Characterization of multiple chemical co.ponents of GuiLingJi by UHPLC-MS and ^(1)H NMR analysis

GuiLingJi(GLJ),a classic traditional chinese medicine(TCM)formula,is co.posed of over 20 herbs,acco.ding to the Pharmaco.eia of the People's Republic of *** to its various activities,GLJ has been used in clinical settings for more than 400 years in ***,the ambiguous chemical material basis limits the development of studies on the quality co.trol and pharmaco.ogical mechanisms of ***,co.prehensive characterization of the multiple chemical co.ponents of GLJ is of great significance for the modernization of this *** the great variety of herbs in GLJ,both UHPLCMS and ^(1)H NMR techniques were employed in this *** addition,solvent extraction with different polarities was used to eliminate signal interference and the co.centration of trace co.ponents.A variety of MS analytic methods were also used,including implementation of a self-built co.pound database,diagnostic ion filtering,mass defect filtering,and co.pound Disco.erer 3.0 analysis *** on the above strategies,a total of 150 co.pounds were identified,including 5 amino acids,13 phenolic acids and glyco.ides,11 co.marins,72 flavones,20 triterpenoid and triterpenoid saponins,23 fatty acids,and 6 other ***,13 co.pounds were identified by ^(1)H NMR *** UHPLC-MS and ^(1)H NMR results supported and co.plemented each *** strategy provides a rapid approach to analyzing and identifying the chemical co.position of chinese herbal *** current study provides basis for further research on the quality co.trol and pharmaco.ogical mechanism of GLJ.

Efficacy and Safety of Lianhua Qingwen for Patients with co.ID-19: A Systematic Review and Meta-Analysis

Background: co.ona virus disease 2019(co.ID-19) has spread around the world since its outbreak, and there is no ascertained effective drug up to now. Lianhua Qingwen(LHQW) has been widely used in China and overseas chinese, which had some advantages in the treatment of co.ID-19. Objective: To evaluate the efficacy and safety of LHQW for co.ID-19 by co.ducting a systematic review with meta-analysis. Methods: A co.prehensive literature search was co.ducted in 12 electronic databases from their establishment to October 30, 2021. Note Express 3.2.0 was used for screening of trials, and the data was independently extracted in duplicate by 2 researchers. The risk of bias of randomized co.trolled trials(RCTs) and retrospective studies were assessed by using the co.hrane co.laboration tool and Newcastle Ottawa Scale, respectively, followed by data analysis using RevMan 5.3. The RCTs or retrospective studies to treat co.ID-19 using LHQW were included. The intervention measures in the experimental group were LHQW alone or co.bined with chemical drugs(LCWC), and that in the co.trol group were chemical drugs(CDs). Outco.e measures included co.puted tomography(CT) reco.ery rate, disappearance rates of primary(fever, co.gh, fatigue), respiratory, gastrointestinal and other symptoms, exacerbation rate and adverse reaction. Subgroup analysis was co.ducted acco.ding to whether LHQW was co.bined with CDs and the different treatment methods in the co.trol group. Results: Nine trials with 1,152 participants with co.ID-19 were included. The CT reco.ery rates of LHQW and LCWC were 1.36 and 1.32 times of CDs, respectively(Pco.pared with CDs, LCWC remarkably increased the disappearance rates of fever, co.gh, fatigue, expectoration, shortness of breath, and muscle soreness(P0.05). co.clusions: LHQW was more

Applying risk management to analytical methods for the desorbing process of ginkgo diterpene lactone meglumine injection

Analysis errors can occur in the desorbing process of ginkgo diterpene lactone meglumine injection(GDMI) by a co.ventional analysis method, due to several factors, such as easily crystallized samples, solvent volatility, time-co.suming sample pre-processing, fixed method, and offline analysis. Based on risk management, near-infrared(NIR) and mid-infrared(MIR) spectrosco.y techniques were introduced to solve the above problems with the advantage of timely analysis and non-destructive nature towards samples. The objective of the present study was to identify the feasibility of using NIR or MIR spectrosco.y techniques to increase the analysis accuracy of samples from the desorbing process of GDMI. Quantitative models of NIR and MIR were established based on partial least square method and the performances were calculated. co.pared to NIR model, MIR model showed greater accuracy and applicability for the analysis of the GDMI desorbing solutions. The relative errors of the co.centrations of Ginkgolide A(GA) and Ginkgolide B(GB) were 2.40% and 2.89%, respectively, which were less than 5.00%. The research demonstrated the potential of the MIR spectrosco.y technique for the rapid and non-destructive quantitative analysis of the co.centrations of GA and GB.

Effect of Zhuyeshigao granule on tumor necrosis factor-α and inter leukins serum protein levels in rats with radiation esophagitis

OBJECTIVE： To investigate the effects of Zhuyesh- igao granule （ZSG） on tumor necrosis factor-a （TNF-a）, interleukin-1 （IL-1）, IL-2, IL-6, and IL-8 in rats with radiation esophagitis. METHODS： Fifty Wistar rats were randomly divid- ed into five groups （10 rats in each group）： co.- trol （without radiation）, saline-treated, and low, medium, and high-dose ISG-treated groups. Rats were given normal saline （10 mL/kg） or 1.15, 2.3, or 4.6 g/kg ZSG by intragastric administration once a day for 7 days. A rat model of radiation esophagi- tis was established by local irradiation of co.0 （490.25 cGy/min, totaling 30 Gy）. The administra- tion of ZSG was co.tinued for another 7 days and on the 7th day post-irradiation, inferior vena cavablood was co.lected. The serum was separated, and TNF-a, IL-1, IL-2, 11_-6, and IL-8 protein levels were determined. RESULTS： Inflammatory response factors were found in the serum of each group. However, levels in ZSG-treated groups were significantly lower than in the saline-treated group （P〈0.05）. co.CLUSION： ZSG may prevent the development of radiation esophagitis, perhaps by inhibiting the generation and release of the inflammatory re- sponse factors TNF-a, IL-1, IL-2, IL-6, and IL-8.

Characterization of aqueous extract in traditional chinese medicine(TCM) is challenging due to the poor retention of the analytes on co.ventional C18 co.umns. This study presents a systematic characterization method based on a rapid chromatographic separation(8 min) on a polar-modified C18(Waters co.tecs T3) co.umn of aqueous extract of co.dyceps sinensis. UHPLC-HRMS method was used to profile co.ponents in both untargeted and targeted manners by full MS/PIL/dd-MS2 acquisition approach. The co.ponents were identified or tentatively identified by reference standards co.parison, fragmentation rules elucidation and available databases search. A total of 91 co.ponents, including 10 nucleobases, 20 nucleosides, 39 dipeptides, 18 amino acids and derivatives and 4 other co.ponents, were characterized from the aqueous extract of C. sinensis. And this was the first time to systematically report the presence of nucleosides and dipeptides in C. sinensis, especially for modified nucleosides. The chemical basis inquiry of this work would be beneficial to mechanism exploration and quality co.trol of C. sinensis and related products. Meanwhile, this work also provided an effective solution for characterization of aqueous extract in TCM.

An online automatic sorting system for defective Ginseng Radix et Rhizoma Rubra using deep learning

Objective:To establish a deep-learning architecture based on faster region-based co.volutional neural networks(Faster R-CNN)algorithm for detection and sorting of red ginseng(Ginseng Radix et Rhizoma Rubra)with internal defects automatically on an online X-ray machine vision ***:A Faster R-CNN based classifier was trained with around 20000 samples with mean average precision value(mAP)of 0.95.A traditional image processing method based on feedforward neural network(FNN)obtained a bad performance with the accuracy,recall and specificity of 69.0%,68.0%,and70.0%,***,the Faster R-CNN model was saved to evaluate the model performance on the defective red ginseng online sorting ***:An independent set of 2000 red ginsengs were used to validate the performance of the Faster RCNN based online sorting system in three parallel tests,achieving accuracy of 95.8%,95.2%and 96.2%,***:The overall results indicated that the proposed Faster R-CNN based classification model has great potential for non-destructive detection of red ginseng with internal defects.

Effects of glossy privet fruit on neural cell apoptosis in the co.tical parietal lobe and hippocampal CA1 region of vascular dementia rats

BACKGROUND： Glossy privet fruit inhibits neural cell apoptosis following the onset of vascular dementia. OBJECTIVE： To co.firm glossy privet fruit effects on neural cell apoptosis in the co.tical parietal lobe and hippocampal CAI region of rat models of vascular dementia using molecular biology techniques. DESIGN, TIME AND SETTING： The neural cell morphology experiment was performed at the Laboratory of Flow Cells and Biochemistry, Academy of Integrative medicine, Fujian University of traditional chinese medicine, and the Basic Room of Pathology, Academy of chinese medicine from December 2006 to May 2008. MATERIALS： A total of 60 Wistar rats were used to establish vascular dementia models using a photochemical reaction method. Glossy privet fruit was purchased from Fujian, China. Hydergine was co.produced by Sandoz, Switzerland and Huajin, China. METHODS： The 60 Wistar rats were randomly divided into 6 equal sized groups （n = 10）, i.e. model, blank, high, moderate and low doses of chinese medicine, and hydergine co.trol groups. Rats in the model group were treated with distilled water （1 mL/100 g） by gavage following model establishment. Rats in the blank group underwent experimental procedures as for the model group, except that rat models were created without illumination. Rats in the high, moderate and low doses of chinese medicine groups, and the hydergine co.trol group respectively received high, moderate and low doses of glossy privet fruit, and hydergine suspension （1 mL/100 g） by gavage, once a day, for 30 days. MAIN OUTco.E MEASURES： Morphology of neural cells from the rat co.tical parietal lobe and hippocampal CA1 region of all groups was observed with an electron microsco.e. Positive neural cells in the injury site of the rat co.tical parietal lobe and hippocampal CA1 region were investigated using the Fas immunohistochemieal method.
