulum(ER) stress-, and autophagy-related proteins were measured by western blot. The results showed that SGY-P reduced fasting blood glucose, pancreatic pathological changes, and islet β-cell apoptosis in db/db mice. Palmitate-induced apoptosis in MIN6 cells was decreased by SGY-P treatment. Hence, SGY-P therapy exhibited a protective effect on pancreatic β-cells by decreasing the expression of cleaved caspase-3, cleaved PARP and Bax, and increasing Bcl-2 by suppressing ER stress(Bip/XBP1/IRE1α/CHOP/Caspase-12) and autophagy(LC3/p62/Atg5) pathways.2/Atg5 pathways.

Do PON1-Q192R and PON1-L55M polymorphisms modify the effects of hypoxic training on paraoxonase and arylesterase activity?

Background:Low levels of antioxidant paraoxonase 1(PON 1)enzyme activity,PON1-Q192R polymorphism(a glutamine(Q)to arginine(R)substitution at position 192),PON1-L55M polymorphism(a leucine(L)to methionine(M)substitution at position 55),and oxidized low-density lipoprotein(oxLDL)are risk factors for coronary heart disease.Aerobic exercise improves PON1 activity,but the effects of hypoxic exercise are yet unclear.The aim of this study was to determine the effects of hypoxic underwater rugby training on PON1 activity and oxLDL levels and the role of the mentioned polymorphisms.Methods:Serum PON1 and arylesterase activities(ARE),PON1,PON3,and oxLDL protein levels(by using the enzyme-linked immunosorbent assays)were determined in an athletic group(42 trained male underwater rugby players;age=21.7±4.2 years,mean±SD)and a control group(43 sedentary men;age=23.9±3.2 years).The polymorphisms were determined from genomic DNA samples.Results:PON1 activity(25.1%,p=0.052),PON3(p<0.001),and oxLDL(p<0.001)of the athletic group,including most genotype groups,were higher than those of the control group.In comparison to the controls,PON1 activity levels(p=0.005)of the PON1-Q192R homozygote QQ genotype group and PON1 activity levels(30%,p=0.116)of the PON1-L55M homozygote LL genotype group were higher,whereas ARE activity values of athletic R allele carrier(Rc=QR+RR)(p=0.005)and LL group(p=0.002)were lower than the control genotype groups related to their polymorphisms.Conclusion:Hypoxic training can cause(1)significant oxidative stress,including oxLDL,and an antioxidant response(increase in PON1 activity and PON3),(2)differences in the activity of PON1 and ARE,which are modified by PON1-Q192R and PON1-L55M polymorphisms,respectively,and(3)improvements in PON1 activity of QQ and LL groups.However,hypoxic training can cause a disadvantage of LL and Rc groups for ARE.
